V této studii jsme podrobně zkoumali sezónní změny koncentrace cukrů, dikarboxylových kyselin a jejich oxidačních prekurzorů v jemném aerosolu na venkovském středoevropském pozadí. Cílem studie bylo identifikovat typické stopové prvky pro různé zdroje organického aerosolu a určit jejich podíl na organickém PM1. zjistili jsme zřetelný rozdíl mezi zimním a letním složením aerosolu PM1 na molekulární úrovni. Následně jsme identifikovali dva biogenní faktory (dominantní v létě), dva antropogenní faktory (hlavní příspěvek v zimě) a jeden faktor pozadí.\nPodrobnější informace bylo možné nalézt v publikaci Vodička et. al (2023). In this study, we have examined in detail the seasonal changes in the concentration of sugars, dicarboxylic acids and their oxidation precursors in fine aerosol at a rural Central European background site. The aim of the study was to identify typical tracers for different sources of organic aerosol and to determine their contributions to organic PM1.We found a clear difference between winter and summer PM1 aerosol composition at the molecular level. Subsequently, we identified two biogenic factors (dominant in summer), two anthropogenic factors (major contribution in winter) and one background factor.\nMore details could be found in Vodička et. al (2023). Open biomass burning (OBB) is a major source releasing large quantities of gaseous and particulate pollutants into the atmosphere, with global, regional and local impacts on air quality, public health and climate (Andreae, 2019, Chen et al., 2017). OBB aerosols mainly consist of carbonaceous matter, and a considerable amount of inorganic salts, which present distinctly different optical properties (Chen et al., 2017, Reid et al., 2005). In this study, we investigate the influence of OBB during the traditional Burning of the Witches (BoW), referred to in Czech as “Pálení čarodějnic”, on the physico-chemical properties of atmospheric aerosols collected at the National Atmospheric Observatory Košetice (NAOK). The BoW is a centuries-old pagan tradition very popular in the Czech Republic. Today, it is celebrated during the night from April 30 to May 1 by lighting bonfires made of woodpiles with fake witches. Atmospheric aerosols (AA) belong to short-lived climate forcers with high spatial and temporal variability. Although the radiative effects of AA are greatest on regional scales, changes in aerosol emissions can induce long-term global climate effects (Szopa et al., 2021). Thus, the measurement of aerosol properties is essential for better understanding of aerosol’s impact on health and climate. Although several international projects and platforms have been monitoring AA properties worldwide (GAW, WMO, ACTRIS, EMEP, EUCAARI, etc., Pandolfi et al., 2018), only PM10 or PM2.5 cutoff sizes have been used for measurements, and information about PM1 particles is missing. \nThe aim of this study is to present aerosol scattering properties with supporting data of absorption and particle number size distribution in PM1 in years 2018-2022 at peri-urban atmospheric site “Atmospheric Observations in LiLLE (ATOLL) in Lille, France. Hodnocení ekosystémových služeb (ES) se v průběhu posledních dvou dekád stalo důležitým koncepčním a modelovým nástrojem využívaným při kvantifikaci příspěvku jednotlivých ekosystémů (včetně globálního)\na biologické rozmanitosti k lidskému blahobytu. I když existuje celá řada koncepčních rámců a široká škála empirických aplikací, chybí podrobnější teoretická vymezení opírající se o ekologické a ekonomické teorie a rovněž\npřetrvává nejednoznačnost v definování pojmu ekosystémová služba. Ad hoc vymezení a obecná formulace tohoto pojmu je pak limitující v interpretaci dosažených výsledků, možnosti srovnání napříč studiemi, včetně provádění přenosu hodnot a funkcí hodnot, či při koncipování ekonomických nástrojů jakým jsou např. platby za ekosystémový služby. V tomto příspěvku proto diskutujeme pojmy jako je konečný produkt, meziprodukt, ekologická produkční funkce a funkce užitku, tedy pojmy, které jsou určující pro korektní vymezení ES z pohledu ekonomické teorie. The assessment of ecosystem services (ES) has become an important concept during the last two decades and a model tool used to quantify the contribution of individual ecosystems (including the global one)\nand biodiversity to human well-being. Although there are a number of conceptual frameworks and a wide variety empirical applications, there is a lack of more detailed theoretical definitions based on ecological and economic theories as well ambiguity persists in defining the term ecosystem service. Ad hoc delineation and general formulation of this the concept is then limiting in the interpretation of the achieved results, the possibility of comparison across studies, including implementation transfer of values and functions of values, or when designing economic instruments such as payments for ecosystem services. Therefore, in this paper we discuss terms such as final product, intermediate product,\necological production function and utility function, i.e. concepts that are decisive for the correct definition of ES from the point of view economic theory. Náplní první části kurzu bude úvod do proteomiky s cílem ukázat současné metody a nástroje použitelné k akvizici a vyhodnocení proteomických dat. V první části probereme ve zkrácené formě všechny kroky experimentálního postupu od 1) designu experimentu tak, aby byl vhodný pro následnou LC/MS analýzu, 2) přípravy vzorku na příkladech imunoprecipitace a komplexního lyzátu až po 3) možnosti měření pomocí Data Dependent nebo Data Independent acquisition. Součástí kurzu bude i kritické hodnocení kvality naměřených dat. Při vyhodnocení se zaměříme na možnosti zpracování dat ve freeware SW typu MaxQuant a DIAnn. Rovněž Vás stručně seznámíme s komerčními programy – Proteome Discoverer, PEAKS, Spectronaut.Druhá část kurzu bude zahrnovat úvod do statistického zpracování kvantitativních proteomických dat v softwaru Perseus. Důraz bude kladen na pochopení důležitých statistických pojmů (distribuce dat, t-test, ANOVA, p-value, FDR, atd.) a porozumění všem nezbytným krokům statistické analýzy, jako je filtrace dat, normalizace dat, statistické testování, apod. K dispozici budou modelová data, která budou účastníci v rámci kurzu zpracovávat. The first part of the course will be an introduction to proteomics with the aim to show current methods and tools used for proteomic data acquisition and evaluation. In the first part, we will discuss in abbreviated form all steps of the experimental procedure from 1) designing the experiment to be suitable for subsequent LC/MS analysis, 2) sample preparation using examples of immunoprecipitation and complex lysate, to 3) measurement options using Data Dependent or Data Independent acquisition. The course will also include a critical evaluation of the quality of the measured data. The evaluation will focus on the possibilities of data processing in freeware software such as MaxQuant and DIAnn. We will also briefly introduce you to commercial programs - Proteome Discoverer, PEAKS, Spectronaut.The second part of the course will include an introduction to statistical processing of quantitative proteomic data in Perseus software. Emphasis will be placed on understanding important statistical concepts (data distribution, t-test, ANOVA, p-value, FDR, etc.) and understanding all necessary steps of statistical analysis such as data filtering, data normalization, statistical testing, etc. There will be model data that participants will process during the course. Simultaneous online determination of nitric acid and nitrates in ambient air

The innovated method for the simultaneous online determination of HNO3/NO3- in ambient air was developed. Due to high sensitivity of CLD (LOD = 5.1 nM in water solution, 3 s/n) no preconcentration method is required. The online methods were compared with the results obtained with the offline method based on sampling using filters and dry denuders (DD) covered by 0.5% NaF with subsequent analysis by ion chromatography (model ICS-2100, Dionex, USA) and capillary electrophoresis (model Agilent 7100, Agilent Technologies, USA). The aim of this study is to characterize the light scattering properties of aerosols at the rural background site National Atmospheric\nObservatory Košetice (NAOK; 49°34’20.787”N, 15°4’48.155”E) in Central Europe, namely the coefficients of total light scattering (σsp) and backscattering (σbsp), the Ångström exponent (SAE), the backscattering ratio (b), and the asymmetry factor (g), with special attention to the influence of meteorology and aerosol size distribution on these properties. We measured σ𝑠𝑝 and σ𝑏𝑠𝑝 at three wavelengths (450, 550, and 700 nm) using the Integrating Nephelometer TSI 3563 (PM10 inlet).
