Structure of an immunoactive polysaccharide isolated from Korean mulberry fruit (Morus alba L.)

A water-soluble polysaccharide (JS-MP-1) was isolated from the water extract of Korean white mulberry fruits (Morus alba L.) by ethanol precipitation and then purified by DEAE-cellulose ion exchange chromatography. Obtained final polysaccharide (1600 KDa) consisted of galactose, arabinose and rhamnose as major neutral sugars. JS-MP-1 also contains galacturonic and glucuronic acids (4:1). Sugar linkage, FTIR and correlation NMR analyses confirmed that it is a rhamnogalacturonan type I (RG I), which contains the alternating sequence of 1,4-alpha-D-GalAp and 1,2-alpha-L-Rhap units in the backbone. Neutral sugar side chains of JS-MP1 were identified as (1 -> 5)-alpha-L-arabinan and arabinogalactan type II (AG II) having the (1 -> 6)-beta-D-galactan core. The arabinan side chains are bound to the backbone at the O-4 position of some alpha-L-Rhap units, while the way of linkage between RG I and AG II chains is unclear. It was demonstrated that JS-MP-1 significantly stimulates murine macrophage RAW 264.7 cells to release chemokines (RANTES and MIP-1 alpha) and proinflammatory cytokines like INF-alpha and IL-6, and induce the iNOS and COX-2 gene expression, which are responsible for the production of NO and prostaglandin PGE2, respectively. These results suggest that JS-MP-1 can act as a potent immunomodulator and these observations may support the applicability of this polysaccharide or the water extracts of mulberry fruit can be used as an immunotherapeutic adjuvant or health beneficial food material. Halogenated organics are emitted into the atmosphere from a variety of sources of both natural and anthropogenic origin (Finlayson-Pitts et al., 2000). Their uptake at the surface of aerosols can affect their reactivity, for example in processes which take part in ozone destruction due to production of reactive chlorine, bromine and iodine radicals. This study presents results of molecular dynamic (MD) simulations carried out to investigate the interaction of short-chain haloalkanes with aqueous surfaces. We report on a free energy minimum that the haloalkanes exhibit at the air/water interface. In all samples of fireworks, K+, Na+, Ca2+ and Mg2+ ions were proved and quantified by the method of capillary zone electrophoresis using water solution of 10 mM 4-aminopyridine, titrated with hydroxyisobutyric acid to the pH 4.3, as background electrolyte and indirect UV-absorption detection at 200 nm. Pb2+ or Ba2+ ions were proved in few samples. The other selected inorganic cations were not proved in any sample of selected fireworks. NH4+ ions were not detected in any pre-blast samples, but they were established in some post-blast (glazed tiles) samples. Open-tubular capillary electrochromatography (OT-CEC) using bare gold nanoparticles-based stationary phase has been applied to separation of tryptic peptide fragments of native and glycated proteins, bovine serum albumin and human transferrin. OT-CEC with bare gold nanoparticles stationary phase was found to be a suitable technique for separation of complex peptide mixtures originating from enzymatic (tryptic) digestion of native and glycated bovine serum albumin and human transferrin, and for glycation studies of these proteins. Two mixtures of human and salmon gonadotropin releasing hormone (GnRH) derived peptides were successfully separated by capillary zone electrophoresis (CZE) in six classical buffer-based BGEs and by carrier ampholytes-based capillary electrophoresis (CABCE) in the same number carrier ampholytes-based isoelectric BGEs of the same or close pH values. In addition, from the pH dependence of peptide electrophoretic mobilities, the isolectric points of separated peptides were determined.
