This contribution is devoted to quantum dots usage for cell imaging in fluorescence microscopy. Fluorescence excitation by evanescence wave (caused by total internal reflection) is compared with conventional epifluorescence microscopy. Příspěvek je věnován využití kvantových teček pro značení buněk ve fluorescenční mikroskopii. Zejména je srovnána excitace fluorescence evanescentní vlnou (vznikající při totálním interním odrazu) s konvenčním epifluorescenčním uspořádáním.

Characterization of a new 8-hydroxyquinoline modifed resin gel for DGT technique

This study deals with a new resin gel based on Spheron-Oxin® ion exchanger with anchored specific groups of 8-hydroxyquinoline for DGT technique. The performance of this new binding phase was tested for determination of Cd, Cu, Ni, Pb and U under laboratory conditions. The new binding phase with a higher selectivity to race metal species than Chelex 100 could potentially provide more information on metals speciation in aquatic systems.

The method of chemical vapour generation of silver by means of sodium tetrahydroborate was futher developed in order to minimize transport losses. The possibility of collection of volatile silver species directly in the atomizer is presented. Metoda generování těkavých forem stříbra pomocí reakce s roztokem tetrahydroborátu byla dále vyvinuta za účelem snížení transportních ztrát. Je prezentována možnost provedení kolekce stříbra přímo v křemenném atomizátoru.

A method of silver chemical vapour generation as a sample introduction technique for analytical atomic spectrometry is presented. Optimization of reaction and atomization parametrs was carried out and the efficiency of the chemical vapour generation was studied by means of radiotracer experiments. V práci je představena metoda chemického generování těkavých forem stříbra jako metoda vnášení vzorku pro analytickou atomovou spektrometrii. Byla provedena optimalizace reakčních a atomizačních podmínek a byla stanovena účinnost generování těkavých forem stříbra pomocí experimentů s radioaktivními izotopy stříbra.

This work represents an improvement of current method of speciation analysis for determination of tri- and pentavalent inorganic, mono-, di-, and trimethylated arsenicals using atomic absorption spectrometry as a detector. Presented study shows that thioglycolic acid is suitable for an on-line pre-reduction which simplifies sample pretreatment and increases sample throughput. Práce představuje zlepšení současné metody speciační analýzy pro stanovení trojmocných a pětimocných sloučenin arsenu za použití detekce atomovou absorpční spektrometrií. Kyselina thioglykolová byla shledána vhodná pro on-line předredukci a ulehčuje tak podstatně proces předúpravy vzorků.

An optimization study of arsine and selenum hydride preconcentration at the quartz surface is presented. A novel apparatus design consisting of a quartz trap integrated with a quartz multiatomizer is described. Práce popisuje optimalizaci podmínek prekoncentrace hydridů arsenu a selenu na křemenném povrchu. Pro tento účel byl navržen nový design aparatury sestávající z křemenného prekoncentračního zařízení integrovaného s křemenným multiatomizátorem.

A novel design of hydride multiatomizer was described. The performence of several modifications of the multiatomizer with variety of optical tubes, especially porous quartz and multiple perforated quartz, has been discussed. Představen byl nový typ multiatomizátoru hydridů. Diskutovány byla jeho funkčnost v závislosti na užití různých typů optických trubic, zejména z porézního přemenného skla a z křemenného skla mnohonásobně děrovaného.

Microfluidic devices are expected to speed up the progress in bioanalysis and separation sciences. Ideally, these Lab-on-chip systems integrate all steps of analysis on a one microdevice; however, before the integration, every single unit operation has to be first developed and tested separately. One of such important unit operations is the cell lysis and the following release of the cellular content. Od mikrofluidických systémů se očekává urychení pokroku v bioanalýze a separačních vědách. V ideálním případě systémy Lab-on-chip integrují všechny kroky analýzy v jednom mikrozařízení; ovšem před integrací musí být každá jednotlivá operace vyvinuta a otestována samostatně. Jednou takovou důležitou operací je buněčná lýza a následné uvolnění buněčného obsahu.
