Vizualization of large biological tissue specimens using confocal laser scanning microscopy

In biology there is often necessary to visualize a biological specimen which size is greater when compared with the field of view of a used optical acquisition instrument. The visualization of such the specimen can be achieved by volume reconstruction. We study biological specimens by using a confocal laser scanning microscope which is able to capture a digital volume representation of the specimen. We investigate great specimens containing, for example, a human tooth pulp, an epithelial layer and a vascular bed of chick embryonic gut or chick embryonic kidneys Objemová rekonstrukce je metoda sloužící k vizualizaci trojrozměrné struktury biologických vzorků, jejichž velikost může být větší než je horizontální či axiální zorné pole snímacího systému (v našem případě laserového konfokálního mikroskopu). Velké vzorky je nutné nařezat na tenké fyzické řezy. Pak se snímají všechny sousedící a překrývající se objemové obrazy v rámci jednotlivých fyzických řezů (kompenzace omezeného horizontálního zorného pole). Volume reconstruction of large biological tissue specimens

Volume reconstruction is a technique for visualization of a biological specimen which is greater than the field of view of a used optical instrument - a confocal laser scanning microscope in our case. The first step of volume reconstruction is acquisition of sets of digital volume images (spatial tiles which overlap) from all studied physical slices. The second step is horizontal merging of overlapping spatial tiles of the same physical slice (mosaicking). The third reconstruction step is vertical merging of digital volumes of successive physical slices of the specimen. The resulting large digital volumes are visualized using a VolumePro hardware board that offers real-time 3D volume rendering. In this paper we show a reconstruction of a chick embryonic kidney Objemová rekonstrukce slouží k trojrozměrné vizualizaci biologických vzorků, jejichž velikost může být větší než je zorné pole snímacího systému (laserového konfokálního mikroskopu).Velké vzorky je nutné nařezat na tenké fyzické řezy. Následně se snímají všechny sousedící objemové obrazy v rámci jednotlivých fyzických řezů (kompenzace omezeného horizontálního zorného pole). Takové obrazy se dále získají pro všechny sériové fyzické řezy (kompenzace omezeného axiálního zorného pole).První krok objemové rekonstrukce představuje složení překrývajících se sousedních zorných polí téhož fyzického řezu. Složené objemové obrazy fyzických řezů je pak nutno složit ve výsledný objemový obraz reprezentující celý biologický vzorek. Stereological metod for the estimation of chloroplast thylakoid surface area

The stereological method using “local vertical windows” applied to the estimation of thylakoid surface area in the chloroplast volume, based on evaluation of chloroplast electron micrographs is presented. Digital volume reconstruction is a technique for rendering and visualization of a biological specimen which is greater than the field of view of a used optical instrument - a confocal laser scanning microscope in our case. Prior to the volume reconstruction, large biological specimens are cut to thin physical slices. The first step of volume reconstruction is acquisition of sets of digital volume images (spatial tiles which overlap) from all studied physical slices. The second step is composition of neighbouring spatial tiles of the same physical slice. The third reconstruction step is registration and merging of digital volumes of neighbouring physical slices of the specimen. The resulting large digital volumes are rendered and visualized using a VolumePro hardware board that offers real-time 3D volume rendering. Schopnost adaptivně reagovat na proměnlivé podmínky dynamicky se měnícího prostředí je vlastností, kterou nacházíme u přirozených i umělých "inteligentních systémů". Naše práce porovnává učení se živých (potkan) a neživých (autonomní mobilní robot řízený evolučním systémem) objektů ve stejném typu prostorových úloh. Výsledky získané "výcvikem" potkanů resp. robota budou pojednány ve zvláštních kapitolách Ability to adapt to a continuously changing environment is inherent both to natural and artificial "inteligent systems". Analysis of microcracks in renal cortex and medulla

Cortex and medula were both damaged after drop shatter test, even in those areas of kidneys, that were free of macroscopic cracks on the surface. The ruptures propagated preferentially through the interstitial connective tissue.
