Aktivity of thioredoxin reduktase in wild and seleniumresistant strains of the alga Scenedesmus quadricauda grown in the presence of inorganic selenium compounds

We have selected resistant strains of Scenedesmus quadricauda able to tolerate high selenium concentration up to 400 mg/L added either as selenate Na2SeO4 (strain SeVI) or selenite Na2SeO3 (strain SeIV). We have investigated activity of selenoprotein thioredoxin reductase (TR) in both wild type and Se-resistant strains. Concomitantly, a total amount of organically bound selenium was evaluated in biomass dry weight. We have found that activity of TR rapidly increases in the SeVI resistant strain in the presence of selenate, while no effect on TR activity was found in SeIV resistant strain and only slight increase in wild type in the presence of selenite Vyselektovali jsme rezistentní kmeny Scenedesmus quadricauda schopné tolerovat vysoké koncentrace selenu až do 400 mg/L přidávaného buď ve formě selenanu Na2SeO4 (kmen SeVI) nebo seleničitanu Na2SeO3 (kmen SeIV). Studovali jsme aktivitu selenoproteinu thioredoxin reduktázy (TR) jak u divokého tak u rezistentních kmenů. Zároveň jsme stanovovali celkové množství organicky vázaného selenu v sušině. Zjistili jsme, že aktivita TR se rapidně zvyšuje u SeVI rezistentního kmene v přítomnosti selenanu, zatímco u SeIV rezistentního kmene jsme nezaznamenali žádný efekt a pouze mírné zvýšení aktivity u divokého kmene v přítomnosti seleničitanu

T. weissflogii was grown under dynamic irradiance conditions (sinusoidal light-dark regime) and then switched to constant, low irradiance. Cellular characteristics (pigment and nutrient content, cell size) indicated synchronization of algal population, furthermore pronounced variations in the aktivity of variable fluorescence, thermoluminescence and in the gene expression (rbcL, psbA, fcp) were observed T. weissflogii byla pěstována v podmínkách dynamické ozářenosti (sinusoidní režim) a poté přenesa na konstantní světlo o nízké intenzitě. Buněčné charakteristiky (obsah pigmentů a nutrientů, velikost buněk) indikovaly synchronizaci řasové populace, dále byly pozorovány výrazné změny v aktivitě variabilní fluorescence, termoluminiscence a v expresi genů (rbcL, psbA, fcp)

Anthropogenic increases in atmospheric pCO2 over the last century are accompanied by higher concentrations of CO2 (aq) in the surface oceans. These changes could influence aquatic primary production including cyanobacterial nitrogen-fixers via the coupling between photosynthesis and nitrogen-fixation. We hypothesized that elevated pCO2 levels may enhance nitrogen fixation and photosynthesis in the bloom-forming, diazotrophic, cyanobacterium Trichodesmium spp. that contributes greatly to primary production in the oligotrophic subtropical and tropical oceans. We acclimated cultures of Trichodesmium IMS101 to 3 pCO2s (pre-industrial -250 ppmv, current - 400, future -900 ppmv) and examined changes in morphology, photosynthesis, nitrogen fixation, and growth. High pCO2 enhanced growth, filament length and nitrogen fixation of Trichodesmium in comparison with both ambient and low pCO2 acclimated cultures Antropogenní zvýšení atmosférického CO2 může ovlivnit i mořské diazotrofní sinice, které fixují atmosferický uhlík. Ukázali jsme, že zvýšení CO2 má pozitivní vliv na rychlost růstu a fixace dusíku u sinice Trichodesmium. Pravděpodobným mechanismem je alokace energie a reduktantů od mechanismů koncentrace uhlíku v buňkách k mechanismům fixace dusíku

Characterization of a restriction modification system from the commensal Escherichia coli strain A0 34/86 (O83:K24:H31)

We have characterised a putative restriction-modification system EcoA0ORF42P in commensal Escherichia coli strain A0 34/86 (O83: K24: H31). This system is a functional member of the Type IB family, whose specificity differs from those of known Type IB enzymes, as proved by immunological cross-reactivity and complementation assay. Using a plasmid transformation method and RM search computer program, we have identified the DNA recognition sequence of EcoA0ORF42P as GGA(8N)ATGC Charakterizovali jsme předpokládaný restrikčně-modifikační systém EcoA0ORF42P komensálního kmene Escherichia coli A0 34/86 (O83:K24:H31). Imunologická analýza a komplementační testy prokázaly, že se jedná o funkční systém patřící do skupiny Typu IB, jehož specifita se liší od dosud známých enzymů této skupiny. Byla identifikovaná specifická sekvence na DNA GGA(8N)ATGC rozpoznávaná enzymem EcoA0ORF42P

In those experimenst we studied the possible impace of CD43 as highly glycosylated molecule in cells exposed to free doxorubicin (Dox) or to conjugates consisting of HPMA copolymer backbone to which the drug is conjugated via enzymatically (PK1 conjugate) or pH sensitive, hydrolytically cleavable bond (hydrazone conjugate, HYD). V těchto pokusech jsme sledovali vliv vysoce glykosylované povrchové molekuly CD43 u buněk vystaveným volnému doxorubicinu nebo doxorubicinu konjugovanému k HPMA polymeru
