Biodiverzita myxozoí v přehradní nádrži Římov zjištěná analýzou environmentální DNA

Myxozoa představují rozmanitou skupinu mikroskopických endoparazitů, kteří infikují převážně ryby. V současnosti zahrnují více než 2600 popsaných druhů. Navzdory vysokému počtu popsaných druhů, zůstává jejich diverzita stále dostatečně neprozkoumána a podhodnocena. Detekce a sledování myxozoí v dosud neprozkoumaných oblastech může odhalit klíčové patogeny spojené s úhynem ryb a objevit biodiverzitu rybomorek. Tato práce se zabývá rozmanitostí parazitů v přehradě Římov, s důrazem na jejich detekci pomocí environmentální DNA (eDNA). Porovnání zjištěné diverzity na různých místech a v různých environmentálních vzorcích v přehradní nádrži Římov odhalilo zajímavé výsledky abundance a počtu OTUs v rámci jednotlivých linií myxozoí. Tato diplomová práce zdůrazňuje důležitost analýzy eDNA při detekci patogenů a posuzování biodiverzity myxozoí. Myxozoans represent a diverse group of microscopic endoparasites primarily infecting fish, including over 2,600 nominal species. Despite the extensive number of described species, their diversity remains still unexplored and underestimated. The detection and surveillance of myxozoan parasites in previously unexamined areas can reveal crucial pathogens linked to fish mortality and discover myxozoans biodiversity. This thesis investigates the parasite diversity in the Římov Reservoir, focusing on myxozoan detection through environmental DNA (eDNA). A comparison of detected diversity at various sites and in different environmental samples across the Římov Reservoir unveiled interesting patterns of read abundance and number of OTUs within each myxozoan clade. This study underscores the importance of eDNA analysis in detecting pathogens and assessing biodiversity in myxozoan parasites.

Intraspecific trait variability of herbaceous plants in organic and conventional olive plantations in region at desertification risk

This thesis is an ecological study in an agricultural context (agroecological research). It is carried out in olive plantations in south Italy, Apulia, a region at strong desertification risk. The effect of agricultural management (organic vs. conventional) is investigated on i) the herbaceous species composition ii) intraspecific trait variability of five selected herbaceous plants iii) soil conditions. Herbaceous plants are here considered as "indicators" of olive plantations' health. Studying if and how species traits and soil parameters vary with management, should reveal how olive understory grasslands are responding to increasing drought conditions and other desertification constraints. Management of grasslands and soils is critical to maintain high ecological functionality and services of olive agroecosystems. Vliv vybraných klíštěcích serpinů na zánětlivou aktivaci různých makrofágových subpopulací

Cílem práce bylo vytvořit tři subpopulace makrofágů z lidských liniových THP-1 buněk pomocí různých stimulačních látek a diferenciačních faktorů. Morfologicky je porovnat a rozlišit na základě exprese povrchových markerů. Dále provést zánětlivou aktivaci pomocí LPS a otestovat vliv dvou vybraných klíštěcích serpinů na zánětlivou aktivaci buněk. The aim of this study was create three subpopulation of macrohages from human cell lineage THP-1 using different stimulating agents and differentiation factors. Morhologically compare and differenctiate them based on the expresion of surface markers. Furhermore, to perform inflammatory activation using LPS and to test the effect of two selected tick serpins on inflammatory cell activation.

This thesis examines the patterns and ecological significance of bird vocalizations during the dawn chorus in the rainforests of Papua New Guinea. Specifically, it aims to identify the species involved, analyze their songs, and understand the influence of environmental factors. To accomplish this, the study employs a combination of field recordings, phylogenetic analysis, and statistical methods to elucidate the complex interactions between avian species and their habitats. The findings of this study contribute to the broader understanding of biodiversity and ecosystem dynamics in tropical rainforests. This thesis examines the patterns and ecological significance of bird vocalizations during the dawn chorus in the rainforests of Papua New Guinea. Jsou osobnostní rysy hraboše polního provázány s denním rytmem pohybové aktivity?

V této studii byl zkoumán vztah osobnostních rysů a denní aktivity jedinců hraboše polního (Microtus arvalis). Třicet čtyři hrabošů bylo odchyceno ve volné přírodě a chováno odděleně za stabilních světelných (L:D, 12:12) a teplotních (19 ? 1 oC) laboratorních podmínek. Hraboši byli podrobeni Open Field (OF) testům pro zjištění jejich osobnostních rysů na gradientech pohybové aktivity a odvážnosti. Poté byli podrobeni sledování ve PhenoTyper boxech, kde byla po celých sedm dní zaznamenávána jejich aktivita. V této diplomové práci bylo zjištěno, že více aktivní a odvážní (bold) jedinci v OF testu jsou také více aktivní ve PhenoTyper boxu, mají větší sklony k periodicitě a pravidelným ultradiánním rytmům a mají větší aktivitu ve tmě. Bázliví (shy) jedinci jsou často neaktivní v určitou dobu (nejčastěji v noci), což oslabuje jejich periodicitu ultradiánních rytmů. Tedy větší odvážnost u hraboše polního vede k větší noční aktivitě a bázlivost spíše k větší noční imobilitě. Osobnostní rysy byly spojeny s denní aktivitou, periodicitou a ultradiánními rytmy. In this study, the relationship between personality traits and daily activity of Common voles (Microtus arvalis) was investigated. Thirty four voles were captured for tests from the wild and kept separately under stable light (L:D, 12:12) and ambient temperature (19 ? 1 oC) laboratory conditions. Voles were subjected to Open Field (OF) tests to determine their personality traits on locomotor activity and shy-bold gradients. They were then monitored in the PhenoTyper boxes, where their activity was recorded for seven days. In this thesis, it was found that more locomotor active and bold individuals in the OF test are also more active in the PhenoTyper box, are more prone to periodicity and regular ultradian rhythms, and have more activity in the dark. Shy individuals are often immobile (most often in the dark), which weakens the periodicity of their ultradian rhythms. Thus, greater boldness in the common vole leads to greater nocturnal activity, and shyness rather to greater nocturnal immobility. The personality traits were associated with daily activity, periodicity, and ultradian rhythms.

Tato diplomová práce se zaměřuje na hudební subžánr grunge, a především na subkulturu, která k němu náleží. Zkoumá vznik a rozvoj subžánru, jeho vlivy i signifikantní znaky. Reflektuje rovněž specifické vizuální aspekty stylu grunge, které odrážely a jistým způsobem i definovaly tehdejší generaci mladých. Analyzuje také poselství a myšlenky spojené se subkulturou, často přinášející nové pohledy na autenticitu, svobodu a individualitu. Práce usiluje o zasazení grunge do nového rámce s tezemi sociologů Dicka Hebdige a Ericha Fromma. Skrze tuto analýzu se snaží porozumět, jaké prvky a hodnoty grunge komunitu formují a jakým způsobem se promítají do života jejích členů. Tímto komplexním přístupem se práce snaží vhodně osvětlit mnohostrannost a hloubku subkultury grunge a ukázat její významné místo v kulturním a sociálním kontextu. This thesis focuses on the musical subgenre of grunge, primarily concentrates on the subculture associated with it. It explores the formation and evolution of the subgenre, its influences, and significant characteristics. Additionally, it reflects the specific visual aspects of the grunge style, which both reflected and defined the youth of that time. The work also analyzes the messages and ideas associated with the subculture, often introducing new perspectives on authenticity, freedom, and individuality. It aims to contextualize grunge within a new framework using the theories of sociologists Dick Hebdige and Erich Fromm. Through this analysis, it seeks to understand the elements and values that shape the grunge community and how they manifest in the lives of its members. Through this comprehensive approach, the thesis endeavors to aptly elucidate the multifaceted nature and depth of the grunge subculture, highlighting its significant position within cultural and social contexts.

Slepé postavy v literárních dílech mohou v rámci literárního kontextu fungovat jako symboly, metafory či reprezentace určitých stereotypů. Tato práce se zaměřuje na vztah a souvislost mezi slepotou, genderem a sexualitou ve vybraných literárních dílech. Tato diplomová práce zkoumá přijetí či odmítnutí genderových stereotypů ve vztahu ke slepotě, analyzuje a porovnává aspekty slepoty aplikované na gender hlavních hrdinů, zkoumá specifické genderové role a stereotypy a jejich vliv na vnímání slepoty v literárních dílech. Blindness has been explored in literature from various perspectives and cultural backgrounds throughout history. Blind characters in literary works can function as symbols, metaphors, or representations of certain stereotypes, depending on the literary context. This thesis focuses on the correlation between blindness, gender, and sexuality in selected literary works. The diploma thesis examines the acceptance or rejection of gender stereotypes in relation to blindness, analyses and compares aspects of blindness as applied to the gender of the main protagonists, and explores the specific gender roles and stereotypes and their impact on the perception of blindness within literary works

Cílem této diplomové práce je představit práci Eugena O'Neilla, Louise Bryant a Johna Reeda v kontextu jejich působení v divadelním uskupení Provincetown Players. Teoretická část je rozdělena do tří rozsáhlých kapitol, kdy první z nich podává obraz americké společnosti na přelomu 19. a 20. století, ukazuje stav, ve kterém se nacházela kultura a především pak divadlo, a s přihlédnutím na předchozí jmenované se zaobírá důvody vzniku jednoho z nejdůležitějším amatérských divadel v Americe. Druhá kapitola v krátkosti představuje historii Provincetown Players, jejich přelomové počiny a nejvýznamnější členy. Teorii pak zakončujeme kapitolou mapující život O'Neilla, Bryant a Reeda před příchodem do Provincetownu, jejich pozdější pobyt v tomto přímořském městě, a především jejich vzájemný vztah, který je ovlivnil na rovině osobní i pracovní. S tímto vědomím pak v analytické části analyzujeme tři vybraná díla těchto autorů, konkrétně The Game Louisy Bryant, The Eternal Quadrangle Johna Reeda a na závěr Thirst Eugena O'Neilla. V této části se zaměřujeme nejen na obsahovou a formální úpravu her, ale hledáme také návaznosti s reálným světem a spojitosti mezi trojlístkem umělců. he main aim of this thesis is to present the work of Eugene O'Neill, Louise Bryant and John Reed in the context of the Provincetown Players. The first chapter presents a picture of American society at the turn of the 20th century, shows the state in which culture and, above all theatre could be found, and, considering the previously mentioned, deals with the reason for the creation of one of the most important amateur theatres in America. The second chapter briefly presents the history of the Provincetown Players, their biggest achievements and the most important members. We then conclude the theory with a chapter mapping the life of O'Neill, Bryant and Reed before coming to Provincetown, their later stay in this seaside town, and also their mutual relationship, which affected them both personally and professionally. With this in mind, in the second part of the thesis, we study three selected works by those authors, namely The Game by Louise Bryant, The Eternal Quadrangle by John Reed and finally, Thirst by Eugene O'Neill. In this last chapter, we not only analyze the plot, motives, themes and form of individual plays, but we will also look for the links with the real world and the connection between the trio of those artists.

Diplomová práce se zabývá klíčovými proměnami image ve 20. století, představuje hlavní módní trendy dominující především v Evropě a Americe a jejich propojení s politickým a společenským děním v průběhu století. Snaží se nabídnout pohled na image jakožto nástroj módního a ideologického diktátu, zmiňuje některé signifikantní osobnosti daných dekád i jejich vliv na oděvní revoluce. Pozornost je věnována také rebelujícím subkulturám odmítajícím masové trendy či temným kapitolám dějin, kdy svou roli hrálo nucené potlačení image na pozadí politického dění. The thesis deals with the key transformations of the image in the 20th century, presenting the main fashion trends dominating mainly in Europe and America and their connection with political and social developments during the century. It seeks to offer a view of image as a tool of fashion and ideological dictate, mentioning some significant figures of the decades and their influence on the clothing revolutions. Attention is also paid to rebellious subcultures rejecting mass trends or dark chapters of history, when the forced suppression of image against the background of political events played a role.
