Využití různých metod izolace entomopatogenních a mykoparazitických hub z různých podmínek prostředí

Abstrakt Hlavním cílem této práce bylo otestovat metody izolace entomopatogenních hub z různých podmínek prostředí. V prním experiment byly inokulovány entomopatogenní houby B. bassiana M. brunneum, I. fumosorosea a L muscarium ve dvojích do sterilního půdního substrátu a inkubovány po dobu 50 dnů. Analýza kompatibility byla prováděna po 14 a 50 dnech inkubace. Výsledky byly porovnávány s výsledky v době 0, kdy byly z kontrolních suspenzí založeny testy, při kterých se hodnotil počet tvořících se kolonií (CFU, colony-forming unists). Výsledky ukázaly, že lze z půdy snadno izolovat entomopatogenní houby za pomoci rutinní metody použití selektivní živné půdy dodine. Zároveň byla z fotografické dokumentace zaznamenána interakce mezi jednotlivými druhy. Druhý experiment byl zaměřen na aplikaci entomopatogenní houby I. fumosorosea a L muscarium a mykoparazitické houby T. virens na rostliny okurek. Z výsledků tohoto experimentu též vyplývá, že jak entomopatogenní houby, tak mykoparazitickou houbu lze i z fyloplánu snadno izolovat entomopatogenní houby pomocí selektivní živné půdy na bázi dodine a mykoparazitickou houbu T. virens po-mocí selektivní živné půdy TSM (Trichoderma selektive medium). Zároveň bylo prokázáno, že relativní vzdušná vlhkost je důležitá pro růst a vývoj entomopatogenních hub po preventivní aplikaci na rostliny okurky seté. Abstract The main objective of this work was to test methods for isolating entomopathogenic fungi from different environmental conditions. In the first experiment, entomopatho-genic fungi B. bassiana, M. brunneum, I. fumosorosea and L. muscarium were inocu-lated in pairs into sterile soil substrate and incubated for 50 days. Compatibility anal-ysis was performed after 14 and 50 days of incubation. The results were compared with those on day 0 when the control suspensions were used to establish tests to assess the number of colony-forming units (CFU). The results showed that entomopathogenic fungi can be easily isolated from soil using the routine method of dodine selective nutrient soil. At the same time, the interaction between species was recorded from photographic documentation. The second experiment focused on the application of the entomopathogenic fungi I. fumosorosea and L muscarium and the mycoparasitic fun-gus T. virens to cucumber plant leaves. The results of this experiment also show that both entomopathogenic fungi and mycoparasitic fungus can be easily isolated even from the phylloplane entomopathogenic fungi using dodine-based selective medium and mycoparasitic fungus T. virens using Trichoderma selective medium (TSM). It was also shown that relative humidity is important for the growth and development of entomopathogenic fungi after preventive application to cucumber plant leaves. This thesis focuses on the influence of wheat varietal mixtures on yield stability, grain quality and flour rheological properties. The aim of this study was to improve wheat yields and includes further mixing of wheat varieties through crossing, interspecific and intergeneric hybridization, biotechnology techniques, wheat mixtures. The growth of wheat mixtures will lead to yield stability, then it will lead to baking quality stability, and the rheological properties of flour will be positively influenced by the growing wheat in mixtures. This thesis focuses on the influence of wheat varietal mixtures on yield stability, grain quality and flour rheological properties. Využití biopreparátů na bázi mikroorganismů k biologickému ošetření osiva vybraných kulturních plodin

Cílem diplomové práce byla zaměřena na biologické ošetření osiva kukuřice, sóji a okurky pomocí entomopatogenní houby Beauveria bassiana, mykoparazitické houby Trichoderma virens a antagonistické bakterie Bacillus mycoides. Pro obalení semen byl použit 0.5% roztok karboxymethylcelulóza, ve kterém byla v 1 ml kon-centrace spor 1x106. Po namoření osiva byl stanoven počet spor vláknitých hub na jednom semeni a zároveň byl sledován vliv mikroorganismů na klíčivost semen a vývoj sazenic. Z výsledků vyplývá, že pozitivní efekt na růst a vývoj sazenic všech testovaných plodin měla antagonistická bakterie B. mycoides a entomopatogenní houby B. bassiana. Naopak mykoparazitická houba T. virens slabě retardovala během klíčení vývoj kořínků a klíčků u kukuřice a sóji. Ošetřená semena okurek pomocí T. virens vykázala rychlejší vývoj obou hodnocených parametrů. Setím mořeného osiva pomocí užitečných mikroorganismů lze snadno vnést tyto mikroor-ganismy do půdy a tím zvýšit supresivitu půd. Klíční rostlinky tak mohou být chráněny proti půdním škůdcům nebo původcům onemocnění rostlin. The aim of the thesis was focused on the biological treatment of corn, soybean and cucumber seeds using the entomopathogenic fungus Beauveria bassiana, mycopara-sitic fungus Trichoderma virens and antagonistic bacteria Bacillus mycoides. A 0.5% solution of carboxymethylcellulose was used for seed coating, in which the concentration of spores in 1 ml was 1x106. After seed coating, the number of spores of filamentous per one seed was determined and also the influence of microorgan-isms on seed germination and seedling development was evaluated. The results show that the antagonistic bacterium B. mycoides and entomopathogenic fungi B. bassiana had a positive effect on the growth and development of seedlings of all tested crops. The mycoparasitic fungus T. virens slightly retarded the development of roots and shoots in corn and soybean during germination. On the other hand, cu-cumber seeds coated by T. virens showed a faster development on both parameters evaluated. By sowing coated seeds with useful microorganisms, which can be easily introduced into the soil in addition increasing soil suppressiveness. Thus, seedlings can be protected against soil-dwelling pests or plant pathogens. Diplomová práce se zaměřuje na komparativní analýzu děl dvou autorek americké jižanské literatury - Flannery O'Connor a Carson McCullers. Zvláštní důraz je kladen na posouzení vztahu gotických a realistických prvků v jejich dílech. Východiskem analýzy je charakteristika jižanské gotické literatury (hlavně místa a prostředí), na jejímž základě jsou stanoveny hlavní znaky tohoto žánru. Samostatné kapitoly jsou věnovány životní zkušenosti obou autorek. Vlastní komparativní analýza je zaměřena na charakteristiku postav a jejich vztahů, funkci prostředí, význam groteskních prvků, spojení tragična a komična, vliv minulosti, úlohu náboženství a otázku dobra a zla. The diploma thesis focuses on a comparative analysis of Flannery O'Connor's and Carson McCullers' Southern Gothic literary works. Special emphasis is put on the relationship between the gothic and realistic features. The comparative analysis is based on the characterisation of Southern Gothic literature (the landscape and environment as the main focus), which leads to the specification of its main features. Individual chapters contain O'Connor's and McCullers' biographies. The comparative analysis itself concentrates on the description of the heroes and their relationships, the function of landscape, the meaning of grotesque motifs, the connection of traditional and comical features, the influence of past, the role of religion and the issue of good and evil. Juvenile hormone (JH) plays a key role in insect development. In larval stage, JH prevents the insect to metamorphose. While JH is unique to arthropods and absent from vertebrates, it represents potential tool for insect control. Combination of cell-based assay and high-throughput screening enabled to find candidate JH agonists (JHags) that activate JH signaling via juvenile hormone receptor (JHR). This thesis aims to elucidate the effect of these JHags on three evolutionary distinct insect species in vivo and possible selectivity of the JHags for individual insect species in vitro. In addition, we provide evidence that JHR from hemimetabolous species Blattella germanica and Prorhinotermes simplex binds JH in vitro. Habitat selection of forest birds in Branišovský les, České Budějovice, Czech Republic was studied by observing habitat preferences in the non-breeding season and exploring possible shifts in preferences due to the changing seasons. Patterns of habitat selection were analysed at multiple levels: considering all bird species as well as comparing between two feeding guilds. Preferences were also studied by specifically monitoring predation rates by birds on artificial caterpillars and exploring changes with habitat and season. Relationship between personality and social network characteristics

This thesis investigated whether an individual's personality (assessed on four personality dimensions: Friendliness, Activity/Excitability, Confidence, Openness) can be related to its social network position in Barbary macaques (Macaca sylvanus). The effect of sex and rank was also evaluated. The network position of individuals was described by individual network characteristics values generated via social network analysis. Additionally, the phenotypic based assortment within the studied group was examined. The results showed that personality is related with the position of the individual in its social group and assortment based on several personality dimensions was found in studied networks. Elucidating the source of bloodstream \kur{Trypanosoma brucei} mitochondrial ATP

For decades, it has been assumed that the reduced function and structure of the bloodstream form Trypanosoma brucei mitochondrion renders it a strictly ATP consuming organelle. Emerging evidence from refocused studies suggest that the bloodstream mitochondrion retains complex bioenergetic pathways that allow the parasite to adapt to various environments. This thesis is focused on the source of bloodstream mitochondrial ATP, with a special emphasis on the role of succinyl-CoA to produce ATP via mitochondrial substrate phosphorylation. We will also discuss alternative bioenergetic pathways present in this life stage of a human pathogen.
