Prions (PrP) are the main cause of neurodegenerative diseases such as Scrapie in sheep, bovine spongiform encephalopathy, chronic wasting disease in deer, and Creutzfeldt-Jakob disease in humans. Although the cellular PrP (PrPC ) is involved in many cellular processes, its precise function still needs to be discovered. The disease is caused by the accumulation of a pathological form of PrP (PrPTSE ), which is caused by direct contact of PrPTSE and PrPC . PrP is anchored in the membrane by GPI and can be transmitted by cell-to-cell contact, tunnelling nanotubes, or extracellular vesicles (EVs). EV factions are divided by different biogenesis into exosomes, microvesicles, and apoptotic bodies. PrPTSE was found in exosomes and microvesicles, but these fractions were never compared to each other. The first aim of the doctoral thesis is a comparison of PrP content, prion-converting activity and infectivity in these fractions on CAD5 and N2a-PK1 cellular models of infection. We isolated a fraction of large EVs (20,000× g) and small EVs (110,000× g) by centrifugation from a conditioned medium. We characterised EVs by cryo-electron microscopy and western blot with Alix, TSG-101, CD63, CD9, and HSP70 markers. The contamination from other cellular compartments was checked by calnexin. Priony (PrP) jsou hlavní příčinou neurodegenerativních onemocnění jako je ovčí klusavka, bovinní spongiformní encefalopatie, chronické strádání jelenovité zvěře a lidská Creutzdfeld- Jacobova choroba. Přestože se buněčný PrP (PrPC ) účastní mnoha buněčných procesů, jeho přesná funkce není dosud známá. Onemocnění vzniká akumulací patologické formy PrP (PrPTSE ), která vzniká přímým kontaktem PrPTSE a PrPC . PrP je ukotvený v membráně pomocní GPI a může být přenášen mezi buňkami kontaktem, tzv. "tunnelling nanotubes" nebo extracelulárními vezikuly (EV). Frakce EV se dělí na exosomy, mikrovezikuly a apoptotická tělíska rozdílnou biogenezí. PrPTSE se nachází v exosomech a mikrovezikulech, ale tyto frakce EV nikdy nebyly mezi sebou porovnány. Prvním cílem dizertační práce je porovnat obsah PrP, prion konvertující aktivitu a infektivitu v těchto frakcích EV na CAD5 a N2a-PK1 buněčných modelech infekce. Centrifugací jsme izolovali frakci velkých EV (20 000× g) a malých EV (110 000× g) z kondiciovaného média. EV jsme charakterizovali pomocí kryo-elektronové mikroskopie a western blotem s markery Alix, TSG-101, CD63, CD9 a HSP70. Kontaminace z jiných buněčných kompartmentů byla ověřena kalnexinem. 4 Abstract The work is based on a revision of collection from several scientific institutions from the Czech Republic and the Federal Republic of Saxony (National Museum in Prague, Chlupáč Museum of Earth History, Senckenberg Natural History Collections Dresden, etc.). The collection is based on samples published by Oskar Lenz and George Bruder from the second half of the 19th century. Both authors also processed the findings of other researchers (e.g. Cotta, Weisse). The belemnite rostra were systematically divided into four families, five genera and seven identifiable genera. One rostrum has not exactly been determined. The age of the samples was established to be the Middle and Upper Jurassic (Bajocian- Kimmeridgian). The length of the expected sedimentation in this area has been prolonged to the past (deeper to the Middle Jurassic), mainly due to the finding of the species Megateuthis gigantea Schlotheim, 1820. Earlier research (especially in the 20th century) assumed the extent of preserved carbonate rocks only in the Oxfordian and Kimmeridgian age, with a not much fossiliferous Callovian Brtníky Formation. The studied sedimentary rocks show a significant influence of Milankovich cycles with repeated alternation of carbonate and non- carbonate layers in the sequence. Preliminary results of isotope... 3 Abstrakt Práce je založena na revizi sbírkového materiálů z několika vědeckých institucí z České republiky a Spolkové republiky Sasko (Národní muzeum Praha, Chlupáčovo muzeum historie Země, Senckenberg Natural History Collections Dresden aj.). Základ sbírky tvoří vzorky publikované Oskarem Lenzem a Georgem Bruderem z druhé poloviny 19. století. Oba čerpali také z nálezů jiných badatelů (např. Cotta, Weisse). Rostra belemnitů byla systematicky rozřazena do čtyř čeledí, pěti rodů a sedmi určitelných rodů. Jedno rostrum nebylo blíže určeno. Stáří vzorků bylo určeno na střední a svrchní juru (bajok-kimmeridž). Délka předpokládané sedimentace v této oblasti byla především díky nálezu druhu Megateuthis gigantea Schlotheim, 1820 posunuta výrazně do střední jury. Dřívější výzkumy (především 20. století) předpokládaly rozsah zachovaných karbonátových hornin jen ve stupních oxford a kimmeridž, s málo fosiliferními brtnickými vrstvami stáří keloveje. Zkoumané horniny vykazují značný vliv Milankovičových cyklů s opakovaným střídáním karbonátových a nekarbonátových poloh ve vrstevním sledu. Title: Sytematics, palaeoecology and trophical structure of bivalve associations within the OAE2 in the Bohemian Cretaceous Basin (Upper Cenomanian): Pecínov quarry Author: Bc. RNDr. Martin Košťák, Ph.D. Abstract: This master thesis provides an updated taxonomic inventory of the bivalve fauna of the Pecínov locality, places the fauna in its palaeobiogeographic setting, and addresses its palaeoecological significance. In the literature research part of the thesis the basic geology of the locality and an overview of the Bohemian Cretaceous Basin are presented. Processes connected to the ocean anoxic events, specifically to the OAE 2 are also discussed. The research part of the thesis describes the systematics of the bivalve assemblage (with the exception of Inoceramidae) and its statistical evaluation with the emphasis on the environment interpretations of the locality. In total, over 350 specimens have been considered and assigned to 20 genera within 15 families and 9 orders. Bivalve functional morphology and guilds are described to interpret palaeoecology. Guilds are based on life habit and feeding category. Four main guilds are recognized: epifaunal suspension-feeders, semi-infaunal suspension-feeders, infaunal... RNDr. Martin Košťák, Ph.D. Abstrakt: Diplomová práce shrnuje poznatky o společenstvu mlžů přítomném na lokalitě Pecínov, jejich taxonomický přehled a paleoekologické vyhodnocení. V rámci řešeršní části je stručně popsána geologie oblasti a lokalita je zařazena do kontextu české křídové pánve. Část práce je také věnována procesům souvisejícím s anoxickými událostmi, zvláště pak s druhou anoxickou událostí na hranici cenoman/turon. Praktická část práce se zabývá systematikou zjištěných společenstev mlžů (s výjimkou inoceramidní fauny) a statistickým zpracováním výsledků s důrazem na paleoekologickou analýzu společenstva a její využití pro interpretaci paleoprostředí. Celkem bylo v rámci práce zpracováno přes 350 jedinců a rozpoznáno bylo 20 rodů z 15 čeledí a 9 řádů. Podrobně je diskutována funkční morfologie a zástupci jsou rozřazeni do čtyř skupin na základě životní strategie: epifauní filtrátoři, semi-infauní filtrátoři, infauní filtrátoři a infauní detritofágové. Manuel Miró Title of the diploma thesis: Automated monitoring of Metridia luciferase released from genetically modified cell line. The diploma thesis reports automatic real-time monitoring of the release of Metridia luciferase from genetically modified cells. A previously optimized method for monitoring the natural release of luciferase from cells was used during the experiment. At the same time, the toxic effect of bile acids (chenodeoxycholic and deoxycholic acid) on changes in cell membrane permeability was tested. Liver epithelial cell line modified with the Metridia luciferase plasmid were used during the experiment. After this genetic modification, the cells were able to synthesize the active Metridia luciferase enzyme. This enzyme generates luminescence upon reaction with substrate coelenterazine. The emitted light was detected in the flow cell of a spectrofluorometer at a wavelength of 485 nm. The measurement was taken in a sequential injection system connected to a 3D printed module containing an insert with the tested cell monolayer. Diplomová práce se zabývá automatickým monitorováním uvolněné Metridia luciferázy z geneticky modifikovaných buněk v reálném čase. Během práce byla použita již dříve optimalizovaná metoda pro monitorování přirozeného přebytku luciferázy z buněk. Zároveň byl testován toxický účinek žlučových kyselin (chenodeoxycholové a deoxycholové) na změny permeability buněčné membrány. Během experimentu byly použity jaterní epiteliální buňky, které byly modifikovány Metridia luciferázovým plazmidem. Po této genetické modifikaci byly buňky schopny syntetizovat aktivní enzym Metridia luciferázu. Tento enzym vykazuje po reakci se substrátem koelenterazinem luminiscenci. Emitované záření bylo následně detekováno v průtokové cele spektrofluorimetru při vlnové délce 485 nm. Měření bylo provedeno za pomocí sekvenční injekční analýzy, která byla propojena přes průtokový systém s 3D tištěnou celou, jenž obsahovala insert s testovanou monovrstvou buněk. Environmental seasonality, and associated fluctuation in resource abundance, affects organisms throughout their life cycles. Periods of resource scarcity can be avoided by seasonal migration, strategy common in animals, including birds. However, the long journeys of migratory birds separate the stages of their annual cycles in space and time, and present them with a variety of challenges. Environmental conditions that migrants experience may have local effects, or may carry over across seasons, and have delayed impacts on individuals and populations. To better understand the interactions between migratory animals and their environment, we integrated individual tracking, environmental measures, and population data within a full annual cycle perspective. We contributed to the understanding that environmental conditions can affect migratory populations both en route and during stationary non-breeding periods, and that their effects can be observed on a continental scale. We also showed, across species and migratory flyways, that stages of the annual cycle are tightly linked during migration, but that these links weaken during prolonged non-breeding stationarity, with implications for individual success and future prospects under ongoing environmental change. To enhance the ability to trace animal... Sezónnost prostředí, doprovázená kolísající dostupností potravy, ovlivňuje organismy v průběhu celého života. Obdobím nedostatku zdrojů se lze vyhnout sezónní migrací, která je u živočichů včetně ptáků běžná. Dlouhé cesty však staví tažné ptáky před řadu problémů, protože jednotlivá období ročního cyklu jsou od sebe oddělena v prostoru i čase. Podmínky prostředí, které tažní ptáci využívají, mohou na ptáky působit lokálně, ale jejich vliv se může projevit i napříč sezónami. Abychom lépe porozuměli interakcím mezi tažnými ptáky a prostředím, propojili jsme data ze sledování migrace s daty o podmínkách prostředí a populačních změnách v rámci celého ročního cyklu. Přispěli jsme tak k poznání, že podmínky prostředí mohou ovlivňovat migrující populace jak během zimování, tak v průběhu tahových zastávek, a že tyto dopady lze pozorovat i v kontinentálním měřítku. Ukázali jsme také, že napříč druhy a tahovými cestami jsou fáze ročního cyklu během migrace úzce provázány, ale že tyto vazby slábnou v průběhu dlouhých období zimování, což má důsledky pro úspěšnost jedinců a pro predikci budoucích scénářů ve světle globálních změn. Title: Electro and thermal magnetotransport in antiferromagnetic systems Author: João Pedro Martins Godinho Abstract: How to electrically identify the Néel vector reversal in PT symmetric systems? The detection of magnetic order reversal has been one open problem in antifer- romagnetic spintronics. More so, in systems like the tetragonal CuMnAs which preserve the combined PT symmetry, since the combined PT symmetry renders odd magnetore- sistance effects such as the anomalous Hall effect to be zero. This thesis focuses on trying to provide an answer to the question above. We present the measurement of second- order magnetotransport effects as a mechanism to identify the reversal of the Néel order. We show it in two different systems, firstly on an out-of-plane synthetic antiferromagnet and later in the tetragonal CuMnAs. We also introduce a scanning thermal gradient microscopy which allows us to exploit different thermoelectric effects depending on the symmetries of the chosen material. For instance, in the collinear CuMnAs we take advan- tage of the magneto-Seebeck effect to image changes in the magnetic structure due to the application of electrical current pulses. In the case of the noncollinear antiferromagnetic Mn3Sn, the anomalous Nernst response is used to map the magnetic order. Keywords:... Detekce obrácení magnetického uspořádání je dosud nevyřešeným prob- lémem v antiferomagnetické spintronice. To platí zejména pro systémy jako tetragonální CuMnAs, které zachovávají kombinovanou PT symetrii, protože ta způsobuje, že liché magnetoresistivní efekty, jako je anomální Hallův jev, jsou nulové. Tato práce se pok- ouší zodpověďet výše uvedenou otázku. Předkládáme měření magnetotransportních jevů druhého řádu jako mechanismus pro identifikaci obrácení Néelova uspořádání. Ukazu- jeme to na dvou různých systémech, nejprve na syntetickém antiferomagnetu a později v tetragonálním CuMnAs. Filip Seitl Modeling and statistics of random tessellations with ap- plications to the study of microstructure of polycrystallic materials We present a parametric statistical methodology for analysing a Laguerre tessel- lation data set viewed as a realization of a marked point process of generators. We study the dependence of the marks and the point process in detail. Further, we suggest two general models. The first one is based on marked Gibbs point processes and leads to a broad class of Gibbs-Laguerre tessellations. Under mild assumptions we prove the existence of the infinite-volume Gibbs measure. Then the choice of energy function for applications is discussed in detail. The sec- ond model is hierarchical, where in the first step the point pattern is modelled and in the second step the marks are modelled conditionally on points, using exponential family models based on geometrical characteristics of the tessella- tion. Statistical tools for the parameter estimation, model selection and fitting are suggested and implemented. We apply our methodology for a 3D Laguerre tessellation data set representing the microstructure of a polycrystalline metal- lic material, where simulations under a fitted model may substitute expensive laboratory experiment. 1 Filip Seitl Modelování a statistika náhodných mozaik s aplikacemi ve studiu mikrostruktury polykrystalických materiálů Práce popisuje statistickou metodologii sloužící k analýze Laguerrových mozaik. Na Laguerrovu mozaiku lze nahlížet jako na realizaci kótovaného bodového procesu generátorů. V práci je podrobně studována závislost kót a bodového procesu. Následně jsou prezentovány dva parametrické modely. První model je založen na kótovaných Gibbsových bodových procesech a vede na širokou třídu takzvaných Gibbs-Laguerrových mozaik. Za určitých předpokladů je dokázána existence nekonečných Gibbsových měr. Podrobně je diskutována volba funkce energie vhodná pro aplikace v materiálovém inženýrství. Druhý model zavádí hierarchický přístup, kdy je v první řadě modelován bodový vzorek poloh ge- nerátorů, a v druhém kroku jsou pak modelovány kóty podmíněně na zna- losti poloh. V obou krocích jsou modely vyjádřené hustotou spadající do ex- ponencíální rodiny hustot. Práce popisuje různé statistické nástroje pro odhad parametrů a výběr vhodného modelu. 1 Contemplated reforms of the U.S. presidential election Abstract This diploma thesis is dedicated to the U.S. presidential election, specifically the greatest attention is paid to the electoral system called the electoral college which is used during the election. The text of my thesis relates to the US presidential election which is generally a very extensive topic therefore it is not possible to analyze in detail all the individual aspects of the process and thus provide a complete thorough analysis within this thesis. As a result of this, my aim is to briefly present the historical development of the electoral college along with its current functioning during the election process of the President of the United States. Subsequently, I am going to focus closely on potential reforms of the presidential elections that could significantly contribute to the improvement of the entire process of selecting (electing) the president of the United States. Uvažované reformy prezidentských voleb v USA Abstrakt Tato diplomová práce se zabývá americkými prezidentskými volbami, největší pozornost je věnována samotnému volebnímu systému používanému během voleb, odborně nazývanému electoral college. Text práce se dotýká amerických prezidentských voleb, které jsou obecně velice obsáhlým tématem, a proto není možné detailně rozebrat všechny jejich jednotlivé oblasti, a tedy i zcela vyčerpat rozsah diplomové práce. Kvůli uvedené skutečnosti patří mezi cíle v prvních dvou částech diplomové práce stručně představit historický vývoje electoral college spolu s jeho současným fungování v rámci volby prezidenta USA. Poslední část práce se detailněji věnuje potenciálním reformám prezidentských voleb, jež by mohly významně přispět ke zlepšení celého procesu výběru, resp. volby amerického prezidenta. The current development of copyright law in the digital environment with the emphasis on the legislation of the EU Abstract This rigorous thesis follows up on the thesis from 2018 titled "Internet and Copyright", which generally dealt with the scope of copyright law in the internet environment and was concluded by a basic presentation of the forthcoming copyright reform in the Digital Single Market in the European Union. The goal of this thesis is therefore to reflect and analyze not only the result of this reform, which was completed in the meantime at the EU level and moved to the implementation phase at the level of the member states, but also other related developments that have taken place in the field of copyright in recent years, with relevance for the digital environment and information society in general. The thesis is systematically divided into two parts. The first, more general part creates the theoretical and legal basis of the thesis, in which the following interpretation is embedded. The first chapter thus briefly presents the basic theoretical-legal basis for copyright and the justification for its existence and offers an overview of its main legal sources relevant to the digital environment at three levels: international, EU and Czech. Aktuální vývoj autorského práva v digitálním prostředí s důrazem na právní úpravu Evropské unie Abstrakt Tato rigorózní práce navazuje na diplomovou práci z roku 2018 na téma Internet a autorské právo, která se obecně zabývala působností autorského práva v prostředí internetu a byla zakončena základním představením připravované reformy autorského práva na jednotném digitálním trhu v Evropské unii. Cílem této práce je proto reflektovat a analyzovat nejen výsledek této reformy, který byl v mezidobí na unijní úrovni završen a přehoupl se do fáze implementační na úrovni členských států, ale i jiný související vývoj, který se v oblasti autorského práva v posledních letech odehrál, s relevancí pro digitální prostředí a obecně informační společnost. Práce je systematicky rozčleněna do dvou části. První, více obecná část, vytváří teoreticko-právní základ práce, do kterého je zasazen následující výklad. První kapitola tak stručně představuje základní teoreticko-právní východiska pro autorské právo a odůvodnění jeho existence a nabízí přehled jeho hlavních právních pramenů relevantních pro digitální prostředí na třech úrovních: mezinárodní, unijní a české. v anglickém jazyce This thesis, while using the knowledge of legal theory, aims to offer a practical legal manual about theatre in Czech Republic well-arranged and understandable even for laymen in the field of both law and theatre. The first part provides a basic description of the intellectual property law, especially the copyright, which is the predominant legal branch in theatre practice. Historical sources of copyright in what is now Czech Republic are mentioned, considering the Theatre Acts which are no longer in effect, as well as international and constitutional bases for the current copyright legislation. Subsequently, the current copyright legislation is introduced with emphasis on explanation of some pivotal provisions of the Copyright Act. The second part defines some basic theatre concepts (such as "theatre play", "theatre production", "theatre performance" etc.) necessary for understanding the rest of the thesis. The third part analyses most frequent types of contracts in the theatre practice. That means the contract of employment and the difference between the employee and the self-employed, the license agreement and, last but not least, the contractor agreement. The fourth part presents the copyright works most commonly present in theatre environment, focusing on their specific... v českém jazyce Tato rigorózní práce si klade za cíl na základě teoretického výkladu nabídnout praktickou právní příručku pro oblast divadla v České republice, která bude přehledná a srozumitelná, aby ji pochopil i právní či divadelní laik. První část poskytuje základní vymezení práva duševního vlastnictví, zejména autorského práva, neboť to je dominantní právní odvětví v divadelní praxi. Jsou zde zmíněny historické prameny autorského práva na území dnešní České republiky s přihlédnutím k již neplatným divadelním zákonům, jakož i mezinárodní a ústavní východiska aktuální autorskoprávní úpravy. Následně je představena aktuální právní úprava autorského práva, s důrazem na výklad některých klíčových ustanovení Autorského zákona. V druhé části jsou vysvětleny některé základní divadelní pojmy (například "Divadelní hra", "Divadelní inscenace", "Divadelní představení", apod.), které jsou klíčové pro další výklad. Ve třetí části jsou rozebrány nejčastější smluvní typy, se kterými se lze v divadelní praxi setkat. Zejména se jedná o pracovní smlouvu, kdy je věnována zvláštní pozornost rozdílu mezi zaměstnancem a osobou samostatně výdělečně činnou, dále o licenční smlouvu a v neposlední řadě smlouvu o dílo.
