Use of Monovalent Copper for Sensitive Detection of Methyl Derivatives of Xanthine

This work is concerned with the electroanalysis of xanthine (Xan) and its methylated derivatives (1-, 3-, 7-, and 9-mXan) on a pencil graphite electrode in the presence of copper ions. The main idea of using copper consists in the in situ formation of the complex Cu(I)-mXan on the electrode surface and improvement of oxidative signals of the corresponding mXan during the detection. Linear sweep voltammetry (LSV) in connection with adsorption stripping techniques was used for the determination of mXan. To enhance the oxidation signals the elimination voltammetric procedure (EVP) was applied. Possibility of DNA probes preparation for want of CMG

The main method of cytogenetics is the DNA fluorescence in situ hybridization (DNA-FISH), where DNA probe links to the specific locus of genome. In our laboratory, DNA probes are prepared from DNA amplified by bacterial vector (PAC/BAC). The following step is exploiting Nick-translation reaction for incorporation of reporter molecule targeted for antibody conjugate with fluorescence molecule. Specificity of prepared DNA probe is increased using Cot-DNA, which suppress the repetitive sequences. Důležitou metodou cytogenetiky je DNA fluorescenční in situ hybridizace (DNA-FISH), kdy dochází ke specifické vazbě DNA sondy na určité místo genomu. V naší laboratoři jsou DNA sondy připravovány z DNA naamplifikované pomocí bakteriálního vektoru (PAC/BAC). Nicktranslační reakcí je do DNA vkládaná reportérová molekula vizualizovatelná protilátkou konjugovanou s fluorescenční látkou. Vystíněním repetitivních sekvencí pomocí Cot-DNA je následdně zvýšena specifičnost připravené sondy. Keywords: cytogenetics; FISH; DNA probe Available at various institutes of the ASCR

In the first part there is given a summary of present knowledge on the origin of life on Earth. Further there are given several views of acknowledged scientists on science. These opinions are confronted with some interpretations of facts in popular articles. The paper warns against the danger of influencing science by ideology. V první části je uveden souhrn soudobých poznatků o vzniku života na Zemi. Dále je uvedeno několik názorů uznávaných vědců na vědu. Tyto názory jsou konfrontovány s některými interpretacemi fakt ve vědecko-populárních článcích. Přednáška upozorňuje na nebezpečí ideologizace vědy. Genomic imprinting represents an epigenetic marking of genes that results in their monoallelic expression, namely in mammals and flowering plants. Here, I also summarize our recent experiments describing the role of DNA methylation and histone H4 acetylation in plant reproductive development. Evolution of electrochemical methods: from virtual capabilities to virtual instruments

Progress in electrochemical methods is tightly bound with the development of electronics and instrumentation. Modern development environments allow one to create software for measurement control, data processing and modelling without deep knowledge of computer programming. Utilisation of one of those environments, the LabView, for modelling the results of adsorption kinetics measurement on mercury drop electrode will be demonstrated in the presentation. Vývoj elektrochemických metod je úzce spjat s vývojem přístrojů a elektroniky. Moderní vývojová prostředí umožňují vyvíjet programy pro řízení měření, zpracování a modelování naměřených dat bez hlubokých znalosti v oboru programování. V příspěvku bude demonstrováno využití jednoho z takových prostředí - LabView - pro modelování výsledku měření kinetiky adsorpce látky na povrchu rtuťové kapkové elektrody.
