Paratrypanosoma is a novel early-branching trypanosomatid

The kinetoplastids are a widespread and important group of single-celled eukaryotes, many of which are devastating parasites of animals, including humans. We have discovered a new insect trypanosomatid in the gut of Culex pipiens mosquitoes. Glyceraldehyde-3-phosphate dehydrogenase- and SSU rRNA-based phylogenetic analyses show this parasite to constitute a distinct branch between the free-living Bodo saltans and the obligatory parasitic clades represented by the genus Trypanosoma and other trypanosomatids. From draft genome sequence data, we identified 114 protein genes shared among the new flagellate, 15 trypanosomatid species, B. saltans, and the heterolobosean Naegleria gruberi, as well as 129 protein genes shared with the basal kinetoplastid Perkinsela sp. Individual protein phylogenies together with analyses of concatenated alignments show that the new species, here named Paratrypanosoma confusum n. gen., n. sp., branches with very high support at the base of the family Trypanosomatidae. P. confusum thus represents a long-sought-after missing link between the ancestral free-living bodonids and the derived parasitic trypanosomatids. Further analysis of the P. confusum genome should provide insight into the emergence of parasitism in the medically important trypanosomatids. Predicting asp and pikeperch recruitment in a riverine redervoir.

Recruitment of two species, asp (Leuciscus aspius) and pikeperch (Sander lucioperca), in a riverine reservoir was studied using a novel statistical approach. Both species are piscivorous and are stocked into reservoirs for biomanipulative purposes to reduce planktivore species. Long-term data series were used, but the number of potential predictors was high. Therefore, a novel informative statistical approach based on dimension reduction methods was applied. Quality of outputs was driven by sampling methods. Main factors affecting asp recruitment were zooplankton abundance, predator density and temperature. In terms of pikeperch fry measured with seine and trawls, the number of predators was the only important factor. Gillnets underestimate small fish and the data were unsuitable for statistical modelling. Orientace ryb má důležité následky pro odhadování jejich skutečné velikosti z horizontálních akustických záznamů. Cílem této studie bylo ověřit předpoklad o náhodné orientaci ryb v jezerních částech nádrže Římov během dne a porovnat distibuce orientace ryb mezi jezerní a přítokovou částí. The orientation of fish has important consequences for estimating their true size from horizontal acoustic records. The aim of this study was to verify the assumption of randomly orientated fish in the lacustrine zone of the canyon-shaped Římov reservoir during daytime and to compare distributions of fish orientation between the lacustrine and tributary (riverine) zones. Raman Microspectroscopy of Individual Algal Cells: Sensing Unsaturation of Storage Lipids in vivo

We have demonstrated the potential of Raman microspectroscopy for the fast and spatially resolved characterization of the composition of selected intracellular regions in individual living cells of three algal species. In particular, we have focused on lipid storage bodies and quantified the degree of unsaturation of algal lipids (iodine value). To verify our results, we developed a method for extraction of storage lipids from highly resistant Trachidiscus minutus cells and determined the fatty acid composition using GC/MS. We have found Raman microspectroscopy to be a fast, versatile, and virtually non-invasive tool for evaluating the potential of different algal species for algal lipid engineering and industry. The prevalence and diversity of Cryptosporidium spp., microsporidia and Giardia intestinalis in orangutans (Pongo abelii and Pongo pygmaeus) at seven different sites on Sumatra and Kalimantan were described using molecular methods. Out of the 298 examined animals, 33 were positive for the tested parasites at least once during the study. Microsporidia(Encephalitozoon spp. and Enterocytozoon bieneusi) were detected in orangutans for the first time and molecular data were obtained on Cryptosporidium spp. and G. intestinalis in this host. Species diversity of Plagiorchis Lühe, 1899 (Digenea: Plagiorchiidae) in lymnaeid snails from freshwater ecosystems in central Europe revealed by molecules and morphology

Tato studie používá molekulární a morfologické přístupy pro identifikaci morfologicky podobných larválních stádií (cerkárií) druhů Plagiorchis (Digenea: Plagiorchiidae), kteří parazitují plže čeledi Lymnaeidae ve sladkovodních ekosystémech střední Evropy. Pět morfologicky homogenních a geneticky odlišných linií druhů Plagiorchis bylo identifikováno spojením molekulárních dat pro mitochondriální gen cox1 spolu s detailními morfologickými údaji. Fylogenetické a srovnávací sekvenční analýzy využívající částečné 28S rDNA a ITS1-5.8S-ITS2 sekvence umožnily molekulární identifikaci tří druhů (P. elegans, P. maculosus a P. koreanus) prostřednictvím shody sekvencí larválních stádií a dospělců těchto digenetických motolic. Součástí předkládané práce je klíč k identifikaci cerkárií rodu Plagiorchis parazitujících populace plovatkovitých plžů ve střední Evropě. This study applies molecular and morphological approaches addressing the identification of morphologically similar larval stages (cercariae) of Plagiorchis spp. (Digenea: Plagiorchiidae) parasitising lymnaeid snail populations in the freshwater ecosystems of central Europe. Five morphologically homogeneous and genetically distinct lineages of Plagiorchis spp. were identified via matching molecular data for the mitochondrial cox1 gene with detailed morphometric data. Phylogenetic and comparative sequence analyses using partial 28S rDNA and ITS1-5.8S-ITS2 sequences allowed molecular identification of three species (P. elegans, P. maculosus and P. koreanus) via matching sequences from larval and adult digenean stages. A key for the identification of the cercariae of Plagiorchis spp. parasitising lymnaeid populations in central Europe is provided. Spatial heterogeneity and seasonal succession of phytoplankton along the longitudinal gradient in a eutrophic reservoir

Spatial distribution and seasonal succesion of phytoplankton along the longitudinal axis of a eutrophic Římov reservoir (Czech Republic) was investigated from April to October 2007, at 1?2 week intervals. Phytoplankton spatial heterogeneity was the most apparent during the summer reflecting a pronounced gradient of environmental parameters from the river inflow to the dam. The riverine zone differed markedly from downstream parts of the reservoir, being dominated by functional groups D and J (large cryptophytes and colonial diatoms). While a dense cyanobacterial bloom (groups H1 and M) occurred in the transition zone, considerably lower biomass of desmids (Group N) was found at lacustrine zone at the same time. A sudden flood event considerably altered nutrient and light availability, and later even resulted in cyanobacterial dominance in the whole reservoir. Tato studie přináší nový symbiotický model ve skupině Hippoboscidae vhodný k porovnávání s dobře charakterizovaným symbiotickým systémem ve skupině Glossinidae. Distribuce symbiontů byla odhalena pomocí elektronové mikroskopie a jejich evoluční původ byl potvrzen rekonstrukcí jejich fylogeneze. Pomocí genomických dat byl zkoumám jejich metabolický přínos hostiteli a osvětlen metabolismus B vitamínů. This study provides a new comparative model of symbiosis in sheep ked to well-characterized tsetse fly symbiotic system. Distribution of symbiotic associates was revealed by transmission electron microscopy and their evolutionary origin was confirmed by reconstruction of their phylogenies. Using genomic data, it explores their metabolic contribution to host and sheds light on B-vitamin metabolism.
