CE-nESI/MS in electrode-free design: narrowing the separation channel to 5 μm

Capillary electrophoresis-nanospray/mass spectrometry (CE-nESI/MS) in electrodefree\narrangement represents a very simple instrumentation for analysis of biomolecules.\nSo far, CE-nESI/MS analyses in this design were limited to 10-75 μm ID of the\nseparation channel. Although the work with narrower dimensions brings several\nchallenges, there is a great potential for better sensitivity, improved separation power\nand lower sample consumption. This work is devoted to some practical aspects of\nexperiments in narrow bore channels and systematic evaluation of CE-nESI/MS\nanalyses conducted in capillaries of 25, 15 and 5 μm ID. Comparative evaluation of protein composition in human breast cancer cells using mass spectrometry

Bottom-up proteomic approach was used for detailed characterization of proteins from\ntwo human tumour cell lines representing major clinically different types of breast\ncancer. The aim was to show the differences between them on proteomic level. Here\nwe present almost 100 unequivocally identified proteins out of which 60 were mutually\ndifferently expressed for MDA-MB-231 and MCF-7. Some well-known breast cancer\nmarkers like annexins A1, A2 and vimentin were found in the MDA-MB-231 cell line.\nOn the other hand, MCF-7 cells were found to be positive for cytokeratins and keratins\nand thus we were able to distinguish both cell lines sufficiently. Hexahistidine labeling of oligosaccharides

Simple route of caspase-3 FRET sensor synthesis using “click chemistry”

Programmed cell death or apoptosis is regulated process of cell suicide. The central role in apoptosis play cysteine proteases called caspases. Caspases recognize tetra-peptide sequences Asp-Glu-Val-Asp (DEVD) on their substrates and hydrolyze peptide bonds after aspartic acid residues. Various techniques for the determination of caspase-3 are commercially available e.g. Enzyme Linked Immuno-Sorbent Assay (ELISA), Western blotting or flow cytometric analysis. The products of the cleavage can be detected by spectrophotometry, fluorimetry, chemiluminescence (CL) or ELISA. In this work, we suggested fluorescent sensor based on easily prepared Förster Resonance Energy Transfer (FRET). We use very simple chemistry called “click”. This type of chemistry takes advantages of quickness, simplicity and cheapness. “Click chemistry” is based on usage of various functional group and cross-linkers to combine individual molecules together. Three-stage labeling strategy allows the preparation of specific conjugates composed of required antibodies and quantum dots as an extremely sensitive bio-probe applicable in varied immunoassays with following optical and electrochemical detection.
