Similarly to other scienti c domains, the expenses related to in silico modeling in pharmacology need not be extensively apologized. Vis a vis both in vitro and in vivo experiments, physiologically-based pharmacokinetic (PBPK) and pharmacodynamic models represent an important tool for the assessment of drug safety before its approval, as well as a viable option in designing dosing regimens. In this contribution, some special techniques related to the mathematical modeling, control and optimization of biochemical networks are presented on a paradigmatic example of enzyme kinetics.

On the 3D Dynamics of the Wake Behind a Circular Cylinder

Flow in the wake of a circular cylinder is studied experimentally using time-resolved stereo PIV method. Special attention is paid to 3D topology of dynamical structures. While the distribution of statistic quantities along the cylinder is uniform, and i.e. 2D, the instantaneous flow structure is fully 3D. Within the velocity fluctuating flow field the structures containing streamwise vorticity and velocity components are dominant. The POD modes connected with von Kármán vortex street are identified.

Excessive extrapolations of Einstein's equations

The standard cosmological model is surprisingly quite thoroughly investigated even though it possesses many paradoxes. We present several arguments indicating why excessive extrapolations of Einstein's equations to cosmological distances are questionable. First, we show how to express explicitly the first of Einstein's 10 partial differential equations to demonstrate their extremely large complexity. Therefore, it would be very difficult to find their solution for two or more bodies to model, e.g., the evolution of the Solar system. Further, we present some unexpected failures of the Schwarzschild and Friedmann solution of these equations. Then we explain why application of Einstein's equations to the whole universe represents incorrect extrapolations that lead to dark matter, dark energy, and several unrealistic situations. Finally, we give 10 further arguments showing why celebrated Einstein's equations do not describe reality well.

The fundamental magnetic characteristics of R2Fe14B intermetallics are highly sensitive to atomic substitutions and interstitial absorption of light elements. In this work, both were combined, and the influence of the substitutions in the R-sublattice and hydrogen absorption on the magnetization of (RR’) 2Fe14BHx (R is Tm or Er, and R’ is Nd) in magnetic fields up to 58 T was studied. By using direct hydrogen absorption, (R0.5Nd0.5)2Fe14BHx compounds were obtained with hydrogen content 0≤x≤5.5. Magnetic measurements were carried out on nanopowders. The strength of the inter-sublattice coupling in (R0.5Nd0.5)2Fe14B and their hydrides is estimated by analysing high-field magnetization data. It is shown that hydrogenation weakens the inter-sublattice exchange interaction up to 45 %.

Stability of a bar influenced by small and large imperfections

The geometrical and physical imperfections of systems can drastically reduce their critical loading. These imperfections are usually of stochastic character and, therefore, they act as random parametric perturbations of coefficients of corresponding differential equations. In this paper, the imperfections are introduced as multidimensional statistics on the set of a large number of realizations of the same system. As far as the amount of information is small or the imperfections themselves cannot be considered small, the convex analysis is preferable. The paper compares results obtained by both stochastic and convex analyses for hyperprism and demonstrates when each of them is more convenient to be used. Besides of the hyper-prism, the possibilities and properties of other modifications of convex method are considered, especially those based on the definition of imperfection zone marked as a centric hyper-ellipsoid or as an eccentric hyper-ellipsoid. The analytical background was brought up to the level when only a few configurations of imperfections are sufficient to be evaluated numerically. These configurations are obtained by means of the convex analysis as points of extreme critical loading using the Lagrange method of constrained extremes.

The experimental set-up for studying dynamical behaviour of the bladed wheel with pre-stressed dry-friction contacts in tie-bosses was built. The numerical solution of the turbine bladed wheel with tiebosses based on 3D finite element method with surface to surface dry friction contact model is proposed. Comparison of experimental and preliminary numerical results of dynamical behaviour and damping estimation of our bladed wheel design yielded a reasonable agreement. Due to dry friction contacts and non-linear solution of 3D FE model it, however, leads to HPC computations and long computation times. The contribution deals with description of computational strategy for damping evaluation and achieved results, too.

This work deals with the preparation and methodology of temperature measurement inside the rubber sample. There is a design of a rubber sample with placed temperature sensors and a description with a scheme of temperature measurement under torsional dynamic stress. The reason for these measurements is to determine the temperature dependence and sensitivity of the parameters of the thermal outputs.
