Interakce povrchových proteinů spermií v reprodukčním procesu

Savčí reprodukce zahrnuje vysoce regulované biochemické interakce mezi komplementárními molekulami lokalizovanými na povrchu spermie a vajíčka a rovněž s látkami přítomnými v přirozeném okolí obou gamet. Proteiny kančí semenné plasmy AQN, AWN spermadhesiny a DQH protein, které jsou vázány na spermii během ejakulace, hrají důležitou roli v procesech reprodukce. Vazba spermie k buňkám oviduktu, rozpoznávání gamet a vazba spermie ke glykoproteinové síti zony pellucidy je zprostředkována protein-sacharidovou interakcí, lektinovou vazbou. Přirozenou funkční formou proteinů kančí semenné plasmy se zdají být jejich agregované formy, při kterých se uplatňuje další typ interakce (protein-protein). Proteinové agregáty tvoří povrchovou vrstvu spermie a její remodelování v samičím reprodukčním traktu je důležité během kapacitace spermie a pro primární vazbu spermie na vajíčko. Mammalian fertilization includes highly regulated biochemical interactions between complementary molecules located on the surfaces of both gametes as well as those present in the natural environment of the gametes. Boar seminal plasma proteins AQN, AWN spermadhesins and DQH protein that are bound to the sperm at ejaculation play important role in the reproduction process. Binding of sperm to epithelial cells, recognition of gametes and binding of sperm to glycoprotein of zona pellucida are mediated by protein-saccharide interaction, lektin-type interaction. Another type of interaction (protein-protein) participates in the formation of aggregated forms, which seem to be the natural functional forms of seminal plasma proteins. Protein aggregates form the sperm coating layers and their re-modeling in the female reproductive tract is imporatnt for the sperm capacitation and for the primary binding of spermto the ovum. Dvě protilátky proti angiotensinovému receptoru AT2 připravené proti dvěma krátkým sekvencím rozeznávaly receptor na Western blotech membrán různých tkání potkana a myši, v nichž se AT2 exprimuje. Obě protilátky rozeznávaly tytéž bílkoviny a po vysycení příslušným antigenním peptidem tuto schopnost ztratily. Jedna protilátka se osvědčila při imunoprecipitaci receptoru. Two antibodies against angiotensin receptor AT2 prepared against two short sequences recognized the receptor on Western blots of several rat and mouse tissues where AT2 is expressed. Both antibodies recognized the identical proteins; they lost this property after immunoabsorption with their antigen peptides. With one antibody, we were able to immunoprecipitate the receptor. Male infertility due to severe azoospermia has been associated with the absence of spermatozoa in ejaculated. We tested the ejaculates (IVF latoratory Pranatal Ltd) from 30 patients with azoospermia for the presence of cells from defferent stages of spermiogenesis. Monoclonal sntibodews against intraacrosomal human sperm proteins were used in the immunofluorescence test for detection of acrosomal proteins in the acrosomal precursors and acrosomes. We describe molecular cloning, expression, purification and properties of two single chain antibody variable fragments (scFv) of potential diagnostic use, namely scFv M75 and scFv Tu-20. The former scFv is derived from a monoclonal antibody M75 specific for a cell surface protein MN/CA IX, strongly associated with many types of human carcinomas. Characterization of peptide proteinase inhibitors isolated from boar seminal plasma

Most of proteins of boar seminal plasma are present in this medium under physiological conditions in the form of aggregates and not of monomers. In the present communication we have shown that not only spermadhesins, but also proteinase inhibitors participate in the formation of associated complexes. Proteinase inhibitors were found after gel chromatographic separation mainly in the fraction with the lowest relative molecular mass. Two different types of serine proteinase inhibitors were isolated from boar seminal plasma ů (i) glycosylated inhibitor with Mr 12 kDa, (ii) non-glycosylated sperm-associated acrosin inhibitor with Mr 8 kDa, and a new type of low molecular weight protein that is probably composed of two subunits coupled by the -S-S- bridge. The proteinase inhibitor with Mr 8 kDa was proved to form a heterodimer with AQN 1 spermadhesin under physiological conditions, whereas inhibitor with Mr 12 kDa forms homodimers.
