Combined effect of temperature and CdO nanoparticles treatment on Picea abies

Three-years old seedlings of Picea abies were pre-treated under two different\ntemperatures (20x35 °C) and subsequently exposed to elevated concentration of CdO\nnanoparticles (CdONPs). Two-week exposure to airborne CdONPs of ecologically\nrelevant size and concentration significantly reduced photosynthetic light use efficiency\nin plants as well as contents of photosynthetic pigments and saccharides. In contrary,\namino acid content was increased under CdONPs. These changes were further\nmodulated by previous growth temperature. Highly sensitive analysis of chlorophenols and sulfonamides in waters by electrophoretic focusion on inverse electromigration dispersion gradient with ESI-MS detection

This work presents a new methodology for high-sensitivity analyses by capillary\nelectrophoresis (CE) with electrospray-ionization mass spectrometric (ESI-MS)\ndetection, based on electrophoretic focusing on an inverse electromigration dispersion\n(EMD) gradient. The technique is based on a robust electrolyte system providing a\ngradient of required properties, suitable for routine analyses of trace amounts of weak\nacids with pKa values between approx. 6.5 and 9. Examples analyses of several\nchlorophenols and sulfonamides are presented, with LODs around 3x10-9 M for spiked\ndrinking water without any sample pretreatment. A novel nanospray liquid junction interface for versatile CE-MS

In this work we present a new polyimide-based liquid junction nanospray interface, combining the advantages of sheath flow and sheathless designs. The liquid junction interface was prepared as a hybrid microfabricated plastic liquid junction sprayer allowing to perform analysis in standard (30-100 micro m ID) separation capillaries. The interface incorporates a self-aligning port for coupling of the separation capillary as well as ports for automated flushing of both the liquid junction and the separation capillary. The interface permits performing CE both with and without electroosmotic flow. The use of large bore capillaries allows injection and preconcentration of larger sample volumes, favoring detection sensitivity. Different compositions of the spray liquid and BGE can be used when needed. Parallel single-cell analysis of active caspase-3/7 in apoptotic and non-apoptotic cells

Caspases are proteases that play key role in the process of apoptosis, the programmed\ncell death. Among them, caspase-3 and -7 are main executioner caspases that cleave\nmany vital proteins during apoptosis and after their widespread activation, the process\ncannot be reversed. To analyze caspase-3/7 activation within single cells, a miniaturized\ndevice for parallel analysis of eight samples was developed. The assay is based on the\nmodified luciferin-firefly luciferase bioluminescence (BL) system. Individual\nsuspended cells were collected and transferred into detection microvials using a\nmicromanipulator. The bioluminescence was detected using a photon counting head\nwith cooled photcathode. The LOD suitable for detection of active caspase-3/7 in both\napoptotic and non-apoptotic cells was reached. In this work we have developed a new sensitive method for quantitation of MDA in\nbiological samples by CE-LIF with LOD about 1.7 nM. First part of the work was\ndevoted to the construction of a new CE-LIF system. Then the derivatization process\nof MDA by TBA was optimized. The ability to use this method for the detection of\nMDA in a biological samples, such as EBC and plasma, was also demonstrated. For the\nfirst time it was possible to detect MDA in EBC samples by CE. Other biomarkers are\nnow being studied. Micro-electromembrane extraction (μ-EME) across multiple aqueous and organic\nsolutions was developed for selective extractions of various analytes from untreated\nbiological samples. The novel approach requires μL volumes of samples and organic\nsolvents, which form consecutive immiscible plugs in a transparent polymeric capillary\nand the extraction process is accelerated by application of d.c. voltage. Number of the\nplugs, their composition, volume and sequence in the extraction capillary can be chosen\nspecifically for each particular application and high variability and selectivity of the\nextraction process can be achieved. In this contribution, we demonstrate suitability of\na five-phase μ-EME system for simultaneous extractions of cations and anions from\nurine and for selective extractions of basic analytes based on their acid-base strength\nfrom plasma.
