Application of pressurized hot water for etching of microfluidics

The subject of this study is a new method of glass etching and its application in microfluidics. Commonly, hydrofluoric acid is used for glass etching. In this study we have tested a new method of glass etching using pressurized hot water. Test motives were prepared using photolitography and metal sputtering on the glass substrate and the etching was performed in a newly developed device capable to compress and heat water to its critical point. Optimization of N-glycopeptides analysis methods and their preliminary application to barley proteins study

N-glycosylation is the most frequently studied plant protein post-translational modification. The analysis of Nglycopeptides after protein proteolytic digestion offers information about the structure of both oligosaccharide and peptide moiety. However, this method has so far been less commonly used. Since glycopeptides hardly ionize during MS analysis in the presence of non-glycosylated peptides, they need to be separated from the complex peptide mixture. In this study, the glycopeptides enrichment, purification and analysis methods were successfully optimized on two standard N-glycoproteins. Concanavalin A (ConA) lectin tips were used for glycopeptide capturing, and obtained fractions were purified on carbon tips and analyzed using MALDI-TOF mass spectrometry. The differences in the CID fragmentation of certain types of glycopeptides were found. This technique was then applied to glycopeptide analysis of barley grain and malt proteins. Several barley glycopeptides were found, however, their identification was very difficult. More proteins separation techniques will be required before this enrichment procedure in further studies. Recent biomedical research prefers analysis of homogenous samples obtained from primary cells to cell lines. Moreover, attention is paid to cells of low number but high biological impact, such as signalling centres in embryonic development, stem cells or tumour cells. To localize cells of interest within an organ/tissue, histological sections are widely used. However, the methods of choice for further analysis of such samples are mostly limited to histochemistry, immunohistochemistry and in situ hybridization. Srovnání výsledků měření a modelování depozičního a stomatálního toku ozonu u smrku ztepilého v lokalitě Bílý Kříž v červenci a srpnu 2008. Zvýšené koncentrace ozonu snižují čistou ekosystémovou produkci, zvláště při vyšších teplotách a vysoké intenzitě globálního záření. Measured and modelled results of ozone deposition and stomatal flux to a Norway spruce forest in locality Bílý Kříž in July and August 2008 were compared. Enhanced concentration of ozone decreased net ecosystem production, especially at higher temperatures and high intensity of global radiation. The especially inhalation chamber is constructed for chronic exposure studies of small animals to nanoparticles under strictly controlled conditions in frame of project „Study of transport of inhalated nano-sized particles (Pb, Cd) and their allocation in organs“. The inhalation chamber builds from a glass and stainless steel and makes possible long-term inhalation procedures (up to 60 days, 24 hours/day) with up to four discrete groups of mice population (up to 60 individuals each) or of rats population (up to 20 individuals each) under controlled illumination, temperature, relative humidity, doses (0.5 x 106 - 5 x 106 particles/cm3) and size (10-60 nm) of nano-sized particles which are synthesized via aerosol route in a hot wall tube flow reactor using metal organic precursors. Aerosoly ve velikostní frakci PM2.5 a PM10 a pouliční prach byly paralelně vzorkovány v Ostravě a Brně během zimy a léta 2009 a 2010. Zdroje aerosolů, včetně resuspendovaného prachu a jeho příspěvku k celkové koncentraci aerosolů, byly identifikovány pomocí metod receptorového modelování. Parallel collection of aerosol particles in size fractions PM2.5 and PM10 and street dust at the same location was performed in Ostrava and Brno during winter and summer 2009 and 2010. Identification of PM sources including resuspended PM and their contribution to overall PM concentration was done by using receptor modelling methods.
