The role of Gaze in Victorian Ghost Stories

Cílem práce je zaměřit se na dvě známá díla z viktoriánské doby od Elizabeth Gaskell a Emily Brontë a poukázat na důležitou roli zraku jako smyslu v různých možných formách, a to v duchařském příběhu "The Old Nurse's Story" a románu Wuthering Heights. Práce se zaměřuje na viktoriánskou dobu a její pohled na duchy, a zárověň známé teorie a pohledy na zrak či pohled. Diplomová práce poté analyzuje daná díla a poukazuje na různé role zraku/pohledu, které se v nich objevují. This thesis aims to focus on two famous works from the Victorian era by Elizabeth Gaskell and Emily Brontë and to highlight the important role of sight as sense in various possible forms, namely in the ghost story "The Old Nurse's Story" and the novel Wuthering Heights. The thesis focuses on the Victorian era and its view on ghosts, and at the same time, well-known theories and perspectives on sight or gaze. The thesis then analyses the works in question and highlights the different roles of sight/gaze that appear in them.

Diplomová práce se zabývá analýzou narativu u vybraných próz českých autorů, které zpracovávají téma vzniku či počátku Státu Izrael. Jde o díla Vítr z hor (1991) a Vepři ve při (1993) Oty B. Krause, V zemi zaslíbené? (1986) Josefa Šebesty, Čelem proti čelu (1947) Františka Gottlieba a Miláček (1969) Arnošta Lustiga. Vznik či vydávání jednotlivých textů od 40. do 90. let 20. století umožňují nahlédnout na to, jak se v daných dekádách proměňovaly vypravěčské strategie v souvislosti s proměnou politicko-společenské situace našeho státu a politických vazeb ke státu Izrael. Knihy se tak zdají být reprezentacemi dobových diskurzů, včetně akcentu na jejich akční potenciál, který měl dobové čtenáře v prezentovaném myšlení utvrzovat či je naopak vést k zamýšlení se nad popisovaným děním. This diploma thesis is focused on the analysis of the narrative of books Vítr z hor (1991) and Vepři ve při (1993) by Ota B. Kraus, V zemi zaslíbené? (1986) by Josef Šebesta, Čelem proti čelu (1974) by František Gottlieb and Miláček (1969) by Arnošt Lustig. These books are about the creation or beginning of the State of Israel. The creation or publication of individual text from the 1940s to the 1990s provides insight into the change of narrative strategies in those decades in connection with the change in the political and social situation of our state and political ties to the State of Israel. The books seem to be representations of contemporary discourses, including an accent on their action potential, which was intended to confirm contemporary readers in the presented thinking, or, on the contrary, lead them to reflect on the events.

Tato diplomová práce se zabývá přeshraniční spoluprací vybraných euroregionů České republiky, konkrétně se zaměřuje na euroregiony Silva Nortica, Šumava, Dunaj-Vltava, Labe, Nisa, Krušnohoří a Egrensis. Nejprve práce vysvětluje klíčové pojmy jako je přeshraniční region a spolupráce, poté se zaměřuje na charakteristiku jednotlivých euroregionů, jejich cílů a projektů. V další části zkoumá historický vývoj a výzvy spolupráce v období před rokem 1989, mezi lety 1989-2020 a 2020-2023. Na závěr zhodnotí veškeré aspekty a budoucí perspektivu. The aim of the thesis is to describe the cross-border cooperation of selected euroregions in the Czech Republic, specifically the euroregions Silva Nortica, Šumava, Dunaj-Vltava, Labe, Nisa, Krušnohoří, and Egrensis. The work first explains key concepts such as cross-border region and cooperation, then focuses on the characteristics of each Euroregion, their goals, and projects. This is followed by the research of the historical development and challenges of cooperation in the periods before 1989, from 1989 to 2020, and from 2020 to 2023. Finally, it evaluates all aspects and future perspectives.

Tato diplomová práce se zabývá současným přístupem učitelů a vedení základních škol k žákům s neurovývojovými poruchami, zejména ADHD, Aspergerovým syndromem a autismem. Cílem této práce je prozkoumat, jakým způsobem mohou školy a rodiče spolupracovat na podpoře hudebního vzdělávání těchto žáků, jaké jsou výzvy a příležitosti této spolupráce a jaký vliv má na celkový vývoj dětí s neurovývojovými poruchami. V teoretické části se práce zabývá problematikou neurovývojových poruch, samotným pohledem na hudební výchovu v RVP ZV, obsahem učiva, alternativními metodami při výuce, podpůrnými opatřeními a domácím vzděláváním. Praktická část diplomové práce je založena na polostrukturovaných rozhovorech s rodiči žáků s neurovývojovými poruchami, na jejich zkušenostech ve smyslu spolupráce se školou. V závěru práce jsou zpracovány dvě kazuistiky. This thesis searches the current approach of primary school teachers and administrators to pupils with neurodevelopmental disorders, such as ADHD, Asperger's syndrome and autism. The aim of this thesis is to explore how schools and parents can work together to support the music education of these pupils, the challenges and opportunities of this collaboration and the impact it has on the overall development of children with neurodevelopmental disorders. The theoretical part of the thesis deals with the issue of neurodevelopmental disorders, the actual view of music education in the educational framework plan of elementary education, the content of the curriculum, alternative methods in teaching, support measures and home education. The practical part of the thesis is based on semi-structured interviews with parents of pupils with neurodevelopmental disorders, their experiences in terms of cooperation with the school. The thesis concludes with two case studies.

Že byl národní socialismus jedinečnou směsí nejstaršího a nejmodernějšího, je již všeobecně známo. Tuto syntézu lze nalézt i v oblasti propagandy. Nejen v osobě a práci ministra pro "lidovou osvětu a propagandu" (RMVP), kde již název vnáší do pojmu rozpor, ale také v textech, které měly zasáhnout širší veřejnost a zdánlivě "nepolitické" obory činnosti, jako jsou domácí práce, film, medicína nebo zbrojní technologie. Tato práce ukazuje a analyzuje tuto "hlubokou dimenzi" nacionálně socialistické propagandy s jejími sublimními, ale také překvapivými ideologickými podtexty. Zkoumaný materiál sahá od stranického programu až po pojmenování ručních zbraní. V úvodní teoretické části je z výzkumného hlediska představena analýza tohoto nacistického jazyka vycházející z tezí Klemperera až po nejnovější literaturu o nacistech. It has often been noted that National Socialism was a strange mixture of the oldest and the most modern. This synthesis can also be found in the area of propaganda. Not only in the person and work of the Minister for "Popular Enlightenment and Propaganda", where the title already brings the contradiction into the term, but also in texts that were intended to have an impact on the broader public and seemingly "non-political" fields of activity such as household work and film, medicine or weapons technology. This work shows and analyses this "deep dimension" of National Socialist propaganda with its sublime but also surprising ideological subtexts. The material examined ranges from the party program to the title of a rifle. In an introductory theoretical part, the analysis of this language from Klemperer's early approach to the latest literature on the Nazis is presented from a research perspective.

Diplomová práce analyzuje vybrané české protireformační texty vydané na konci 16. a v první polovině 17. století. Pozornost soustředí především na náboženské polemiky z pera katolických autorů, které byly namířeny proti jednotě bratrské nebo luterské reformaci. Půjde zejména o texty, jejichž autory byli Jakub Horčický z Tepence, Jindřich Ondřej Hoffman a Georg Scherer, jehož díla překládal především Václav Brosius. Stranou pozornosti nezůstanou ani homiletické tisky, které měly apologetický podtext nebo se snažily ovlivnit veřejné mínění. Spolu s polemickou rolí těchto textů, které si již povšimlo starší bádání, se student zaměří na komunikaci jednotlivých textů mezi sebou navzájem pomocí odkazů nebo identických argumentů. Zmíněné texty se pokusí vnímat jako specifický typ média, které mělo nejen polemizovat s oponenty, ale také přesvědčit váhající a utvrdit souvěrce. Kvalifikační spis si rovněž povšimne symbolické řeči studovaných textů v komunikaci mezi duchovními a jejich posluchači nebo čtenáři. The thesis will analyze Czech Counter-Reformation texts published at the end of the 16th and the first half of the 17th century. The student will focus primarily on religious polemics of Catholic authors directed against the Unitas Fratrum or Lutherans. The primary texts will include works by Georg Scherer in the translation of Václav Brosius, Jakub Horčický of Tepence, and Jindřich Ondřej Hoffman. Homiletic prints with an apologetic undertone or those aiming to influence public opinion will not be neglected either. In addition to the polemical role of these texts, which has already been noted by earlier research, the student will focus on the communication between the texts themselves through references or identical arguments. The mentioned texts will be perceived as a specific type of media intended not only to polemicize with opponents but also to persuade the hesitant or to reinforce the faithful. The thesis will also take note of the symbolic language of the studied texts in the communication between the clergy and their listeners or readers.

Předkládaná diplomová práce formou případové studie zkoumá průběh šlechtických zásnub v předbělohorské době. Jako příklad slouží kauza třináctileté šlechtičny Elišky ze Žerotína, které se v letech 1601 až 1604 dvořili dva nápadníci, Petr II. ze Švamberka a Ladislav Berka z Dubé. Když se Eliška ze Žerotína rozhodla pro Ladislava Berku z Dubé, vyvolal Petr II. ze Švamberka soudní spor s Eliščinou matkou Ludmilou Žerotínskou z Kolovrat. Podle jeho názoru byl s Eliškou ze Žerotína zasnouben on, a tak pro něj její svatba s jiným urozeným mužem byla urážkou na cti. Výpovědi svědků v následném soudním sporu slouží jako pramenná základna diplomové práce. Kromě rituálu dvoření a zásnub v předbělohorském Království českém se práce zabývá také průběhem soudního sporu mezi Petrem II. ze Švamberka a Ludmilou Žerotínskou z Kolovrat. Neopomíjí také majetkovou stránku celého konfliktu. The submitted dissertation uses the form a case study to examine the process of betrothal of Czech nobility before the White Mountain. Thirteen years old Eliška of Žerotín serves as an example because she was courted by two aristocrats, Petr II. of Švamberk and Ladislav Berka of Dubé, in the years 1601 to 1604. When Eliška of Žerotín decided to marry Ladislav Berka of Dubé, her second suitor Petr II. of Švamberk started a litigation process against her mother Ludmila Žerotínská of Kolovraty. According to his opinion he was already betrothed to Eliška of Žerotín and so her marriage to another man was an insult to Petr's honour. The witness statements in the ensuing litigation served as the sources for the dissertation. Apart from the ritual of courtship and betrothal in the Czech kingdom before the White Mountain, the entire litigation between Petr II. of Švamberk and Ludmila Žerotínská of Kolovraty is also examined in the dissertation. The question of the estate of all three participants is not neglected either.

Diplomová práce se zabývá vytvářením svatého Jana Nepomuckého v prostoru barokních slavností pořádaných k uctění uvedeného světce především ve dvacátých a třicátých letech 18. století. Autor upře pozornost zejména na kázání coby médium, které se zásadně podílelo nejen na konstrukci obrazu mučedníka zpovědního tajemství, ale rovněž celé slavnosti. Proto se zaměří na rozličné komunikační strategie a postupy kazatelů využívané v průběhu svatojánských festivit k ovlivňování imaginace svého obecenstva. V souvislosti s vytvářením obrazu svatého Jana Nepomuckého z výšin kazatelny se v zorném úhlu diplomové práce rovněž ocitnou aktuální kulturně-historická témata jako například fenomény těla, emocí, smyslů, gest, ale také lidské řeči a jazyka. The master thesis deals with the fabrication of Saint John of Nepomuk in the space of Baroque festivals organized to honour the saint, especially in the 1720s and 1730s. In particular, the author focuses on the sermon as a medium, that was fundamentally involved not only in the construction of the image of the martyr of the seal of confession, but also in the whole festivity. Therefore, he will focus on the various communicative strategies and practices preachers used during the festivities to influence the imagination of their audiences. In connection with the fabrication of the image of the Saint John of Nepomuk from the heights of the pulpit, current cultural and historical themes such as the phenomena of the body, emotions, senses, gestures, as well as human speech and language will also be in the scope of the thesis.

Genetická struktura populací pstruha obecného v oblasti šumavských ledovcových jezer

Tato práce navazuje na předchozí ichtyologické průzkumy v oblasti šumavských ledovcových jezer. Na podzim roku 2022 byl během elektrolovu odebírán genetický materiál pstruha obecného (Salmo trutta L.), ze kterého byly v genetické laboratoři určeny základní populačně genetické charakteristiky. Pro ucelený obraz o prostředí byly zaznamenány i environmentální parametry studovaných toků. This master's thesis is based on ichthyological investigations in the area of the Bohemian Foret's glacial lakes. In the autumn 2022, genetic material of brown trout (Salmo trutta L.) was collected during electrofishing, from which basic population genetic characteristics were determined in the genetics laboratory. Environmental parameters of the study streams were also recorded in order to obtain a comprehensive picture of the environment.

Kininase-like metalloprotease regulates tick salivation

Ticks are obligate hematophagous ectoparasites that require a blood meal to complete their development and reproduction. To facilitate blood feeding, they secrete saliva, a complex mixture of bioactive compounds that modulate the host's immune responses. To evade host immunity, ticks have evolved their salivary proteins to exhibit low immunogenicity. However, in some less adapted hosts, compounds of tick saliva can evoke antibody-mediated responses. Angiotensin-converting enzyme-like kininase (Ir-ACE), the main subject of this thesis, was identified as one of the most dominant salivary antigens of the tick Ixodes ricinus. The aim of this thesis was to investigate the molecular and functional properties of this enzyme; an objective that has not yet been accomplished. To achieve this, RNAi and repeated feeding experiments, proteomic analysis of salivary glands and expression of recombinant Ir-ACE used for the vaccination experiments and substrate profiling were performed.
