Anaplasma phagocytophilum je významný patogen přenášený klíšťaty, který kolonizuje granulocytární buňky obratlovců. Vyskytuje se v Severní Americe a v několika evropských zemích. Způsobuje závažná onemocnění domácích zvířat a potenciálně smrtelné onemocnění lidí zvané lidská granulocytární anaplazmóza (HGA). Vzhledem k rostoucí prevalenci A. phagocytophilum je nutné zavedení zvířecích modelů. Tato studie se zabývá přenosem kmene bakterie A. phagocytophilum NY-18 dvěma druhy klíšťat (Ixodes ricinus a Ixodes scapularis) a zavádí laboratorní model přenosu A. phagocytophilum klíštětem I. ricinus u myší. Práce dále porovnává oba přenosy i dynamiku infekce. Anaplasma phagocytophilum is a significant tick-borne pathogen that colonizes granulocytic cells in vertebrate hosts. It is found in North America and in several European countries. It causes severe diseases in domestic animals and potentially fatal disease in humans called human granulocytic anaplasmosis (HGA). The implementation of animal models is necessary due to the increasing prevalence of A. phagocytophilum. This study addresses the transmission of the human isolate of the bacterium A. phagocytophilum NY-18 by two tick species (Ixodes ricinus and Ixodes scapularis) and implements a laboratory model for the transmission of A. phagocytophilum by the tick I. ricinus in mice. The thesis also compares both transmissions and infection dynamics. V této studii jsme porovnávali koncentraci dvou genotypů (I a III) parazita Encephalitozoon cuniculi v experimentálně vyvolaných zánětlivých ložiscích v kontrolovaném experimentálním prostředí. Výsledky této studie mohou přispět k pochopení šíření parazita v těle hostitele a pomoci při vývoji cílené léčby. In this study we compared the concentration levels of two genotypes (I and III) of the parasite Encephalitozoon cuniculi in experimentally induced inflammation foci in a controlled experimental setting. The results of this study may provide insights into the understanding of the parasite's spreading within host body and help in the development of targeted treatments. Influence of different buckwheat flours on selected qualitative parameters of cookies

common buckwheat (fagopyrum esculentum Monech) consumption has many of the health benefits due to its high levels of phenolic compounds in groats and high antioxidant activity. The aim of the work was to test the possibility of cookies production with graduated proportion of different buckwheat flours from sprouts and evaluate their selected qualitative properties. Blends of wheat flours and buckwheat sprout flours containing 0%,5%, 10%,and 20%buckwheat sprouts , on a replacement basis , were prepared .flour characteristics as water absorption capacity , bulk density forming capacity , swelling capacity were determined .Physical ( diameter ,thickness, spread ratio, spread factor, weight hardness and colour)and sensory characteristics of cookies ( colour and appearance , flavour, taste texture and overall acceptability ) were also determined .The addition of buckwheat sprout mainly affected the colour properties of the cookies .. of the properties of flour enriched with buckwheat sprouts, water absorption capacity was affected only by the addition of 20% buckwheat sprout and swelling capacity by the addition of 15 and 20% sprouts .of the sensory properties of the cookies , the taste and flavour of the cookies was negatively affected only by 20% share of buckwheat sprout in flour ,therefore , we can recommend the addition of buckwheat sprouts to wheat flour up to 15% for the production of cookies . The significant of a well- balanced diet in the prevention of numerous disease is a well known , consequently , individual nutrition should be aimed at the disease prevention , improving the quality of life and attenuating the process of body ageing. common buckwheat has been a crop of the secondary importance in many countries . it is a sources source of nutrient in general and has many health benefits mainly due to high phenolic compounds in groat and high antioxidant activity. since buckwheat is a gluten free ,thus ,replacing wheat flour in product will dilute the gluten protein and thereby help in preventing celiac disorder . Buckwheat act as good replacement for wheat flour in unleaving product eg cookies.it belong to the family polygonacea . this plant is generally referred to as buckwheat rhrubarb or sorrel family. The common buckwheat plant is a branching habit . common buckwheat was domesticated possibly around 6000BCE when it was first cultivated in southeast Asia .the use of sprout as an ingredient in food product is an interesting method of improving the nutritional value of food , as well as a safer way to ingest sprout , since food processing can reduce microbial contamination and consequently decrease the occurrences of food-borne disease . the germination process is accepted by customers , as well as the product made from the germinated grains or from their flour. Tato práce zkoumá vědecký základ agroekologických praktik, metod a struktur doporu-čené permakulturou, jak uvedené na dotazník České permakulturní asociace s účelem kvalifikovat permakulturní projekty jako takové čí jako projekty s permakulturními prv-ky. Pro praktiky, metody a struktury uvedené byla provedená literární rešerše na Web of Science. Přes tuto rešerši bylo možné identifikovat různé publikace potvrzující vhod-nost těchto doporučených praktik, i když pro některé z nich, zejména aplikace průmys-lových hnojiv a bezorebné systémy, vědecké výsledky naznačí pozitivní vlivy, dokud správně aplikované. Dále práce s tímto dotazníkem sugeruje, že permakultura není prostě agroekologická hnutí, ale její dnešní ohnisko je širší, hledající udržitelná řešení v mnoho aspektech života, včetně ubytování, získávání energie a vody a také socio-ekonomických aktivit lokálních a regionálních komunit. This thesis attempts to verify whether agroecological practices recommended by per-maculture are scientifically founded. For this purpose, a questionnaire used by the Czech permaculture association (Permakultura CS) to qualify projects as permaculture showcase projects or projects with permacultural elements, was used, and for the listed practices, methods and structures a literature search on Web of Science was per-formed. By the literature search we could identify several publications confirming the suitability of the recommended practices, although for some of them, in particular the avoidance of tillage as well as industrial fertilizers, scientific evidence suggests that, as long as correctly applied, these may even have a positive impact. Furthermore, the re-view of the questionnaire confirms that permaculture is not per se an agroecological movement, but its focus is nowadays broader, searching for sustainability solutions in most aspects of life, including housing, energy and water sourcing, as well as in socio-economic aspects of local and regional communities. This study aims to explore the principle of the interdependence and mutual influence of the Dark web and cryptocurrency. Specifically, it explores the terms "the Dark web" and "cryptocurrency", as well as distinguishes cryptocurrency from normal currency. Furthermore, it establishes a connection between the Dark web and cryptocurrency. In order to reach the aim of this study, multiple sources of literature had been analyzed, including books and online articles. The analysis of literature shows that there is a strong correlation between the existence of the Dark web and demand on cryptocurrency. Thus, the results of this paper conclude that the Dark web and cryptocurrency exist in symbiosis - cryptocurrency allows the Dark web to exist for its anonymity and impunity, while the Dark web guarantees demand on cryptocurrency. Hlavním cílem této práce bylo zhodnotit proveditelnost výroby udržitelných a biologicky odbouratelných filmů pro balení potravin vyrobených z rostlinných surovin metodou odlévání. Specifické cíle byly následující: a) Optimalizace kroků odlévací metody pro přípravu filmů na základě použití různých druhů mouk, stanovení jejich fyzikálních vlastností (barva, soudržnost, pružnost). b) Příprava biofilmů z vybraných druhů mouk, vyhodnocení jejich fyzikálních a mechanických vlastností (vizuální a hmatové aspekty, barva, tloušťka, vlhkost). c) Zkoumání biologické rozložitelnosti získaných materiálů na základě enzymatické degradace ve vodním prostředí a ve vodě samotné. The main aim of the thesis was to evaluate the feasibility of the production of sustainable and biodegradable films for food packaging made from raw plant materials using casting method. Specific objectives were: a) Optimization of the steps of the casting method for films preparation based on the use of different types of flours, determination of their physical properties (colour, cohesiveness, elasticity). b) Preparation of biobased plastic material from selected types of flours, determi-nation of their physical and mechanical properties (visual and tactile aspects, colour, thickness, moisture content). c) Investigation of the biodegradability of prepared materials based on enzymatic degradation in water suspension and water itself. Účinnost mykoparazitických a entomopatogenních hub proti houbovým původcům onemocnění rostlin

Entomopatogenní a mykoparazitické houby mají potenciál být využívány na místní i mezinárodní úrovni jako účinná a ekologicky šetrná strategie boje proti škůdcům a chorobám rostlin způsobeným houbovými patogeny. V bakalářské práci byla tes-tována účinnost užitečných hub v laboratorních podmínkách. Metodika zahrnovala izolaci a popis užitečných druhů hub a následné testování užitečných hub s poten-cionálními mykoparazitickými a antagonistickými účinky. Účinnost hub byla tes-tována na patogenech, které byly převážně odizolovány z osiva nebo nemocných ros-tlin luskovin. V rámci pokusů bylo zjištěno, že všechny kmeny Trichoderma izolo-vané z půd ČR prokázaly mykoparazitický efekt na vybrané patogeny. Nicméně, účinnost kmenů závisela na druhu houbového patogenu. Hodnocena byla i produkce spor kmenů Trichoderma jak v kontrolní variantě, tak i ve vzájemné kombinaci s patogeny. Výsledky opět prokázaly rozdíly v produkci spor u jednotlivých kmenů Trichoderma. Zároveň byl hodnocen mykoparazitický účinek entomopatogenních hub Isaria fumosorosea a Akanthomyces attenuatus proti patogenu Botrytis cinerea. Výsledky ukázaly, že ačkoli I. fumosorosea a A. attenuatus vykazovaly významný mykoparazitický účinek proti Botrytis cinerea, nejvyšší účinnost proti patogenu prokázala mykoparazitická houba T.virens. Zjištění naznačila, že jak mykoparazitické, tak entomopatogenní houby jsou udržitelnou alternativou umělých pesticidů v boji proti houbovým původcům chorob rostlin. Entomopathogenic and mycoparasitic fungi have the potential to be used both locally and internationally as an effective and environmentally friendly strategy for the man-agement of pests and plant diseases caused by fungal pathogens. In this bachelor thesis, the efficacy of beneficial fungi was tested under laboratory conditions. The methodology involved isolation and description of useful fungal species and subse-quent testing of useful fungi with potential mycoparasitic and antagonistic effects. The efficacy of the fungi was tested on pathogens that were mainly isolated from seeds or diseased legume plants. The experiments revealed that all Trichoderma strains isolated from soil of the Czech Republic showed mycoparasitic effect on se-lected pathogens. However, the efficacy of the strains depended on the type of fun-gal pathogen. Spore production of Trichoderma strains was also evaluated both in the control variant as well as the mutual interactions with pathogens. The results again showed differences in spore production among the Trichoderma strains. The myco-parasitic effect of the entomopathogenic fungi Isaria fumosorosea and Akanthomyces attenuatus against the pathogen Botrytis cinerea was also evaluated. The results re-vealed that although I. fumosorosea and A. attenuatus showed a significant mycopar-asitic effect against Botrytis cinerea, the mycoparasitic fungus T. virens showed the highest efficacy against the pathogen. The findings indicated that both mycoparasitic and entomopathogenic fungi are sustainable alternatives to artificial pesticides in the fight against fungal agents of plant diseases. Characterisation of novel mitochondrial proteins in \kur{Trypanosoma brucei}

This study analyses and characterises novel mitochondrial proteins in the parasitic protist Trypanosoma brucei. Applying phylogenetic analysis was described the evolutionary origin of ZapE protein in eukaryotes, using a newly developed proximity-dependent biotinylation approach (BioID2) we identified ZapE interaction partners like Oxa1. We also discovered a relationship when distribution of mitochondrial ZapE is restricted only to organisms with Oxa1, respiratory complexes, and a mitochondrial genome. TbPams were detected by phylogenetic analyses as orthologs of corresponding proteins in Opistokonts. We analyse the function of TbPam18 and TbPam16 in the replication of the mitochondrial DNA and determine, how the TMDs of TbPam18 and TbPam16 are essential for their functions. Finally, we evaluated a set of putative mitochondrial proteins of the heterolobosean N. gruberi defined by Localisation of Organelle Proteins by Isotope Tagging (LOPIT) and analyse the origin of mtFfh and mtFtsY.
