Genetic analysis of dicyemid infrapopulations suggests sexual reproduction and host colonization by multiple individuals is common

This study explores the infrapopulation structure of Dicyema moschatum in its host Eledone moschata with the use of microsatellite markers. In order to assess the infrapopulation diversity, set of eight microsatellite markers was selected and respective oligonucleotides were designed for this population genetics study. Fragmentation method was used to analyze various microsatellite markers and the results were analyzed in bioinformatic programs that focus on standard population genetic metrics. Obtained results proved to provide new information about D. moschatum mating system and infection strategy. Práce vychází z institucionálního přístupu ke studiu muzeálního fenoménu v minulosti. Soustředí se na vykreslení jednoho konkrétního místa ve světě českých muzeí v době státního socialismu a jeho podoby za Pražského jara a v prvních letech normalizace. Tímto místem je někdejší Oblastní/Okresní a dnes Prácheňské muzeum v Písku, které právě tehdy napsalo jednu z nejzajímavějších kapitol své historie. Cílem práce je vymezit a popsat obsahy dokumentační a prezentační činnosti píseckého muzea v předmětném období. Zohledňována je přitom dobová normativní a metodická literatura i vztah této činnosti k dobové politice a ideologii. Práce se opírá o rozbor zejména dobových výročních zpráv muzea, různých příležitostných tisků a publikací, novinových zpráv, muzejní dokumentace a evidence, ale také příslušné dobové odborné a metodické literatury. Záměrem je přiblížit nejen osudy jednoho konkrétního muzea, ale prostřednictvím případové studie se vyslovit k roli a místu českých muzeí ve společnosti, která sebe samu označovala za socialistickou. Předmětem zájmu v této práci je především oblast těch aktivit muzea, které souvisely s plněním vymezené úlohy a funkce. Stranou až na pár výjimek zůstaly otázky řízení a kontroly muzea ze strany nadřízených orgánů, recepce muzea a jeho aktivit širší veřejností, stejně jako vztah muzea a jeho produkce a činnosti k naplňování muzeálního fenoménu. The thesis is based on an institutional approach to the study of museum phenomena in the past. It focuses on depicting one specific place in the landscape of Czech museums during the period of state socialism and its appearance during the Prague Spring and in the first years of normalization. This place is the former Regional / District and today Prácheň Museum in Písek, which just then wrote one of the most interesting chapters in its history. The aim of the work is to define and describe the contents of documentation and presentation activities of the Písek Museum. The contemporary normative and methodological literature as well as the relation of this activity to contemporary politics and ideology are taken into account. The work is based on the analysis of the period's annual reports of the museum, various occasional prints and publications, newspaper reports, museum documentation and records, but also the relevant period and professional literature. The intention is not only to approach the fate of one particular museum, but also to comment on the role and place of Czech museums in a society that called itself socialist through a case study. The subject of interest in this work is mainly the area of those activities of the museum, which were related to the fulfillment of the defined task and function. With a few exceptions, the issues of management and control of the museum by superior bodies, the reception of the museum and its activities by the general public, as well as the relationship of the museum and its production and activities to the fulfillment of the museum phenomenon remained aside. Disentangling the nonlinear effects of habitat complexity on functional responses

Habitat structural complexity modifies trophic interactions by providing refuges and altering predator and prey behavior. The quality and intensity of predator prey interactions can be described by functional responses, that is by the relationship between the number of preys consumed by a predator and initial prey density. We combine two complementary methods of functional response estimation to examine the effects of habitat complexity on the predation of Aeshna cyanea dragonfly larva on Chaoborus obscuripes glassworm larvae. We show that habitat complexity has nonlinear effects on functional response parameters (attack rate and handling time), despite the prevalence of stepwise responses reported in literature. Our results suggest that detailed understanding of habitat complexity effects on food webs requires complementary experimental approaches across a gradient of habitat complexity and should also consider predator and prey behavior. Tato práce odhaluje výsledky NGS analýzy mykobiomu z FFPE vzorků pacientů s Crohnovo chorobou, ulcerózní kolitidou a zdravých kontrol. Rozděluje fungální rody do tří experimentálních skupin, určuje jejich příslušnost ke zdraví a nemoci, navrhuje interakce mezi fungálními rody a ukazuje specifické fungální rody související s určitou diagnózou. This thesis reveals results of NGS mycobiome analysis from formalin-fixed, paraffin-embedded samples of Crohn's disease, ulcerative colitis patients and healthy controls, divides fungal genera into three experimental groups, determines their affiliation to health and disease, suggests fungal genera interactions, and shows specific fungal genera related to the diagnosis. Studium prevalence a subtypové diverzity střevního prvoka rodu Blastocystis sp. ve zdravé lidské populaci v České republice

Blastocystis sp. je běžný střevní prvok kolonizující lidské střevo. Jeho role v ekosystému lidského střeva zůstává nejasná. Zde jsme se zaměřili na rozšíření znalostí o epidemiologii Blastocystis sp. ve střevě zdravých lidí v České republice, včetně distribuce jeho podtypů, korelace mezi jeho výskytem a několika faktory, jako je životní styl, kontakt se zvířaty, věk a pohlaví. Celkem bylo získáno 288 vzorků stolice od asymptomatických dobrovolníků a 136 vzorků od zvířat, se kterými byli lidé v častém kontaktu. Všechny vzorky byly vyšetřeny paralelně pomocí PCR a xenické in vitro kultury. Blastocystis sp. is a common intestinal protist colonizing the human intestine. Its role in the human gut ecosystem remains unclear. Here, we aimed to expand the knowledge of the epidemiology of Blastocystis sp. in the gut-healthy humans in the Czech Republic, including the distribution of its subtypes, the correlation between its occurrence and several factors such as lifestyle, contact with animals, age, and sex. A total of 288 stool samples were obtained from asymptomatic individuals over the entire age-range and 136 samples from animals with which the volunteers were in frequent contact. All samples were examined in parallel by PCR and xenic in vitro culture. Climate change may accelerate the Urban Heat Island (UHI) effect with many consequences for the well-being of human populations in cities. Green and blue infrastructures (GBI) are presented as a way to mitigate the UHI effect. In our study, the influence of GBI (primarily less studied types) on surface temperature (ST) was tested using a thermal camera. Various types of GBI (e. g. tree shade and lawn) and their combination were compared. Climate change may accelerate the Urban Heat Island (UHI) effect with many consequences for the well-being of human populations in cities. Green and blue infrastructures (GBI) are presented as a way to mitigate the UHI effect. Počítačová mikro-tomografie (mikro-CT) je výjimečná zobrazovací technika schopná vizualizace jak externí, tak interní morfologie členovců, jako jsou klíšťata. Hlavním nedostatkem mikro-CT je potřeba dodatečného kontrastování měkkých biologických tkání před jejich vizualizací. Standardně používanými kontrastujícími metodami je v případě klíšťat vysoušení a máčení ve vodném roztoku obsahujícím prvek s vysokým atomárním číslem. Obě tyto metody jsou ovšem zdrojem výrazné deformace a smršťování vzorků. Cílem této studie bylo vyvinout nový způsob kontrastování klíšťat, který by nedostatky současně používaných technik překonal. Podle našich zjištění je možné dospělé samice Ixodes ricinus vykrmených in vitro na krvi obohacené o zlaté nanočástice s polyethylenovým obalem skenovat s vynikajícím kontrastem. Díky využití popsané metody založené na hematofágním způsobu života klíšťat jsme byli schopni pozorovat lumen a epitelium střev, a rektální vak v jejich přirozeném tvaru. Vyjma toho jsme byli schopní spočítat objem střeva. Micro-computed tomography (micro-CT) is an exceptional non-destructive imaging modality, which can be used to visualize both external and internal morphology of arthropods like ticks. The major micro-CT drawback is the soft tissues visualization, which require pre-examination contrasting steps. The commonly employed tick contrasting methods are drying and immersion in a solution containing a high-atomic number element, which both cause major deformation and shrinkage of examined specimens. The aim of this study was to develop a new tick contrasting approach to bypass these limitations. We show that adult Ixodes ricinus females engorged in vitro on blood enriched with gold nanoparticles with polyethylene glycol coating can be scanned with excellent contrast. By utilizing this technique based on the hematophagous nature of the ticks, we were able to observe midgut lumen, midgut epithelium and rectal sac in their natural state. In addition, we were able to precisely calculate the midgut volume. Tato rigorózní práce se zabývá populační strukturou pěti druhů hnědásků - hnědáska jitrocelového, hnědáska kostkovaného, hnědáska květelového, hnědáska podunajského a hnědáska černýšového, sdílící dvě lokality Národního parku Podyjí - Havranické vřesoviště a Mašovická střelnice. Obě tyto lokality jsou v současnosti ohroženy postupnou sukcesí a zarůstáním. Díky tomu byly vybrány pro zavedení volné pastvy velkých herbivorů (Exmoorští pony). Tato práce popisuje demografickou strukturu, mobilitu a habitatové nároky dospělců pěti druhů hnědásků před zavedením volné pastvy velkých herbivorů. This thesis focuses on the population structure of five frittilary butterfly species - Melitaea athalia, M. cinxia, M. didyma, M. britomartis and M. aurelia - co-occurring on two xeric grassland localities of the Podyjí National Park, Czech Republic - Havranické vřesoviště and Mašovická střelnice. Recently, both of these sites are threatened by succession and degradation processes. Due to that, they have been chosen for whole season grazing by a large herbivore (Exmoor pony). This study describes adult demography structure, adult mobility and adult habitat requirements of the five butterflies prior to the grazing management establishment.
