2013 - English
To improve the localizing yield of interictal discharges, identification of specific features of the discharges generated only within epileptogenic region is required. We have developed self-clustering algorithm which discriminates distinct populations of interictal epileptiform discharges according to the morphology of their waveforms. Results demonstrate the ability of the algorithm to separate interictal epileptiform discharges according to their morphological features Keywords: epilepsy; interictal epiletiform discharges; morphological clustering Available at various institutes of the ASCR

2012 - English
Posttranslational modifications of proteins are important reactions, which significantly affect the function of proteins in the organism. In principle, they can be divided into enzymatic and non-enzymatic modifications. Non-enzymatic reactions include glycation (earlier called nonezymatic glycosylation), which plays an important role in the development of chronic complications of diabetes mellitus, uremia, in the process of aging and degeneration of the brain. This work deals with the study of glycated albumins (human serum albumin and bovine serum albumin). Methodologically we used nano-liquid chromatography coupled to Q-TOF mass spectrometer. In vitro modified proteins were cleavaged by trypsin and arising peptides were separated on C18 nano column with trap-column. Peptides and their modifications were analysed by high-resolution Q-TOF mass spectrometer MaXis with precision determination of mass below 2 ppm. We found some modifications of proteins. Besides well known carboxymethyllysine new ones were determined - create mass shift 78, 132 and 218. Origin of these modifications is discussed and possible structure is presented. All found modifications were allocated to the structure of proteins and reactivity to various oxo-compounds was also examined Keywords: non-enzymatic glycation; serum albumin Available at various institutes of the ASCR

Application of morphology filters to compensation of lateral illumination inhomogeneities in confocal microscopy images
Michálek, Jan; Čapek, Martin; Mao, X. W.; Kubínová, Lucie
2010 - English
Due to multiple distortion effects, confocal laser scanning microscopy (CLSM) images of even homogeneous specimen regions suffer from irregular brightness variations. In this paper, a fast correction method based on estimating a spatially variable illumination gain, and multiplying acquired CLSM images by the inverse of the estimated gain, is presented. The method does not require any special calibration (reference) images since the gain estimate is extracted from the CLSM image being corrected itself. The proposed approach exploits two types of morphological filters: the median filter and a morphological operator called the upper Lipschitz cover Keywords: CLSM; illumination inhomogeneities Available at various institutes of the ASCR

Elastic registration of biomedical images on CUDA-Supported graphics procesor units
Michálek, Jan; Čapek, Martin; Janáček, Jiří; Kubínová, Lucie
2010 - English
Elastic registration is a task of finding the matching of two images, using geometric and elastic transformations, so that objects in images have the same size, position and orientation. We apply elastic registration in the framework of volume reconstruction, where an object acquired from parallel physical sections is composed and mutual positions of the sections including deformations caused by their cutting have to be found. The method lies in optimizing a functional consisting of two parts: first, discrete total variation as a measure of roughness and, second, L1 norm as a measure of dissimilarity of images. As a parallelizable optimization strategy we apply a potential-based equivalent transformation of a (max,+)-labelling problem. CUDA-based implementation of the described elastic registration algorithm is reasonably fast, requires seconds to minutes of calculations, provides good results and, thus, can be used for practical tasks dealing with alignment of biomedical images Keywords: volume reconstruction; elastic alignment; CUDA Available at various institutes of the ASCR

Bláha, L.; Vyvadilová, M.; Janáček, Jiří
2009 - Czech
The basic aim of presented work was analysis of root development of different oilseed rape cultivars at two localities. Obtained results show large influence of localities on the root development. The genotypes with good germination under unfavourable conditions (lack of water, drought, high temperature) develop stronger root system i.e. volume and length of the roots after sowing and during following vegetation period at studying localities Keywords: winter oilseed rape; root system; stress tolerance Available at various institutes of the ASCR

Janáček, Jiří
2009 - Czech
Práce je věnována řešení úloh analýzy obrazu minimalizací funkce složené z totální variace obrazu a Lp ztrátové funkce. Minimalizaci lze provést iterační metodou největšího spádu s výpočtem kroku založeným na nalezení minimálního řezu v grafu obrazových prvkù. Jako příklad je uvedena regularizace zašuměného obrazu a registrace mikroskopických obrazů fyzických řezů A solution of various problems in image analysis using concurrent minimization of total variation and Lp loss function is presented. Multi-photon excitation microscopy
Chernyavskiy, Oleksandr; Kubínová, Lucie
2008 - English
Multiphoton excitation microscopy (MPM), being some time ago rather exotic than an easy-to-handle tool, becomes now more widely used due to advances in laser technique. This promising, yet expensive technique provides such advantages, as deeper penetration depth, lower photodamage, especially important in live cells imaging for extended periods of time Multifotonová excitační mikroskopie se z exotické techniky postupně stává stále více používaným nástrojem díky pokroku v laserové technice. Tato slibná i když náročná technika poskytuje výhody, jako je průnik do větší hloubky, nižší fotopoškození, jež jsou zvláště užitečné při dlouhodobém zobrazování buněk Keywords: two-photon excitation; multiphoton excitation; second-harmonic generation Available at various institutes of the ASCR

The goal of this study was to fellow up the effect of n-3 and n-6 polyunsaturated fatty acids (PUFA) and the adaptation to chronic intermittent hypoxic (CIH) on fatty acid (FA) composition of serum and heart lipids in rats. The results of this study showed that PUFA diets and CIH distinctly modified FA profile in serum and heart lipids; both interventions considered to be cardioprotective Cílem práce bylo sledovat změny srdečních a sérových lipidů potkana po dlouhodobé dietě obohacené o n-3 a n-6 polynenasycené mastné kyseliny (PUFA) a adaptaci na chronickou intermitentní hypoxii (CIH). Studie ukázala, že jak dieta obohacená o PUFA tak CIH zásadně modifikují složení mastných kyselin v sledovaných lipidech a mohou souviset s potenciálními kardioprotektivními účinky obou intervencí Keywords: heart; chronic hypoxia; lipid diet Available at various institutes of the ASCR
