(A)sexual Life of Liverworts

2015 - Czech This thesis comprises of two published papers and one accepted manuscript, focused on various aspects of liverwort reproduction. Treated aspects include patterns of asexual reproduction, sex ratio and sex-specic pattern in vegetative growth, and patterns of genetic variation and spatial genetic structure of populations differing in availability of substrate on localities and the population connectivity, and consequently in size, density, and prevailing reproductive mode. These characteristics were studied on representatives of the family Scapaniaceae s.l., belonging to the largest liverwort order Jungermanniales. The results showed that asexual propagules were formed and present in course of the whole growing season and can be considered as a sufficient substitution for sexual reproduction. In contrast with the female-biased sex ratio observed earlier in most dioicous bryophytes, unexpectedly high male-biased sex ratio was observed in the aquatic liverwort, which was speculated to represent a strategy to overcome sperm dilution in aquatic environment. In addition, no size differences between female and male shoots were detected, although the evidence for higher cost of sexual reproduction in females was found. The study of population genetic structure has shown that even small and predominantly asexually reproducing populations are important sources of genetic variation. However, we were able to demonstrate notably low levels of gene flow among populations where habitat fragmentation poses a significant barrier to dispersal of diaspores. The fine scale study of spatial genetic structure revealed a strong aggregation of genotypes, particularly in smaller populations, and at the same time showed that asexual reproduction is an efficient mean of maintaining the populations at not only the short distances, given the spatial extent of clones spanning dozens of meters. Reading experiences of Czech authoress Karolina Světlá

2015 - Czech The aimof this thesis is a research of reading experiences of Karolina Světlá (1830- 1899), one of the significant Czech authoress and representative of women´s emancipation in the second half of the 19. century. One of the main sources for this research are published editions of personal correspondence and memoirs, where wecan find particular references to home and foreign-language literature and affirmative or negative appreciation single works or writers, which she knows from her reading, follow up the changes of them and focus on the parts talking about a function of reading in the girl´s education and proper literature for women. Another important source is a personal archival file of this authoress, which is stored in the museum in Český Dub in northern Bohemia. Here we can discover, except 366 books from possession of Karolina Světlá and her husband, some personal hand-writing documents, which are illustrating her reading preferences (extracts from some books or bills from different bookstores). This thesis would like to create a reading biography of followed authoress, show the atmosphere and Czech literary world in the second half of the 19. century from the alternative point of view and reveal the way of thinking and ideas of important person with many cultural and social activities. Předmětem disertační práce je výzkum čtenářských zkušeností Karoliny Světlé (1830- 1899), výrazné představitelky české literatury a ženského emancipačního hnutí druhé poloviny 19. století. Jedním z hlavních pramenů pro tento výzkum jsou její vydané memoárové texty a korespondence, kde můžeme nalézt konkrétní zmínky o jednotlivých autorech či autorkách i přečtených dílech, sledovat názory na ně a jejich proměny a zaměřit se na pasáže o četbě jako důležité součásti dívčího vzdělávání a výběru vhodné četby pro ženy. Druhým bohatým zdrojem informací k danému tématu je osobní fond Karoliny Světlé ve sbírkách Podještědského muzea v Českém Dubu, kde kromě souboru 366 svazků z majetku manželů Mužákových nalezneme také rukopisné památky dokládající její čtenářské preference (výpisky z četby, sešit opsaných německých básní nebo dochované knihkupecké účty). Cílem práce je vytvoření čtenářské biografie sledované autorky a prostřednictvím jejích čtenářských zkušeností a názorů na přečtené texty i konkrétní autory či funkci četby ve vzdělávacím procesu ukázat českou literární scénu a atmosféru druhé poloviny 19. století z jiného úhlu pohledu a nahlédnout do myšlenkového světa významné, kulturně i společensky aktivní osobnosti své doby. Holders of economical politics on the regional level, their role and possibilities in the optimal adjustment of old age pensions scheme

2015 - Czech Ph.D. thesis called "Holders of economical politics on the regional level, their role and possibilities in the optimal adjustment of old age pensions scheme" deals with the possibilities of a general solution for pension insurance in the scope of the region. The possibilities of the regional pension insurance system in the real conditions of the Czech Republic are described and analyzed, as it is difficult to transfer pension insurance systems from one country to another as a whole. Possible roles and possibilities of the region when dealing with the problem of old age pension insurance were defined on the basis of performed analyses. A specific outcome of this project represents the definition of conditions which must be satisfied if the public fund of Regional old age pension system should function. Its operation in the framework of second pillar of pension system seems like the optimal solution. The specific solution contains also the mathematical model of functioning of the regional pension scheme under defined conditions, which simulates possible development of the fund and the amount of old age pension received by the contributor provided he satisfies the defined requirements for entitlement to old age pension. Předložená disertační práce nazvaná "Nositelé hospodářské politiky na regionální úrovni, jejich role a možnosti v optimálním nastavení důchodového zabezpečení" se zabývá možnostmi plošného řešení důchodového zabezpečení v rámci regionu. Vzhledem k obtížné přenositelnosti důchodových systémů jako celku jsou mož-nosti regionálního důchodového zabezpečení popisovány a analyzovány v rámci reál- ných podmínek České republiky. Na základě provedených rozborů a analýz byly definovány možné role a možnosti regionu při řešení problematiky zajištění občanů ve staří. Konkrétním výsledkem je pak definování podmínek fungování veřejného fondu regionálního důchodového zabezpečení, přičemž jako optimální se jeví jeho působení v rámci druhého pilíře důchodového zabezpečení. 2014 - Czech The doctrine of predestination in Western theology had for centuries been burdened by the dualistic conception of Saint Augustine. The bishop of Hippo held that it is God?s intention to reveal His mercy on a small number of those predestined to salvation and His justice on a far larger mass of rightly damned sinners. He came to be convinced that salvation is an utterly undeserved gift and consequently he negated the importance of human freedom. His interpretation would rather give rise to anxiety than the hope Scripture speaks about. Vladimír Boublík dared to oppose Augustine?s authority and presented an entirely different solution based on modern exegesis of the Pauline epistles. He defined predestination as an irreversible divine decision the content of which is the participation of all humanity on divine life in Christ. The present paper shows that Boublík?s theology stems from not just academic, but above all existential search for the answer to the question of human destiny marked with sin, suffering and death. It is already apparent on the pages of the spiritual diary the author kept during his studies. All his writing shows the intention to address his contemporaries and lead them to Christ who stands in the centre of the author?s thinking. This paper attempts to follow up on the work of the Czech exile theologian. It presents his conception of predestination and further develops it in some respects. Besides other things, it throws light on the previously unclarified problem of Christ?s primacy in predestination. Vladimír Boublík?s thesis is confronted with objections that have been raised against it and its content is compared with the content of today?s contribution to the topic. Nauka o předurčení byla v západní teologii po staletí zatížena dualistickou koncepcí svatého Augustina. Biskup z Hippo se domníval, že Božím záměrem je zjevit své milosrdenství na malém počtu předurčených ke spáse a svou spravedlnost na nepoměrně větší mase po právu zavržených hříšníků. Nabyl přesvědčení, že spása je naprosto nezasloužený dar a následně popřel význam lidské svobody. Jeho výklad v člověku vzbuzoval spíše úzkost než naději, o které hovoří Písmo. Vladimír Boublík se odvážil postavit Augustinově autoritě a předložil docela odlišné řešení, opírající se o novodobou exegezi pavlovských listů. Předurčení definoval jako neodvolatelné Boží rozhodnutí, jehož obsahem je účast celého lidstva na Božím životě v Kristu. Přítomná práce ukazuje, že Boublíkova teologie vychází nikoliv jen z akademického, ale především z bytostného hledání odpovědi na otázku lidského osudu, poznamenaného hříchem, utrpením a smrtí. Rýsuje se již na stránkách duchovního deníku, který si autor vedl v době svých studií. Celá jeho tvorba prozrazuje záměr oslovit současného člověka a vést ho ke Kristu, který stojí ve středu autorova uvažování. Tato práce je pokusem na českého exilového teologa navázat. Představuje jeho pojetí předurčení a v některých bodech ho dále rozvíjí. Mimo jiné vnáší světlo do dříve nevyjasněného problému Kristova prvenství v předurčení. Teze Vladimíra Boublíka je konfrontována s námitkami, jež proti ní byly vzneseny, a její obsah je porovnán s obsahem dnešního příspěvku k danému tématu. Protein Control Over Carotenoid Spectroscopy and Functions

2013 - Czech The photophysics of pigments is influenced, to an extent depending on its structure, by the properties of the environment. Proteins represent a very specific environment at least in two aspects: i) they are native to most of the pigments in living systems; ii) they facilitate modifications of pigment configuration, leading to changes not only in its spectroscopic properties, but also in its functional abilities. In studies presented in this thesis, femtosecond pump-probe spectroscopy was used to study predominantly the photosynthetic antenna complexes of bacteria and algae. Based on spectroscopic evidence, the structural modifications of pigments imposed by the protein were deduced or hypothesized, together with their functional relevance. Strategy of Regional Development and Rural Development supported by the EU and their implementation in the Czech Republic

2014 - Czech The Doctoral dissertation, the Strategy of Regional Development and Rural Development supported by the EU and their implementation in the Czech Republic, deals with the evaluation of a link between prosperity of a municipality based on the population or migration growth or population decline and drawing subsidies from the European Union. The aim of this work is to determine whether the thriving municipalities are more successful in drawing of subsidies than the unprofitable municipalities. A sub-goal of this Doctoral dissertation is to analyse the impact of human potential of municipalities on drawing of EU funds. The author, in the practical part of the Doctoral dissertation first focused on testing the prosperity of 100 randomly selected rural municipalities and their drawing of EU subsidies during the period 2007 - 2011. For these 100 municipalities was calculated the modified HDI which represents the human potential of municipalities and was compared with municipalities with respect to their prosperity. Finally, there was determined the dependence of drawing money from EU funds on the human potential of municipalities using the correlation and regression analysis. Předkládaná disertační práce, Strategie regionálního rozvoje a rozvoje venkova podporované EU a jejich uplatnění v ČR, se zabývá zhodnocením souvislosti mezi prosperitou obce vymezenou na základě přirozeného nebo migračního přírůstku nebo úbytku obyvatel, a čerpáním dotací z Evropské unie. Cílem disertační práce je zjistit, zda jsou v čerpání dotací prosperující obce úspěšnější než obce neprosperující. Dílčím cílem je analýza vlivu lidského potenciálu obcí na čerpání fondů EU. Autorka se v aplikační části zaměřila nejdříve na testování prosperity 100 náhodně vybraných venkovských obcí s jejich čerpáním dotací z Evropské unie v období 2007 ? 2011. Pro těchto 100 obcí byl vypočten modifikovaný HDI, který představuje lidský potenciál obcí a byl porovnáván s obcemi z hlediska jejich prosperity. Nakonec byla zjišťována závislost čerpání dotačních prostředků z EU na lidském potenciálu obcí pomocí korelační a regresní analýzy.
