Utilization of polymerase chain reaction, PCR-RFLP technique and sequence analysis for determination of homozygotes, intraspecific heterozygotes and interspecific hybrids of the genus Salvelinus in breeding facilities

2017 - Czech The methodology provides relevant information and reliable instruments for determination of the most frequently bred fish of the genus Salvelinus in the Czech Republic and in European countries. It includes the optimized process of identification of four species (Salvelinus fontinalis, Salvelinus alpinus, Salvelinus umbla, Salvelinus namaycush) in the homozygous and heterozygous state and in their hybrid combinations. The methodology describes the present situation in identification of the above mentioned fish species in the Czech Republic including its drawbacks and at the same time proposes a new and reliable solution utilizing polymerase chain reaction, PCR-RFLP restriction technique and sequencing. Visualization and separation of diagnostic fragments is performed either on agarose gel or on the Fragment AnalyzerTM. Cílem metodiky je poskytnout relevantní informace a spolehlivé nástroje k determinaci nejčastěji chovaných ryb rodu Salvelinus v ČR a v evropských státech. Metodika obsahuje optimalizovaný postup identifikace čtyř druhů (Salvelinus fontinalis, Salvelinus alpinus, Salvelinus umbla, Salvelinus namaycush) v homozygotním i heterozygotním stavu a v jejich hybridních kombinacích. Metodika popisuje aktuální stav identifikace uvedených druhů ryb v ČR včetně jejich nedostatků a současně navrhuje nové a spolehlivé řešení s využitím polymerázové řetězové reakce, restrikční techniky PCR-RFLP a sekvenace. Vizualizace a separace fragmentů je prováděna buď na agarózovém gelu anebo na přístroji Fragment AnalyzerTM. Kvalita prostředí a potravní chování kapra ve vztahu k přikrmování v polointenzivní rybniční akvakultuře

2017 - Czech Kombinací čtyř metod terénního výzkumu (telemetrie, elektrolov, potravní analýzy a měření parametrů vody) se ukázalo, že ryby nejsou v polointenzivním rybničním chovu rozmístěny rovnoměrně. Dominantní jedinci se koncentrují v krmných místech v hustotě, která může snížit obsah kyslíku až k hranici fyziologického minima, což vede k horšímu využití krmiv. Menší ryby se zdržují spíše mimo krmná místa a v okrajích rybníka a živí se převážně přirozenou potravu. Kromě menší hmotnosti mají i nižší obsah tuku. Rozdělení obsádky kapra na větší jedince s vyšším obsahem tuku, významně se orientující na krmná místa, a na menší s nižším obsahem tuku živící se více přirozenou potravou vede k doporučení vhodnosti rozšíření aplikace krmiv na více krmných míst, případně jejich střídání. Morphological differences in farm groups of salmonid fishes

2016 - Czech The aim of this study was examination of a influence species/line and stock conditions. This paper record first results form three groups of salmonid fishes – rainbow trout (from Italy and Denmark) and brook trout hybrides (S. fontinalis x S. alpinus). There were choosen morphological parameters observed – body contruction, weigth of intestine organs, epidermal and gill structure and erythrocyte size. Cílem studie je posouzení vlivu druhu/linie a podmínek chovu. Tato práce zachycuje první průběžné výsledky získané na základě tří skupin lososovitých ryb – pstruha duhového původem z Itálie, z Dánska a hybridních jedinců sivena (S. fontinalis x S. alpinus). Sledovány byly vybrané morfologické parametry – stavba těla, hmotnost vnitřních orgánů, struktura pokožky a žáber a velikost krvinek. 2016 - Czech The screening survey of fish stock composition was performed at the pilot sites of future monitoring, namely in the facilities of the Kroměříž Castle (reservoirs and ponds in the garden and park), Lednice Castle (park pond), Ploskovice Castle (park pond), Červené Poříčí (park pond) and Němčice reservoir (flood protection). Screeningový průzkum rybích obsádek byl proveden na lokalitách zámek Kroměříž (nádrže a rybníky v zahradě a parku), zámek Lednice (Růžový rybník v zámeckém parku), zámek Ploskovice (rybník v Zámeckém parku), zámek Červené Poříčí (rybník v zámeckém parku) a nádrž Němčice (poldr).
