Identifikace a vzdělávání nadaných dětí v mateřské škole

2024 - Czech The bachelor thesis focuses on the issue of identification of a gifted child in kindergarten and pedagogical procedures used by the teacher in subsequent work with a gifted child. The aim of the thesis is to find out how kindergarten teachers contribute to the development of gifted children and the full use of their potential. The theoretical part of the thesis in the introduction pays attention to the general introduction to giftedness, then it deals with the characteristics of gifted children and their subsequent identification. In the conclusion, the legislative documents related to the education of gifted children are presented, followed by the possibilities of development and education in the kindergarten environment and the essentials of the teacher's work with gifted children. The empirical part of the thesis analyses the manifestations of potential giftedness in children in kindergarten and aims to identify the pedagogical methods applied when working with gifted children. The research was conducted using a mixed-methods approach, specifically a sequential combining design (QUAL-quan scheme) that employed a case study design and a questionnaire survey. Bakalářská práce se zabývá problematikou identifikace nadaného dítěte v mateřské škole a pedagogickými postupy, které jsou využívány učitelem při následné práci s nadaným dítětem. Cílem práce je zjistit, jakým způsobem vyučující mateřských škol přispívají k rozvoji nadaných dětí a plnému využití jejich potenciálu. Teoretická část práce v úvodu věnuje pozornost obecnému seznámení se s nadáním, dále se zabývá charakteristikou nadaných dětí a jejich následnou identifikací. V závěru jsou uvedeny legislativní dokumenty spojené se vzděláváním nadaných dětí, na ně navazující možnosti rozvoje a vzdělávání v prostředí mateřské školy a náležitosti práce pedagoga s nadanými dětmi. Empirická část práce analyzuje projevy potencionálního nadání u dětí v mateřské škole a cílí na zjištění pedagogických metod uplatňovaných při práci s nadanými dětmi. Výzkum byl zpracován smíšeným přístupem, konkrétně pomocí sekvenčního kombinování (schéma QUAL-quan), který využíval design případové studie a dotazníkového šetření. Studium struktury a interakcí proteinů pomocí strukturní hmotnostní spektrometrie

2024 - Czech Transmembrane channels and transporters of the ClC protein family are present across all living organisms. They are found on the cytoplasmic and lysosomal membranes of the cells, where they participate in maintaining ion homeostasis. When dysfuncional, they lead to serious health complications. To develop treatment for these diseases, it is essential to describe transport mechanism of ClC proteins. The antiporter ClC-ec1 from E.coli is used as a model protein for the entire ClC protein family. This homodimeric protein, which transports one proton against two chloride ions, has a separate transport path in each monomer. Based on the crystal structure, it is believed that during transport the protein alternates between outward and inward-facing conformations. Conversion to the outward-facing conformation of the protein is accompanied by the protonation of three glutamates located in the transport path. To study these conditions, a QQQ mutant was designed that has these glutamates replaced by glutamines. Until now, the study of the transport mechanism of ClC-ec1 has mainly relied on studies based on X-ray crystallography. Crystallography provided static images, which did not contain sufficient information about protein dynamics. Therefore, to study transport mechanism of ClC-ec1, we chose a dynamic... Transmembránové kanály a přenášeče z ClC proteinové rodiny se vyskytují napříč všemi živými organismy. Nacházejí se na cytoplasmatických a lysosomálních membránách buněk, kde se podílí na udržování homeostázy iontů. Narušení jejich funkce vede k vážným zdravotním komplikacím. Pro vývoj efektivní léčby těchto onemocnění je potřeba objasnit transportní mechanismus ClC proteinů. Antiporter ClC-ec1 z E.coli je modelovým proteinem celé ClC proteinové rodiny. Tento homodimerní protein, který transportuje proton proti dvěma chloridovým iontům, má transportní dráhu v každé z podjednotek. Na základě krystalové struktury se přepokládá, že během transportu se protein střídavě otevírá na obě strany membrány. Otevření proteinu směrem ven je umožněno protonací tří glutamátů, které se nacházejí v transportní dráze a jsou pro transport iontů klíčové. Pro studium tohoto stavu byl navržen QQQ mutant, který má tyto glutamáty nahrazeny za glutaminy. Dosud se studium transportního mechanismu ClC-ec1 opíralo převážně o studie založené na rentgenové krystalografii. Krystalografie poskytla statické obrázky, které neobsahovaly dostatečné informace o dynamice proteinu. 2024 - English This bachelor's thesis aims to explore the unconventional register used in Sapphire's novel PUSH. The practical section of the thesis is dedicated to translating the first chapter and the final two paragraphs of the novel from the English language into the Czech language. The theoretical section of the thesis focuses on stylistic analysis of the translation, examining various textual elements such as register, vulgarisms, idiomatic expressions, and place names. The practical section also considers and explains the decisions made during the translation process. The thesis also includes chapters dealing with Sapphire's biography and a brief introduction to translation theory, discussing the theories of Jiří Levý, Dagmar Knittlová, Theodore Savory and Eugene Nida. Tato bakalářská práce si klade za cíl prozkoumat nekovenční řečový registr obsažený v Sapphiřině románu PUSH. Praktická část práce je věnována anglicko-českému překladu první kapitoly a závěřečných dvou odstavců knihy. Teoretická část práce se zaměřuje stylistickou analýzu překladu a zkoumá různé textové prvky, jako je řečový registr, vulgarismy, idiomatické výrazy či místopisné názvy. V praktické části jsou dále zohledněna a vysvětlena rozhodnutí učiněná během samotného procesu překladu. Součástí práce jsou také kapitoly zabývající se autorčiným životopisem a stručným úvodem do teorie překladu, v němž jsou mimo jiné rozebrány teorie Jiřího Levého, Dagmar Knittlové, Theodora Savoryho a Eugena Nidy. 2024 - Czech This work deals with the issue of future planning in families with a child with mental disabilities. The theoretical part outlines the definition of a family, the specifics of families with a child with mental disabilities, social services for adults with mental disabilities, and the specifics of future planning for people with mental disabilities. Within the practical part of the bachelor thesis, the methodology of the chosen approach is defined, and the results of the research survey are interpreted. The aim of the work is to find out how families with a child with mental disabilities approach future planning. The chosen research method is semi- structured interviews with ten respondents. The testimonies of parents and siblings of children with mental disabilities provided insight into the approach to future planning, the difficulties associated with this topic, and the unique stories of individual families. Tato práce se zabývá problematikou plánování budoucnosti v rodinách s dítětem s mentálním postižením. V teoretické části je vymezena definice rodiny, specifika rodiny s dítětem s mentálním postižením, sociální služby pro dospělé lidi s mentálním postižením a specifika plánování budoucnosti lidí s mentálním postižením. V rámci praktické část bakalářské práce je vymezena metodologie zvoleného přístupu a interpretovány výsledky výzkumného šetření. Cílem práce je zjistit, jakým způsobem přistupují v rodinách s dítětem s mentálním postižením k plánování budoucnosti. Zvolenou metodou výzkumu je polostrukturovaný rozhovor s deseti respondenty. Výpovědi rodičů a sourozenců dětí s mentálním postižením poskytly náhled do přístupu a plánování budoucnosti, obtížích s tímto tématem spojených a jedinečných příběhů jednotlivých rodin. 2024 - Czech This bachelor thesis aims to describe the history of the creation and subsequent second life of two selected monuments in Chodsko. This includes a description of the initial idea regarding the creation of the monuments, the course of fundraising activities, the selection of the author, the unveiling ceremony, and the subsequent second life of the monument during the World War II. The thesis also looks at how the buildings were received by the general public and places the selected monuments in the context of the period. The primary sources of information are mainly contemporary documents, periodicals, correspondence from individual committees, chronicles and photographs. In describing the events or persons to whom the monuments are dedicated, the work is based primarily on scholarly literature. Tato bakalářská práce si klade za cíl popsat historii vzniku a následný druhý život dvou vybraných památníků na Chodsku. To zahrnuje popis prvotní myšlenky ohledně vzniku monumentů, průběhu sbírkové činnosti, výběru autora, slavnostního odhalení a následného přiblížení druhého života pomníku za druhé světové války. Práce se také věnuje tomu, jak byly stavby přijaty širokou veřejností a zasazuje vybrané památníky do kontextu doby. Přičemž hlavním zdrojem informací jsou převážně dobové dokumenty, periodika, korespondence jednotlivých výborů, kroniky a fotografie. Při popisu událostí či osob, jimž jsou pomníky věnovány, vychází práce primárně z odborné literatury. 2024 - Czech Hemoproteins are diverse and very important group of proteins involved in a wide range of biochemical processes. Their typical roles in living organisms include oxygen storage and distribution, enzymatic catalysis of redox reactions and electron transport in the respiratory chain. In relatively recent times, a fourth group of hemoproteins, the so-called heme sensor proteins, has been discovered. This group includes, among others, proteins that have the ability to reversibly bind heme, which in such cases acts as a signaling molecule. This reversible interaction is essential for cells in certain signalling pathways. As it was found, one of the heme sensor proteins is the p53 protein, which is an important transcription factor The first phase of this diploma thesis was focused to the preparation of p53 family proteins including some of their deletion mutants. Subsequently, the task for selected proteins was to study the influence of heme on their oligomeric state. The essential part of the thesis consists of spectrophotometric studies, which aimed to reveal potential interactions of the studied proteins with heme and to characterize the interactions. Keywords: heme, heme sensor proteins, p53 protein family, transcription factors, signal transduction Hemoproteiny jsou rozmanitou a velmi významnou skupinou proteinů podílející se na celé řadě biochemických pochodů. Mezi jejich typické role v živých organismech patří uskladňování a distribuce kyslíku, enzymová katalýza redoxních reakcí a transport elektronů v dýchacím řetězci. V relativně nedávné době byla objevena čtvrtá skupina hemoproteinů, tzv. hemové senzorové proteiny. Do této skupiny náleží mimo jiné proteiny, které disponují schopností reversibilně vázat hem, jenž takových případech plní funkci signální molekuly. Tato reversibilní interakce má pro buňky zásadní význam v některých signalizačních drahách. Jak bylo zjištěno, jedním z hemových senzorových proteinů je také významný transkripční faktor, protein p53. První fáze předkládané diplomové práce byla věnována přípravě proteinů rodiny p53 včetně některých jejich delečních mutantů. Následně bylo úkolem u vybraných proteinů prostudovat vliv hemu na jejich oligomerní stav. Zásadní část diplomové práce je tvořena spektrofotometrickými studiemi, které si kladly za cíl odhalit potenciální interakce studovaných proteinů s hemem, popřípadě dané interakce charakterizovat. 2024 - Czech The bachelor thesis deals with the wives of Sigismund of Luxembourg, Maria of Anjou and Barbara of Cilli. Through the study of sources, especially chronicles and secondary literature, domestic and foreign, it provides a closer look at her life destinies. The thesis is divided into two stages, each is about one of the wives. At the beginning of each section, there is a description and a closer look to the family of Anjou and family of Cilli, i.e., two important families which both women come from. In the chapters themselves, the work deals with the wives from their childhood to being at the ruler's side, or even their existence after Sigismund's death. In the case if the second wife, Barbara of Cilli, her second life is also mentioned here with an overlap into the present. It is explained how today's society looks at her and for what reason it is so. Their female position in the late medieval society is also mentioned. Prostřednictvím studia pramenů, zejména kronik a sekundární literatury, domácí a zahraniční, blíže přibližuje jejich životní osudy. Práce je rozdělena do dvou stěžejních částí, kdy se každá zabývá právě jednou z žen. Na začátku každého úseku dochází k popisu a bližšímu přiblížení rodu Anjouovců a hrabat z Celje, tedy významných rodin, ze kterých obě ženy pocházejí. V kapitolách samotných se práce zaobírá manželkami od dětství až po působení po boku vládce. V případě Barbory z Celje je zmiňován i její život po Zikmundově smrti a takzvaný druhý život s přesahem do současnosti. Je vysvětleno, jak se na ní dívá dnešní společnost a z jakého důvodu tomu tak je. V neposlední řadě je zkoumáno postavení ženy v tehdejší pozdně středověké společnosti.
