Clustering of genes with similar regulation patterns during monocyte and granulocyte differentiation of HL-60 cells

In the present study, a high density-microarrays of human cDNA containing 19,000 genetic elements were used to search for differences in gene expression of human promyelic leukaemia cell line HL-60 during monocyte (MD) and granulocyte differentiation (GD). For differentiation to monocytes (granulocytes), the TPA, 12-O-tetradecanoylphorbol-D-acetate (DMSO) were used, respectively. A total number of 4625 (4760) genes were found to be regulated during MD (GD). The genes have been divided into groups according to their kinetics of regulation (R-groups). The genes of the same group are not distributed randomly throughout the genome but form clusters on chromosomes (R-clusters). Thus, functionally interconnected genes are frequently localized near to each other on the DNA molecule. V předkládané studii byly pomocí vysokokapacitních cDNA čipu obsahujících 19,000 sekvencí, provedeny studie expresních profilů promyelotických leukemických buněk HL-60 diferencujících do monocytů (MD) a granulocytů (GD). Jako diferenciační činidlo pro monocytární diferenciaci bylo použito TPA (12-O-tetradecanoylphorbol-D-acetate) a pro granulocytární proces DMSO. Studiem expresních profilů bylo zjištěno že v průběhu MD signifikantně měnilo expresi 4625 genů a v průběhu GD 4760 genů. Geny obou diferenciací byly rozděleny do podle typu kinetiky s jakou se účastní diferenciace do tzv. R-skupin. Bylo zjištěno, že geny z jednotlivých R-skupin nejsou nenáhodně rozprostřeny na chromosomových teritorií, ale formují na chromosomech klastry. Skupiny genu s podobnou kinetikou regulace tedy můžeme najít na chromosomech blízko sebe.

Image analysis of the movement of sub-cellular structures

Analysis of time-lapse series of images of living cells requires automation. This contribution concerns the main features of the software package developed for this purpose in our laboratory. Some improvements of our recently published method for tracking of sub-cellular structures based on graph theory are presented. The behaviour of the method is evaluated on images of centromeric heterochromatin defined by HPlbeta-GFP fusion protein. The results show that the method is very well applicable for this type of data. Analýza časových sérií obrázů živých buněk vyžaduje automatizaci. Tento příspěvek popisuje hlavní rysy softwarového balíku, který byl k tomuto účelu vytvořen v naší laboratoři. Prezentuje několik vylepšení námi nedávno publikované metody pro sledování pobybu buněčných struktur. Chování metody je vyhodnoceno na obrázcích heterochromatinových oblastí definovaných HP1beta-GFP fúzním proteinem. Výsledky ukazují, že pro tento typ dat je navržená metoda velmi vhodná.

The aims of the present study were to determine potencies of a series of environmentally relevant PCB congeners to inhibit GJIC invitro, and to screen for intrcellular signaling pathways potentially involved in GJIC downregulation by PCBs. It was found that a number of nonplanar PCB congeners are efficient inhibitors of GJIC in vitro, and that a model compound, PCB153, does not act via activation of PKC or ERK1/2, which is contrast with the effects of protoypical inhibitors of GJIC, such as TPA or EGF.

The article concentrates on the latest knowledge concerning the role of epigenetic mechanisms in sex determination in model dioecious species Silene latifolia. Results suggest that the locus suppressing development of female organs in male plants is hypersensitive to the action of agents influencing level of DNA methylation (5-azacytidine) and histone acetylation (trichostatin A). According to the results obtained via inter-specific hybridisation and the ability of the smut Ustilago violacea to activate development of male organs in female plants, it can be deduced that there is a locus suppressing male organ development in standard female plants. Function of this locus is not influenced by the drugs mentioned above.

Genomový imprinting je definován jako reverzibilní proces, kdy specifická modifikace genů v parentální generaci vede k funkčním rozdílům mezi paternálními a maternálními genomy v diploidních buňkách potomstva. Genomic imprinting represents an epigenetic marking of genes that results in their monoallelic expression, namely in mammals and flowering plants. Here, I also summarize our recent experiments describing the role of DNA methylation and histone H4 acetylation in plant reproductive development.

Sex chromosomes of plant species Silene latifolia are the ideal model for evolutionary biology studies because they represent early stages of X and Y chromosomes history. We used fluorescence in situ hybridization (FISH) with bacterial artificial chromosome (BAC) clones in studies of chromosome structure to demonstrate that a majority of genes are located in subtelomeric regions of all chromosomes. Our results also indicate the absence of accumulation of main classes of repetitive sequences - retroelements and centromeric tandem repeats - in non-recombining parts of the sex chromosomes.
