This thesis investigates the systematics and biology of Neotropical Cassidinae and summarizes available knowledge of all tribes occurring in the Neotropics. Circadian clock genes in insects

This thesis focuses on molecular characterization of circadian clock genes in insects. It explores genetic diversity of circadian clock genes by molecular characterization of several insect species including two dipteran flies (Musca domestica and Drosophila melanogaster), two cockroach species representing ancestral insects, and the linden bug, Pyrrhocoris apterus. Furthermore it considers various roles of circadian clock genes in insect physiology. Application of molecular-biology methods in Pyrrhocoris apterus, non-model insect species, enable us to investigate involvement of circadian clock genes in photoperiod induced physiological responses. Application of molecular-biology methods in Periplaneta americana and Blattella germanica was used to explore involvement of circadian clock genes in magnetoreception. Nutritional requirements of ticks.

Ticks acquire nutrients only by a parasitic nature of feeding on animals, including humans. During this process, a wide array of pathogens is transmitted. Ticks of the Ixodidae family receive exactly one blood meal in each active developmental. Knowing the trophic dependence of tick metabolism on the host blood meal components may enable discovering processes essential for the tick physiology and development. Exploiting a membrane system of tick feeding and whole blood fractionation, we have revealed that adult ticks need to acquire host haemoglobin-derived haem so that they can produce viable larvae, and reproduce. Haem is not further catabolised in ticks, and iron is thus acquired via independent route with the host serum transferrin as a source molecule. Using RNA-seq, we compared transcriptome compositions between guts of blood- and serum-fed ticks. We identified fifteen gut transcripts that change their levels with respect to the presence/absence of dietary red blood cells. Glutathione S-transferase, one of the identified encoded molecules, shows a clear haeminresponsive expression at both transcript and protein levels. Its apparent haem-binding properties suggest that this protein is directly involved in haem homeostasis maintenance within the tick gut. The ultimate goal of such research is to identify and verify targets that, when blocked, would render the acquisition and/or distribution system of haem in ticks nonfunctional. Genomic and Cellular Integration in the Tripartite Nested Mealybug Symbiosis

The PhD thesis is composed of three publications on genomic, metabolic, and cellular integration between the host and its symbionts in the tripartite nested mealybug system. The articles revealed a path to an intimate endosymbiosis that can be compared to what we think happened before (and to some extent after) bacterial ancestors of key eukaryotic organelles, mitochondria and plastids, became highly integrated into their host cells. I argue that these much younger symbioses may tell us something about how the mitochondria and plastids came to be, at the very least by revealing what types of evolutionary events are possible as stable intracellular relationships proceed along the path of integration. V této práci jsou studovány spektrální charakteristiky fotosyntetického aparátu smrku ztepilého pomocí spektroskopie cirkulárního dichroismu a nízkoteplotní fluorescenční spektroskopie a porovnány vůči vybraným modelovým rostlinám (huseníček rolní, ječmen jarní). Hlavním cílem této práce bylo získat informace o pigment-proteinových komplexech smrku, jejich makro-organizaci a stabilitě v thylakoidní membráně. In this thesis, spectral characteristics of Norway spruce photosynthetic apparatus are studied using circular dichroism and low temperature fluorescence spectroscopy and compare to those of model plants Arabidopsis thaliana and spring barley. Main goal of this thesis was to obtain new information about the pigment-protein complexes of spruce, their supramolecular organization and stability in the thylakoid membranes. Diplomová práce pojednává o principech a zásadách uplatněných v řízení před Arbitrážním soudem. Tato práce se zabývá analýzou a kontrolou PLC programu. Její soucástí je nástroj pro automatické generování binárních rozhodovacích diagramu a stavových automatu pro promenné, které jsou použity v PLC programu. Hlavním cílem práce je prevést program napsaný v Ladder Diagramu prímo do BDD a stavového automatu. Kontrola programu je provedena prevedením do UPPAAL a následnou verifikací.This thesis is about analyzing and verification of PLC program. There is also an instrument for automatical generating of binar decision diagram and for state machines for variables which are used in PLC program. Main result of thesis is translation of program in Ladder Diagram into BDD and state machine. Verification of the program is made by translation of state machine into UPPAAL. Táto diplomová práca sa zaoberá konaním o svojprávnosti do doby, než súd dospeje k meritórnemu rozhodnutiu. V práci je okrem metódy analytickej, použitá metóda komparačná s predchádzajúcou právnou úpravou, konkrétne s § 186 a § 187 občianskeho súdneho poriadku účinného do 31. 12. 2013. V úvode práce je zdôraznený význam Úmluvy o právach osôb so zdravotným postihnutím, ktorá je v Českej republike platná už od roku 2009 a v požiadavkách ktorej tkvie základ súčasnej právnej úpravy svojprávnosti. Ďalej je v práci podrobne rozobraná napríklad možnosť súdu svoju príslušnosť preniesť, použitie pojmu zdravotný ústav, výsluch posudzovaného, výsluch znalca alebo obligatórne zastupovanie posudzovanej osoby procesným opatrovníkom. V dôsledku prijatia novej právnej úpravy bolo do zákona o zvláštnych konaniach súdnych zaradené ustanovenie, ktoré hovorí o povinnosti súdu počas celého priebehu konania zvažovať využitie miernejších a menej obmedzujúcich opatrení. Tejto problematike sa venuje posledná kapitola so zameraním na voľbu opatrovníka bez obmedzenia svojprávnosti. This diploma thesis deals with proceedings on legal capacity until the court arrives at a decision on the merits. Analytical method is used in this thesis as well as comparative method with previous legal regulations, more specifically, with § 186 and § 187 of Civil Procedure Code effective till 31st December 2013. Importance of Convention on the Rights of Persons with Disabilities is emphasized at the beginning of this thesis, which has been effective since 2009 in the Czech Republic and whose requirements are the base of current legal capacity legislation. The following conceptions are also described within the thesis: the option of court to carry its venue jurisdiction, use of term health institution, the hearing of person whose legal capacity is decided, the hearing of an expert witness or compulsory procuration of a person whose legal capacity is decided by the process guardian. Following the adoption of the new legal regulations, a provision of a law has been included in the Act on Special Judicial Proceedings, which regulates commitment of the court to consider milder and less restrictive measures during the proceedings. This issue is analyzed in the last chapter focusing on the choice of guardian without restriction of legal capacity.
