The fungal enzyme pyranose 2-oxidase (EC 1.1.3.10) is the key catalyst for production of 2-ketoglucose, a glucose derivative of industrial interest. To prevent formation of inclusion bodies during its heterologous expression in E. coli, several fermentation protocols were developed to ensure proper folding and recovery of the enzyme activity at high levels Enzym pyranosa-2-oxidasa z hub (EC 1.1.3.10) je klíčovým katalyzátorem pro produkci 2-keto-glukosy, průmyslově zajímavého derivátu glukosy. Aby bylo možno eliminovat tvorbu inklusních tělísek při jeho heterologní expresi v E. coli, byly vypracovány fermentační protokoly, které umožnily správné skládání proteinu a vedly k získání vysokých enzymových aktivit

This method increased the detection sensitivity more than ten times and allowed us to quantify doxorubicin cleaved from the conjugates and intercalated into nuclear DNA Jedním z hlavních mechanismů účinku anthracyklinového antibiotika doxorubicinu je jaderná akumulace a interkalace do jaderné DNA. Kvantifikace jeho akumulace v jádře nepřímou metodou pomocí snížení fluorescence Hoechst 33342 je výrazně citlivější a přesnější technikou než přímá detekce, založená na fluorescenci doxorubicinu

Using combination of bulk solution and single-molecule assays we have investigated the influence of several mutations in the PD-(D/E)-X-K catalytic motif of HsdR on the DNA translocation and ATPase properties of EcoR124I, the restriction-modification enzyme Type I. The gained results have given us the insight into the possible interdomain interactions between helicase and nuclease domains of the HsdR subunit and the effect of their interactions on DNA translocation Kombinací hromadných experimentů (stopped-flow) s experimenty s jednotlivou molekulou enzymu (magnetická pinzeta) byl studován vliv několik mutaci v katalytickém motivu PD-(D/E)-X-K podjednotky HsdR na DNA translokační a ATPasovou aktivitu restrikčně-modifikačního enzymu EcoR124I. Získané výsledky ukazují na možné interakce helikásové a nukleasové domény podjednotky HsdR a na efekt těchto interakcí na translokaci DNA

We demonstrated that the mitogen-activated protein kinases (MAPKs) play a crucial role in cancer cell death and survival, however their role seems to be indirect. We confirmed the previously published results showing that different intracellular processes are underway in cancer cells treated with Dox.HCl compared to those incubated with HPMA conjugates Prokázali jsme, že mitogenem aktivované signalizační dráhy (MAPKs) hrají důležitou roli při smrti a proliferaci buněk, ačkoli je jejich role nepřímá. Potvrdili jsme předchozí výsledky, které ukazují, intracelulární procesy buněk inkubovaných s volným doxorubicinem se liší od procesů buněk inkubovaných s HPMA konjugáty

Studies of structure and function of promoters in C. glutamicum were used in graded expression of genes involved in biosynthetic pathway of valine (ilvBNC, ilvD, ilvE) and in competing or branching-off pathways (ilvA, leuA). Combination of particular promoter mutants resulted in tuning the whole pathway to overall higher flux of the intermediates through the pathway and in high production of valine Byla studována struktura a funkce promotorů v C. glutamicum a výsledky využity k regulaci exprese genů biosyntézy valinu (ilvBNC, ilvD, ilvE) a genů konkurenčních nebo odvětvujících se biosyntetických drah (ilvA, leuA). Kombinace mutací v určitých promotorech vedly k modulaci celé biosyntetické dráhy s následným zvýšením toku meziproduktů a zvýšenou produkcí valinu
