On UV-Vis Spectroelectrochemistry of Luteolin-7-O-Glucoside

The UV-Vis spectroelectrochemical cell containing an optically transparent platinum net used as the working electrode was tested in two regimes of electrochemical measurement, cyclic voltammetry and chronoamperometry, respectively. A three electrode setup used silver quasi reference electrode and platinum electrode serving as a counter electrode. The calibration of the spectroelectrochemical cell was performed for one-electron oxidation processes as well as for two-electron oxidations. The spectral changes during the oxidation of luteolin-7-O-Glucoside gave the information about the electroactive chromophores in molecule. Resulted spectra were compared with luteolin. The identification of products of oxidation was done by complementary analytical techniques HPLC-DAD and HPLC-ESI-MS/MS. Charge Transport in Single Oligophenylene Molecular Wires with Different Anchoring Groups

This work compares single molecule conductance measurements of selected organic systems containing identical oligophenylene molecular wires and different tripodal anchoring groups. Single molecule conductance G was obtained by a scanning tunneling microscopy break junction technique complemented by theoretical calculations based on the density functional theory and non-equilibrium Green’s function formalism. Two molecules were compared where the same oligophenylene wire is connected to one of the electrodes via a tripod substituted on each leg by a thiol group either in the meta or para position. By combined experimental and theoretical analysis it was possible to confirm that single molecule conductance in the metal-molecule-metal junction of both molecules corresponds to a fully extended molecular wire, which is attached to one of the electrodes by all three thiolate bonds of the tripod. Experimental results confirmed that G value of meta-connected molecules is lower than that of para, whereas junction formation probability was higher for meta functionalization. Voltametrie na uhlíkové pastové elektrodě modifikované silikagelem

Kompositní uhlíkové elektrody s obsahem silikagelu 1,2 se začaly používat již v 90. letech minulého století, a to ve variantě pevné (s obsahem ceresinu) nebo pastové (s parafinovým olejem). Přítomnost silikagelu v uhlíkové pastě vede ke zlepšené amperometrické detekce látek, které lze na povrchu modifikovaném silikagelem nahromadit, a to jak látek organických, tak iontů kovů 3. Reprodukovatelnost povrchu a tím i vlastního stanovení je nejlépe zabezpečena použitím pastové elektrody se speciálně vyvinutým teflonovým držákem s pohyblivým pístem 4. Možnosti charakterizace uhlíkových pastových elektrod byly dále popsány v přehledném článku 5.\nVlastnosti uhlíkové pasty modifikované silikagelem byly dokumentovány při oxidaci flavonoidů a oxidaci kadmia v přítomnosti cysteinu a fytochelatinu PC2. Voltammetry using silica gel modified carbon paste electrode (SiO2-CPE) for oxidation of selected flavonoid showed better sensitivity and reproducibility than oxidation using glassy carbon or unmodified carbon paste electrode. Only SiO2-CPE is useful for study of cadmium complexation with cysteine containing ligand as phytochelatin or metallothionein. Voltammetry on boron doped diamond electrode showed only peak of sulfur compound oxidation, which decreased with the addition of cadmium ions. Adhesion and growth of adipose tissue-derived stem cells on fibrin assemblies with attached growth factors for tissue engineering of heart valves

Currently used xenogeneic biological heart valve prostheses are decellularized and crosslinked with glutaraldehyde. These grafts usually undergo degeneration and calcification. Pericardium-based heart valve prostheses, re-seeded with autologous cells, i.e. Adipose tissue-derived cells (ASCs) and endothelial cells, could have longer durability and biocompatibility. In order to improve the adhesion of cells and their ingrowth into decellularized pericardium, various fibrin (Fb) layers were developed, i.e. Fb, Fb with covalently bound heparin (H), Fb with either vascular endothelial growth factor (VEGF) or fibroblast growth factor 2 (FGF) in various concentrations (1 ng/ml, 10 ng/ml, 100 ng/ml) or with both VEGF and FGF (100 ng/ml). Growth factors were attached onto Fb via heparin or were adsorbed. ASCs were seeded on theses layers in a DMEM medium supplemented with 2 % of fetal bovine serum, TGFβ1 and BMP-4 (both 2.5 ng/ml), and with ascorbic acid. Cell adhesion and growth/viability was assessed by counted cell number/MTS evaluation. ASCs were stained for differentiation markers of smooth muscle cells, such as alpha-actin, calponin, and myosin heavy chain. On day 7, ASCs on Fb-H-VEGF layers produced both calponin and alpha-actin. An increased FGF concentration caused reduced calponin staining of ASCs. Lack of heparin in fibrin assemblies with growth factors inhibited the production of both alpha-actin and calponin in ASCs. The highest ASCs density/viability was found on Fb-H-VEGF-FGF layer. The proper formulation of fibrin coatings could be favorable for ASCs growth and differentiation and could subsequently support endothelialization of cardiovascular prostheses with endothelial cells. Atmosférický aerosol (AA) ovlivňuje nejen vznik oblačnosti, ale také další vlastnosti oblaků. Interakce mezi AA a oblačností může být zkoumána i u mlh, resp. nízké oblačnosti. Příkladem stanice vhodné k takovému výzkumu je stanice Milešovka, kde se mlha (nebo nízká oblačnost) nachází 55 % času (Fišák, Tesař, & Fottová, 2009). Zde proto bylo provedeno měření za účelem popisu vlivu mlh na početní velikostní distribuce AA a aktivovaných nukleačních jader.\n\n Processes between atmospheric aerosol (AA) and clouds, source of large uncertainty in weather and climate changes estimations, were described on fogs at Milešovka, meteorological observatory of the Institute of Atmospheric Physics. For the description of the AA properties, online measurement of outdoor particle number size distribution (PNSD) in the size range 10 nm – 20 μm was conducted using SMPS and APS spectrometers. The sampling system consisted of a heated whole air inlet, and PM2.5 sampling head, being switched by an automatic valve. From the difference between PNSD sampled by whole air inlet and by PM2.5 inlet, PNSD of activated particles (aPNSD) was calculated. The aPNSDs differ with hydrometeor type and depend on air mass history, with a stronger influence on freezing fog AA. Structural characterization of fatty acids with triple bond and unusual double bond positions by HPLC/APCI-MS2

Fatty acid methyl esters (FAMEs) were analyzed by reversed-phase HPLC coupled to atmospheric pressure chemical ionization (APCI) mass spectrometry. Acetonitrile was used as mobile phase. The gas-phase reactions of acetonitrile and unsaturated FAMEs in the APCI source provided [M + C3H5N]+• adducts. Subsequent MS/MS analysis of these ions gave diagnostic fragments. The formation and fragmentation of the acetonitrile-related adducts were utilized for the structural characterization of FAME with isolated double bonds in (Z)-9,19-hexacosadienoic acid methyl ester, FAME with conjugated double bonds in (Z)-9,11-octadecadienoic acid methyl ester and FAME with a triple bond in 9-octadecynoic acid methyl ester. Rybářské hospodaření na malých vodárenských nádržích typu Hamry

Cílem metodiky je poskytnout správcům a uživatelům malých vodárenských nádrží typu Hamry na základě současné úrovně poznatků a realizovaného výzkumu při řešení projektu metodickou pomůcku pro optimalizace monitoringu kvality vody v těchto nádržích. Dále pak návrh metodiky monitoringu rybího společenstva a optimalizaci zásahů pro omezení nežádoucích druhů ryb a posílení stavu vhodných rybích druhů pro dosažení topdown efektu a pro udržení nebo zlepšení kvality vody v těchto vodárenských nádržích. V předložené metodice jsou shrnuty poznatky dosažené při sledování a hodnocení vybraných parametrů kvality vody, planktonních a rybích společenstev na vodárenských nádržích Hamry za poslední roky. Zároveň obsahuje vyhodnocení aplikovaných zásahů, zaměřených zejména na rybí obsádku. Jednalo se o odlovy nežádoucích druhů ryb a vysazování ryb dravých se snahou o optimalizaci složení rybího společenstva pro pozitivní ovlivnění kvality vody v nádrži běžné označované jako účelová rybí obsádka. Metodika zahrnuje doporučení pro monitoring kvality vody v nádrži, a to včetně monitoringu planktonního společenstva, monitoring ryb a doporučené manipulační zásahy v rámci rybí obsádky. Metodika poskytuje pokyny a informace pro optimalizaci monitoringu a zásahů pro malé vodárenské nádrže typu Hamry. The aim of the methodology is to provide managers and users of small water reservoirs Hamry type with methodological tools for optimization of the water quality monitoring based on the current level of knowledge and research. Furthermore, provides a design of a methodology for fish community monitoring and optimization of treatments to reduce unwanted fish species and strengthen the status of suitable fish species with the aim of achieving the topdown effect and maintaining or improving water quality in these water reservoirs. The presented methodology summarizes the findings obtained during the monitoring and evaluation of selected parameters of water quality, planktonic and fish communities in water supply reservoirs Hamry in recent years. It also contains an evaluation of applied treatments, focused mainly on fish stock. Unwanted fish species were caught and the predatory fish were stocked with the aim of optimizing the fish community composition and positively influencing the quality of the water in the reservoirs. The methodology contains recommendations for monitoring of the water quality in the reservoir, including monitoring of planktonic communities, monitoring of fish and recommended manipulation with fish stock. The methodology provides guidance and information for optimization of monitoring and treatments for small water supply reservoirs of the Hamry type.
