A route to a series of novel carbocyclic nucleosides locked in North conformation with bicyclo[3.2.1]octane scaffold was developed. Prepared nucleosides served as a starting material for the synthesis of modified oligomers [d(GCATATCAC), r(GCAUAUCAC), and A9]. Biological effects of the prepared nucleosides as well as the hybridization properties of the appropriate duplexes were evaluated. Bi-substrate inhobitors of human pyrimidine 5’-nucleosidases

The work is aimed at the synthesis of bi-substrate inhibitors of 5’-nucleosidases. The pyrimidine nucleobase of lead compound was derivatized in the 5-position with various phosphonic acids differing in conformational flexibility and in the length of alkyl side chain carrying phosphonate group. The phosphonic acid with optimal linker can reach the phosphate binding side and make strong interaction with the enzyme. Inhibition activity of such inhibitors and crystal structure of inhibitor-enzyme complex will be presented. Voltammetric Behavior of 4-Aminophthalimide Label using Hanging Mercury Drop Electrode

4-aminophthalimide (API), covalently attached to nucleotide (dCTP or dATP) has recently beendesigned as a fluorescent label for the detection of DNA-protein interaction. However API is electrochemically reducible and therefore it is possible to use alternatively the API-modified nuclotides as DNA redox labels. The electrochemical behavior of labeled nucleotides and labeled DNA was studied using cyclic voltammetry and transfer stripping cyclic voltammetry at hanging mercury drop electrode. Observed CVs showed irreversible reduction signals of 4-aminophthalimide label without production of consequently oxidizable/reducible species. Voltametrická analýza nukleosidtrifosfátů a oligonukleotidů značených antrochinonem na zlatých elektrodách

Elektrochemická aktivita nukleových kyselin byla objevena v 50. letech 20. století a od té doby je používána ke studiu struktury a interakcí přirozených i modifikovaných molekul nukleových kyselin i syntetických oligonukleotidů. Přestože je přirozená DNA sama o sobě elektrochemicky aktivní a je možné ji studovat na různých typech elektrod, je pro řadu analytických aplikací praktické použít DNA značenou elektroaktivními molekulami (například komplexy přechodných kovů, amino- nebo nitroskupinami, antrachinonem apod.). Tyto látky podléhají redoxním reakcím a dávají takto modifikované DNA nové elektrochemické vlastnosti, které je možné využít pro studium struktury a interakcí nukleových kyselin a v oblasti DNA diagnostiky. DNA labelling is used for increase of sensitivity of electrochemical detection. When the DNA or oligonucleotides (ONs) are chemically modified, their electrochemical responses can be changed depending on electrochemical activity of the introduced moiety. One option of DNA labeling is incorporation of chemically modified nucleotides using corresponding deoxynucleotide triphosphates (dNTPs) into DNA molecules with using primer extension (PEX). In our study we used anthraquinone labeled dNTPs and ONs with enzymatically incorporated anthraquinone labeled nucleotides. Thus synthesized ON stretch bearing the anthraquinone tags was electrochemically analyzed using voltammetry at gold electrodes. Separation of nonpolar lipids from vernix caseosa

The aim of this work is to develop a separation method for characterization of unknown nonpolar lipids in vernix caseosa. Here we optimized adsorption chromatography coupled to mass spectrometry detection with exact mass measurement. A long silica gel column (250 + 250 x 4.6 mm, particle size: 5 mu m) provided the best separation of nonpolar lipids present in vernix caseosa.
