Circadian rhythms of photosynthesis in marine diatom Thalassiosira weissflogii under nitrogen replete and deplete conditions

T. weissflogii was grown under dynamic irradiance conditions (sinusoidal light-dark regime) and then switched to constant, low irradiance. Cellular characteristics (pigment and nutrient content, cell size) indicated synchronization of algal population, furthermore pronounced variations in the aktivity of variable fluorescence, thermoluminescence and in the gene expression (rbcL, psbA, fcp) were observed T. weissflogii byla pěstována v podmínkách dynamické ozářenosti (sinusoidní režim) a poté přenesa na konstantní světlo o nízké intenzitě. Anthropogenic increases in atmospheric pCO2 over the last century are accompanied by higher concentrations of CO2 (aq) in the surface oceans. These changes could influence aquatic primary production including cyanobacterial nitrogen-fixers via the coupling between photosynthesis and nitrogen-fixation. We hypothesized that elevated pCO2 levels may enhance nitrogen fixation and photosynthesis in the bloom-forming, diazotrophic, cyanobacterium Trichodesmium spp. that contributes greatly to primary production in the oligotrophic subtropical and tropical oceans. We acclimated cultures of Trichodesmium IMS101 to 3 pCO2s (pre-industrial -250 ppmv, current - 400, future -900 ppmv) and examined changes in morphology, photosynthesis, nitrogen fixation, and growth. High pCO2 enhanced growth, filament length and nitrogen fixation of Trichodesmium in comparison with both ambient and low pCO2 acclimated cultures Antropogenní zvýšení atmosférického CO2 může ovlivnit i mořské diazotrofní sinice, které fixují atmosferický uhlík. Ukázali jsme, že zvýšení CO2 má pozitivní vliv na rychlost růstu a fixace dusíku u sinice Trichodesmium. Characterization of a restriction modification system from the commensal Escherichia coli strain A0 34/86 (O83:K24:H31)

We have characterised a putative restriction-modification system EcoA0ORF42P in commensal Escherichia coli strain A0 34/86 (O83: K24: H31). This system is a functional member of the Type IB family, whose specificity differs from those of known Type IB enzymes, as proved by immunological cross-reactivity and complementation assay. Using a plasmid transformation method and RM search computer program, we have identified the DNA recognition sequence of EcoA0ORF42P as GGA(8N)ATGC Charakterizovali jsme předpokládaný restrikčně-modifikační systém EcoA0ORF42P komensálního kmene Escherichia coli A0 34/86 (O83:K24:H31). Imunologická analýza a komplementační testy prokázaly, že se jedná o funkční systém patřící do skupiny Typu IB, jehož specifita se liší od dosud známých enzymů této skupiny. Fluorescence kinetic microscopy: 2D imaging of variable chlorophyll fluorescence from single algal cells with spectral resolution

The fluorescence kinetic microscopy (FKM) provides new dimension in application of chlorophyll fluorescence techniques. It combines classical fluorescence microscopy with 2D kinetic detection systems. FKM allows to examine photosynthesis of individual algal cells or on the cellular level of macroscopic structures. FKM is well suited to study heterogeneity in photosynthetic response among individual cells in algal cultures or in natural mixed populations. Special microscopic chambers allow to maintain live cells under optimal conditions for several hours or days and to follow evolution of photosynthesis in individual cells during the course of their growth cycle.
