Voltammetric Determination of Psilocin and Psilocybin at Carbon-fiber PLA Electrode

Psilocin and psilocybin are two neuroactive drugs. They can be used in experimental treatment in psychiatry, moreover, also as illicit drugs to achieve hallucinations and relaxation. Easy access to these two alkaloids indicates that their monitoring in the environment and the body fluids is necessary. Electrochemistry offers fast, simple, and easy methods of their determination using laboratory-manufactured electrodes from 3D printed carbon-fiber polylactic materials. This technique of electrode fabrication can be characterized by many benefits, e.g., desired shape or diameter of the electrode, well-developed voltammetric signals, and low background currents. Analysis of the Hydrated ions Clusters Impact on the Liquid/Liquid Interface

Open circuit potential (OCP) measurements of the partition of tetrapentylammonium chloride (TPeACl) at the water/1,2-dichloroethane (DCE) interface reveal two remarkable processes. The first one is characterized by a decay of the OCP from its expected equilibrium value. This process has been ascribed to the extraction of the salt driven by its accumulation in the hydrated salt clusters in the DCE phase. The second process is characterized by the short-term potential spikes overlapping the long-term OCP records. These spikes are assumed to be associated with the collision and fusion of the hydrated salt clusters produced by the former process with the water/DCE interface. Fused deposition modeling 3D printing employing high-resolution extrusion nozzles and specialized manufacturing protocols have recently attracted attention as an inexpensive tool to create microfluidic devices. In this work, we have applied such an approach to create measurement cells with integrated microchannels of systematically varied dimensions. The real width of channels was inspected by two techniques, namely (A) impedance spectroscopy employing an aqueous sensing electrolyte and (B) optical microscopic imaging of silicone imprints hardened in the interior of printed structures. Both techniques have demonstrated that fused deposition modeling 3D printing enables precise manufacturing of channels with truly microfluidic dimensions. Obsah železa a kvalita organických látek jako faktor určující stabilizaci uhlíku a koloběh živin ve sladkovodních ekosystémech.

Studie ukazuje, jak geochemické vlastnosti povodí, reprezentované obsahem Fe a hodnotami pH a s tím související kvalitou organických látek v půdách, ovlivňuji stabilizaci organického uhlíku a živin v sedimentech mělkých nádrží. V rámci prezentované studie bylo sledováno 12 rybníků z různých lokalit České republiky. Rybníky byly rozděleny na základě geologie povodí do dvou skupin i) povodí s kyselými půdami (pH 5,1 - 6,3) bohatými na železo (H_Fe) a ii) povodí s neutrálními půdami (pH = 6,8–7,6) a nízkým obsahem železa (L_Fe). Ve sledovaných rybnících byly porovnávaný parametry vodního sloupce, pórové vody a extrakce sedimentů z hlediska stabilizace organické hmoty a navazujících živinových koloběhů. Bylo ukázáno, jak vyšší koncentrace hydratovaných oxidů Fe, spolu s aromatickým charakterem organických látek přispívá k celkové stabilizaci sedimentu. Naopak nízké koncentrace Fe byly doprovázeny nižší mineralizaci sediment a vyšší rizikovosti pro vodní ekosystém z hlediska uvolňovaní živin nebo rozvoje anoxii. The effect of watershed geochemistry, in terms of soil pH, Fe concentration and organic matter quality, on the stability of organic carbon and nutrients level in freshwater sediments of shallow lakes was studied. Twelve fishponds were divided into two groups based on the typical soil pH: i) watershed with acidic soils (pH 5.1 – 6.3) rich in Fe called H_Fe and ii) watershed with neutral pH soils (pH 6.8 – 7.6) poor in Fe called L_Fe. The parameters of the water column, pore water and sediment chemical extractions were compared. A higher concentration of hydrated Fe oxides, together with the aromatic character of organic substances were shown to highly contribute to the overall stabilization of the sediment. Conversely, low Fe concentrations were accompanied by lower mineralization of sediments and a higher risk for the aquatic ecosystem in terms of nutrient release or the development of anoxic conditions. Radon v jezeře - odhad množství podzemní vody.

V roce 2020 byla realizována bilanční studie aktivity radonu v Plešném jezeře a jeho přítocích s cílem zpřesnění odhadu zdrojů podzemní vody. Bilance je založena na kvantifikaci zdrojů, monitorování množství vody odtékající z jezera a koncentrace olova ve vrchních vrstvách sedimentů. A balance study of radon activity in Lake Plešný and its tributaries was carried out in 2020 to refine the estimate of groundwater resources. The balance is based on quantification of sources, monitoring of the amount of water draining from the lake, and lead concentrations in the overlying sediments. Těžiště kurzu bude v MALDI hmotnostním zobrazování. V první části budou shrnuty poznatky a postupy přípravy vzorků, nanášení matrice, měření a vyhodnocováním MALDI MSI analýz. Následovat bude blok věnovaný praktickým zkušenostem MALDI MSI lipidů, metabolitů a dalších analytů na tkáňových řezech a jiných planárních površích. Závěr bude věnován molekulovému zobrazování s využitím dalších používaných ionizačních technik (DESI, DAPPI, LAESI, atd.). The course will focus on MALDI mass spectrometry imaging. Knowledge and procedures of sample preparation, matrix application, measurement, and evaluation of MALDI MSI will be summarized. The first part will be followed by a block dedicated to the practical experience of MALDI MSI of lipids,metabolites, and other analytes on tissue sections and other planar surfaces.
