Mechanism of retrograde transport in contact-dependent inhibition (CDI) toxins through the bacterial translocon

Due to the constant pressure, strategies to allow competition and communication have been developed within bacterial communities. In this respect, Contact Dependent Growth Inhibition (CDI) is often of relevance. The aim was to elucidate a newly found pathway of transport of the CDI toxins across the inner bacterial membrane via Sec translocon. Since all proteins are transported via Sec translocon in an unfolded state, and there is no obvious mechanism which would drive or facilitate this transport in the retrograde fashion, the hypothesis that the effector domain of CDI toxins evolved an anisotropic energy landscape of mechanical unfolding was constructed. That would allow the toxin to be mechanically more labile in the direction of translocation and mechanically stable in the orthogonal directions. This anisotropy would permit efficient translocation and overall thermodynamic stability at the same time. The unfolding landscape was assessed by a molecular dynamics simulation combined with umbrella sampling. Developed methodology, complemented with a rational design of "circular permutants", helped to estimate the forces required to unfold the toxins under different geometries. 2023 - English Hlavním cílem práce je analyzovat posledních padesát let energetické politiky v Dánsku a získat představu o tom, jak se Dánsko stalo jedním z evropských lídrů v dekarbonizaci ekonomiky a rozvoji obnovitelných zdrojů energie. Práce má rešeršní povahu a vychází z širokého přehledu akademických článků a knih o decentralizované výrobě energie a změně klimatu a zpráv vládních a mezinárodních organizací. Část věnovaná analýze a výsledkům je rozdělena do tří částí, které pokrývají tři fáze dánské energetické transformace a analyzují různé vládní politiky prováděné v těchto fázích. - První fáze: Prvotní nárůst výroby energie z obnovitelných zdrojů do energetického systému. - Fáze druhá: Hledání řešení nestability dodávek obnovitelných zdrojů energie, když je výroba energie z obnovitelných zdrojů mezi 40 a 50 %. - Třetí fáze: Hledání řešení pro ukládání v systému 100 % obnovitelné energie. Výzkum vedl k některým důležitým zjištěním, které jsou užitečné pro další státy, jež se snaží dekarbonizovat své ekonomiky. Práce je uzavřena vybranými politickými doporučeními. The main aim of the thesis is to analysis the past fifty years of energy policy in Denmark to gain insight on how Denmark has become one of the European leaders in decarbonising its economy and developing renewable energy resources. A thorough review of academic articles, government and international institutional reports, and books on decentralised energy and climate change were essential for the research required for this work and make up the literature review section. The analysis and results section is divided into three parts that cover the three phases of the Danish energy transition and analyses the different government policies implemented during the different stages and how well they were in achieving the government's goals. - Phase one: Adding renewable energy production into energy system. - Phase two: Seeking solutions to the intermittency of renewable energy technologies when renewable energy production is between 40-50%. - Phase three: Seeking storage solutions in an 100% renewable energy system. The research led to some important insights that may be useful for other states seeking to decarbonise their economies within their own contexts and some policy recommendations conclude the paper. 2023 - English Práce je zaměřena na variabilitu pozice adjektiv v substantivních frázich ve staré a střední angličtině. Tato problematika je zkoumána na podkladu šesti staroanglických a středoanglických homílií a kázání. Práce nejprve popisuje tyto texty s ohledem na gramatické změny, které se v období staré a střední angličtiny udály, se zaměřením na ztrátu flektivních koncovek a ústup silné a slabé deklinace. Dále se v rámci substantivní fráze zaměřuje na souhru faktorů, kterými jsou adjektivní pozice, silná a slabá deklinace (především ve staré angličtině), určitost a neurčitost substantivní fráze a význam. Práce se také věnuje spojení mezi pozicí adjektiv a informační strukturou textu. Práce se krátce věnuje dvěma případům podobnosti pozice adjektiv v substantivní frázi v minulosti a dnes. The thesis is focused on the variability of adjectival position in noun phrases in the Old English and Middle English periods, which is analyzed on the textual samples of six Old English and Middle English homilies and sermons. It describes these texts with regards to their general characteristics and focuses on the grammatical changes that occurred in the Old English and Middle English periods, especially with regards to leveling of inflectional endings and the gradual loss of strong and weak inflection. Subsequently, it focuses on the interplay of the factors of adjective position, strong or weak inflection (particularly in Old English), definiteness or indefiniteness of the noun phrase, and meaning. It also explores the connection of adjective position and information structure of the texts. It devotes brief attention to two cases of similarity between adjective placement nowadays, and in the past. The portrayal of class in Charles Dickens' works

2023 - English Bohatství a společenské postavení mělo ve viktoriánské Anglii velký vliv na vnímání a sudbu člověka takřka ve všech směrech. Charles Dickens v tomto období vyrůstal jako člen právě té nižší sorty lidí, na kterou toto pojetí společnosti mělo ten největší dopad. Kvůli dluhům svého otce žil dlouhou dobu spolu s rodinou ve vězení pro dlužníky, a kvůli matce se mu nedostávalo důkladného vzdělání. Ve své podstatě jim křivdu nikdy nedokázal odpustit, a to, spolu s dalšími faktory, se také odráží v jeho pozdější tvorbě. Cílem mé bakalářské práce je studium zobrazování tříd v Dickensových dílech v kontextu s jeho životem a dobou, jejíž názory tak ovlivnily jeho literární příběhy. Za pomocí postav z jeho knih, které zastupují jednotlivé sociální skupiny, se pokusím vykreslit Charlesovo vnímání, pojetí a následné zakomponování témat společenské nerovnosti do jeho tvorby. Wealth and social status in Victorian England had a great influence on perceiving a person in nearly all directions. Charles Dickens grew up in this time period as a member of the lower class of people on which this perception of society had a huge impact. Thanks to his father's debts, he lived, together with his family, for quite a long time in a debtor's prison, and he lacked a sufficient amount of education because of his mother. In essence, he was never able to forgive them, which got portrayed, as well as many other factors, in his later works. The aim of my bachelor's degree thesis is a study of the depiction of social classes in Dickens' writings in the context of his life and period, whose opinions influenced his literary stories. With the help of the characters from his books, who represent each social class, I will try to picture Charles' conception and inclusion of the topics of social inequality in his work. Applying CA to the Understanding of How Interactional Teaching Strategies Contribute to the Development of Communicative Competence: A Case Study of Online English Lessons Delivered to a Group of Four Mature Students

2023 - English Zvládnout vztah se svými studenty je jedním z nejdůležitějších úkolů učitele. Pokud je vztah mezi ním a studenty dobře vybudován a stabilně nastaven, může se učitel dále soustředit na to, jakým způsobem řídit interakci ve třídě. V tomto ohledu však nelze dospět k jednomu univerzálnímu závěru, proto cílem práce bylo zmapování samotného průběhu interakce. Studie je zaměřena hlavně na takové části hodin, kdy vyučující usilovala o obecný rozvoj nejen řečových dovedností v cizím jazyce, ale i plynulosti mluveného projevu. Zkoumání proběhlo v rámci případové studie provedené na čtyřčlenné skupině studentů angličtiny jakožto cizího jazyka. Všichni byli ve věku 50+ a lišili se nejen úrovní jazyka, ale i motivací pro studium; jednalo se o malou heterogenní skupinu dospělých studentů. Výuka probíhala kompletně v online prostředí v období pandemie Covid-19. Z jedenácti výukových hodin o přibližné délce 60-90 minut bylo shromážděno 50 vzorků dat, které byly podrobeny rozboru a interpretaci. Analýza dat byla provedena za pomoci metodologie konverzační analýzy, konkrétně konverzační analýzy pro výuku cizího jazyka (CA-for-SLA). Prostřednictvím této metodologie došlo k samotnému sběru dat skrze audionahrávky, jejichž části pak byly doslovně přepsány za použití zjednodušeného transkripčního systému. Přepisy byly dále zkoumány a rozebírány za účelem zachycení interakčních strategií užívaných učitelem v rámci procesu výuky cizího jazyka, skrze něž se projevovala jeho rozvinutá interakční kompetence přispívající k rozvoji komunikační kompetence studentů. Maintaining relationships with students is one of the most important tasks for a teacher. If the relationship is well-built and steady, the teacher can start focusing on the way of managing classroom interaction. In this regard, there is no single universal conclusion, therefore this thesis will aim to map the process of interaction itself. The study focuses mainly on the parts of sessions in which there occurred the successful development of language skills in the foreign language as well as the development of fluency in the spoken language. The research takes the form of a case study of a group of four students of English as a foreign language. All of them were more than fifty years old. Each student had not only a different level of language skills but also different motivations for studying English-it was a small heterogenous group of adult learners. The teaching process was done fully in the online environment during the Covid-19 pandemic. 50 data samples were gathered from 11 sessions, each spanning approximately 60-90 minutes. These became the subject of analysis and interpretation. Data analysis was done using the conversational analysis methodology, specifically conversational analysis for second language acquisition (CA-for-SLA). Via this methodology, the process of data collection was done through audio recordings. Parts of them were transcribed word-by-word, using a simplified transcription system. The transcripts were further studied and analysed to capture interactional strategies used by a teacher during the language acquisition process through which developed interactional competence was shown, and how this competence contributed to the development of learners' communicative competence. Literární použití bezejmennosti slouží k rozvoji postav, zkoumání struktur moci a výzvě tradičním představám o identitě a přináležitosti. Autorka využívá různé teoretické perspektivy, včetně těch Josefa Campbella, Carla Gustava Junga, Michela Foucaulta, Gérarda Genetta, Jane Blissové a Lao Tzua, aby tento fenomén analyzovala. Tím, že čerpá z těchto různorodých perspektiv a aplikuje různé metody analýzy, práce zdůrazňuje interdisciplinární povahu studie, vycházející z teorií v dějepisu, mytologii, psychologii, filosofii a literární kritice. The thesis explores the significance and implications of namelessness in fantasy literature on the example of A Song of Ice and Fire by George R. R. Martin and the Earthsea series by Ursula Le Guin. The literary use of namelessness serves to develop characters, examine power structures, and challenge traditional notions of identity and belonging. The author uses various theoretical perspectives, including those of Josef Campbell, Carl Jung, Michel Foucault, Gérard Genette, Jane Bliss, and Lao Tzu to analyze the phenomenon. By drawing on these diverse perspectives and applying different methods of analysis, the thesis highlights the interdisciplinary nature of the study, drawing from theories in history, mythology, psychology, philosophy, and literary criticism. This master's thesis examines the significance of collocations in the English language teaching at the corresponding level of secondary schools, i.e., B1 and B2 levels according to the Common European Framework of Reference for Languages. The thesis deals with the linguistic aspect of collocations in the theoretical part and is based on the methodological approaches to vocabulary teaching. It analyses selected English language coursebooks: English File, Headway, New Success, High Note, and Maturita Solutions, in order to gain insight into the methodological approaches to developing collocational competence, and the variety of exercises related to collocations. The research offers valuable resources and activities designed to support deeper understanding and practical application of collocational competence among language learners. V první části bakalářská práce krátce představí autorův život nejen jako kritika, ale také jako esejisty a novináře, začátek jeho literární kariéry a proces psaní zmíněných děl. Prostřední část práce se bude věnovat utopii a dystopii: její definici, historii a vývoji vliteratuře jako fiktivním žánru aideologii totalitarismu. Vposlední, klíčové části bakalářské práce přistoupíme přímo k analýze děl Farma zvířat a 1984 a budeme zkoumat jejich žánrové zařazení. The purpose of the bachelor's thesis Genre analysis of anti-utopia in George Orwell's works is to analyse the anti-utopian works Animal Farm (1945) and 1984 (Nineteen Eighty-Four, 1949) written by British writer George Orwell. The first part of this bachelor thesis will shortly introduce the author's life not only as a critic but also as an essayist and journalist, the start of his literary career, and the process of writing the literary works mentioned earlier. The middle part of the thesis is dedicated to utopia and dystopia: its definition, history, and its evolution in literature as a fiction genre and ideology in totalitarianism. In the last, crucial part of the bachelor's thesis, we will proceed to the analysis of the works Animal Farm and Nineteen Eighty-Four and we will be examining their genre classification. Feminism in The Scarlet Letter and comparing Hester Prynne with Margaret Fuller's writings in Woman in the Nineteenth Century

2023 - English Tato práce zkoumá otázku feminismu jako jednoho z přístupů k postavě Hester Prynne z románu Šarlatové písmeno od Nathaniela Hawthorna. Tento román je považován za feministický nejen kvůli tomu, jak je v něm Hester vykreslena, ale také kvůli faktu, že kniha byla napsána v době vzrůstu feministického hnutí a také kvůli tomu, že měl Hawthorne blízko k Margaret Fullerové, která byla jednou z předních feministek své doby. Na základě srovnání poznámek z knihy Fullerové Žena v Devatenáctém Století a diskuze o dalších přístupech k románu tato práce ukazuje, že Hester Prynne, ačkoli vykazuje známky feminismu, není ani tak moc feministkou jako spíše transcendentalistkou. Toto je zřejmé, když si čtenář uvědomí, že Hester nezměnila pohled společnosti na nezávislé ženy, které usilují o své cíle, ale že se snaží žít sama pro sebe v souladu s Bohem a přírodou. This thesis explores the issue of Feminism as one of the approaches to Hester Prynne from Nathaniel Hawthorne's novel The Scarlet Letter. The novel is considered feminist not only because of how Hester is portrayed in the book, but also because it was written during the rise of the feminist movement and also because Hawthorne was close to Margaret Fuller who was one of the leading feminists of her day. Having compared notes from Fuller's Woman in the Nineteenth Century and having discussed other approaches to the novel, this paper shows that Hester Prynne, although she does show signs of it, is not as much a feminist rather than a transcendentalist. This judgment is evident when the reader realizes Hester did not change how society views independent women pursuing their goals, but rather trying to live for herself in harmony with God and nature.
