Mathematics and Optimal control theory meet Pharmacy: Towards application of special techniques in modeling, control and optimization of biochemical networks

Similarly to other scienti c domains, the expenses related to in silico modeling in pharmacology need not be extensively apologized. Vis a vis both in vitro and in vivo experiments, physiologically-based pharmacokinetic (PBPK) and pharmacodynamic models represent an important tool for the assessment of drug safety before its approval, as well as a viable option in designing dosing regimens. In this contribution, some special techniques related to the mathematical modeling, control and optimization of biochemical networks are presented on a paradigmatic example of enzyme kinetics. Metoda rentgenové mikro-tomografie je stále více využívána mimo jiné při výzkumu materiálových charakteristik a lomového chování stavebních materiálů. Poskytuje slibné výsledky v oblasti trojrozměrného pozorování a kvantifikace prvků mikrostruktury, včetně objektů lomu. Se standardním přístupem k získávání tomografických dat je však dosažitelné rozlišení výrazně omezeno velikostí zkoumaného objektu, což má za následek omezení nebo dokonce úplnou ztrátu užitečných informací týkajících se například tvaru a velikosti lomové procesní zóny. Z tohoto důvodu byly v této práci testovány možnosti tomografického cílení na zájmovou oblast s vyšším dosažitelným rozlišením, než je obvyklý limit tomografických systémů. The method of X-ray micro-tomography is increasingly used in the research of material characteristics and fracture behavior of building materials. It brings promising results in the field of three-dimensional observation and quantification of microstructure elements, including fracture objects. However, with the standard approach to the tomographic data acquisition, the achievable resolution is severely limited by the size of the examined object resulting in a reduction or even complete loss of useful information regarding, for example, the shape and size of the fracture process zone. For this reason, the possibilities of tomographic targetting of the region of interest with a higher achievable resolution than the usual limit of the tomographic systems have been tested in this work. Grantový projekt, jehož výstupem je kniha, je pokračováním a prohloubením tohoto výzkumu. Důraz je kladen na mezinárodní spolupráci a na přeshraniční a transnacionální perspektivy. The introductory chapter of the book presents the history of research into the phenomenon of forced service to the inhabitants of the occupied countries in the German armed forces. The grant project, the output of which is the presented book, is a continuation and deepening of this research. Emphasis is placed on international cooperation and on cross-border and transnational perspectives. On the influence of diffusion stabilization in Oldroyd-B fluid flow simulations

This work presents some numerical tests of finite element solution of incompressible Oldroyd-B fluid flows. The effect of numerical stabilization using artificial stress diffusion is investigated in detail. The limits of Weissenberg number We for which it is possible to obtain the numerical solution were studied depending on the Reynolds number Re and the diffusion parameter. Series of numerical tests were performed for steady two-dimensional Oldroyd-B fluid flow in corrugated channel (tube). The numerical results clearly proved the advantage (higher attainable We) of stabilized numerical method over the classical formulation without the artificial stress diffusion. Středověký autor se vymyká jak modernímu pojetí autora, tak i pojetí strukturalistickému zejména proto, že je zakotven ve zcela odlišné historické realitě: charakter středověkého textu, používání psaného záznamu, ani způsob šíření díla neodpovídají tomu, co dnes považujeme za literaturu, a ani pojem či status autora se s moderní představou příliš nepřekrývá. Počátky moderního autorství lze ale sledovat již ve středověku, zejména definujeme-li autora ve vztahu k textu, dílu a čtenáři, a to jednak statusem autora, který mu přiznali čtenáři a jednak subjektivitou, tj. vědomím sebe sama jako tvůrce. Na příkladu básníka a hudebního skladatele Guillauma de Machaut popisuje tato studie jak řadu fenoménů typických pro středověk, kdy dominovala orální performance, objednávka mecenáše a kdy byla umělecká tvorba plně integrována do běžného života, tak i zrození subjektivity autora, která se utváří ve vztahu k mecenáši a je důsledkem reflexe vlastní sociální role v dvorském prostředí. The medieval author eludes both the modern concept of the author and the structuralist concept, because he belongs to a different historical reality: the character of a medieval text, the use of a written record and the way in which a work was disseminated do not correspond to what we consider literature today, and neither the concept of the author nor his status coincide with the modern notion. The origins of modern authorship can be traced as early as the Middle Ages, if we define the author in relation to the text, work and reader, in terms of the status granted to an author by his readers, and also in terms of his subjectivity. Taking as its example the poet and composer Guillaume de Machaut, this paper describes a number of phenomena typical of the Middle Ages, when oral performances and commissions by patrons dominated. It also describes the genesis of the author’s subjectivity, which was formed in relation to his patron and reflects Machaut´s social role. On the 3D Dynamics of the Wake Behind a Circular Cylinder

Flow in the wake of a circular cylinder is studied experimentally using time-resolved stereo PIV method. Special attention is paid to 3D topology of dynamical structures. While the distribution of statistic quantities along the cylinder is uniform, and i.e. 2D, the instantaneous flow structure is fully 3D. Within the velocity fluctuating flow field the structures containing streamwise vorticity and velocity components are dominant. The POD modes connected with von Kármán vortex street are identified. Excessive extrapolations of Einstein's equations

The standard cosmological model is surprisingly quite thoroughly investigated even though it possesses many paradoxes. We present several arguments indicating why excessive extrapolations of Einstein's equations to cosmological distances are questionable. First, we show how to express explicitly the first of Einstein's 10 partial differential equations to demonstrate their extremely large complexity. Therefore, it would be very difficult to find their solution for two or more bodies to model, e.g., the evolution of the Solar system. Further, we present some unexpected failures of the Schwarzschild and Friedmann solution of these equations. Then we explain why application of Einstein's equations to the whole universe represents incorrect extrapolations that lead to dark matter, dark energy, and several unrealistic situations. Finally, we give 10 further arguments showing why celebrated Einstein's equations do not describe reality well. Laminar-Turbulent Transition in View of Flow Separation from Isothermal and Heated Cylinder

This work presents insight to the laminar-turbulent transition in flow around cylinder through a numerical simulation, both in isothermal and heated case. The flow is modelled directly as a solution to the Navier-Stokes and Navier-Stokes-Fourier system. Reynolds numbers are considered in range 100-20000, ratio of cylinder-wall and upstream temperature is up to 1.5. Curves representing evolution of separation positions in time provide novel insight to vortex formation. Various regimes are recognized and some of them can be assigned to phenomena described formerly in experiment.
