Výzkum stability kalibračních faktorů při GC-FID analýze kvality alkoholických nápojů metodami vnitřního standardu

This thesis focuses on measuring calibration factors and the dependence of their stability on experimental conditions for the purpose of quality control and the safety of alcoholic beverages using gas chromatography with flame ionization detection (GC-FID). Ten volatile compounds (acetaldehyde, methanol, methyl acetate, ethyl acetate, 1-propanol, 1-butanol, 2-butanol, isobutanol, acetal, 3-methyl-1-butanol) were monitored according to the legal regulations of the European Commission. These target compounds were analysed in prepared solutions under 27 different measurement conditions. Taguchi's multifactorial statistical method was used to determine the influence of individual measurement parameters on the stability of calibration factors. The chosen variables for measurements were matrix composition, concentration of volatile compounds, GC injector temperature, sample injection volume, injection split ratio, and detector temperature. Within the experiments, each of these variable factors took one of three possible values. Relative response factors (RRF) for each analysed compound were compared using two internal standard (IS) methods. Předkládaná diplomová práce se věnuje měření kalibračních faktorů a závislosti jejich stability na experimentálních podmínkách pro účely kontroly kvality a bezpečnosti alkoholických nápojů metodou plynové chromatografie s plamenově ionizační detekcí (GC-FID). Bylo sledováno deset těkavých látek (acetaldehyd, methanol, methyl-acetát, ethyl-acetát, 1-propanol, 1-butanol, 2-butanol, isobutanol, acetal, 3-methyl-1-butanol) podle právní legislativy Evropské Komise. Tyto cílové látky byly v připravených roztocích analyzovány při 27 rozdílných podmínkách systému. Ke zjištění vlivu jednotlivých parametrů měření na stabilitu kalibračních faktorů byla použita Taguchiho multifaktoriální statistická metoda. Proměnnými měření byly zvoleny složení matrice, koncentrace těkavých látek, teplota GC injektoru, objem nástřiku vzorku, rozdělovací poměr nástřiku a teplota detektoru. Každý z těchto proměnných faktorů nabýval v rámci experimentů jednu ze tří možných hodnot. Relativní faktory odezvy (RRF) pro každou analyzovanou látku byly porovnány u dvou metod vnitřního standardu (IS). Adaptace dvouletých dětí v dětských skupinách

In my master thesis, I deal with the specific development of two-year-old children. Specifically, physical, cognitive, social and emotional development. The second to third chapters focus on the history of the inclusion of two-year-olds in preschool, experts' opinions on whether it is appropriate for children to enter preschool at the age of two, and what options exist in the country for the inclusion of two-year-olds. The fourth chapter looks at the options available to parents in the Czech Republic when they want to place their two-year-old child in pre-school. The last - fifth chapter deals with adaptation as such. The practical part of the master thesis begins by setting the objectives, which are to analyse the adaptation plan of a particular children's group, to find out how the adaptation period is going for the 11 children from the particular children's group who were involved in the research. Last but not least, the research aims to figure out what could make the adaptation to the children's group easier for two-year- old children and their parents. In order to accomplish my goals, I drew on children's group attendance records, questionnaires from the children's parents, and most importantly, my participant observation. I also address general quality standards as well as the quality... Ve své diplomové práci se zabývám specifickým vývojem dvouletých dětí. Konkrétně jde o fyzický, kognitivní, sociální a emocionální vývoj. Druhá až třetí kapitola je zaměřena na historii zařazování dvouletých dětí do předškolních zařízení, názory odborníků na to, zda je pro děti vhodný nástup do předškolního zařízení ve věku dvou let, a jaké existují v ČR možnosti zařazení dvouletých dětí do dětského kolektivu. Čtvrtá kapitola se věnuje možnostem, které mají rodiče v ČR, když chtějí umístit své dvouleté dítě do předškolního zařízení. Poslední - pátá kapitola pojednává o adaptaci jako takové. Praktická část diplomové práce začíná stanovením cílů, jimiž jsou analýza adaptačního plánu konkrétní dětské skupiny, zjištění, jak probíhá adaptační období u 11 dětí z konkrétní dětské skupiny, které byly zapojeny do výzkumu. V neposlední řadě je cílem výzkumu přijít na to, co by dvouletým dětem i jejich rodičům mohlo adaptaci do dětské skupiny zjednodušit. Na cestě k naplnění cílů jsem čerpala z docházkových záznamů dětské skupiny, dotazníků od rodičů dětí, a především ze svého zúčastněného pozorování. This master's thesis is focused on development of a new analytical method based on photochemical generation of volatile species (PCVG) of rhodium as an efficient sample introduction technique for inductively coupled plasma mass spectrometry. At first, the conditions affecting the PCVG efficiency were optimized, namely carrier gas flow rate, irradiation time, composition of the reaction medium as well as the impact of transition metal ions as potential sensitizers was examined. The combination of Cu2+ and Co2+ was found optimal, enhancing the PCVG efficiency more than 60-fold. Additionally, a serious effect of the residence time of volatile species in the reaction medium, after exiting the irradiated part of the photoreactor, was found. The PCVG efficiency was doubled by modifying the output part of the photoreactor and another two-fold increase in the PCVG efficiency was achieved using the addition of nitrate anions to the reaction medium. Under chosen optimal conditions, the limit of detection and quantification were 13 pg dm-3 (3σ, n = 11) and 42 pg dm-3 (10σ, n = 11), respectively, and the PCVG efficiency was determined to be 13.5 ± 0.1%. Tato diplomová práce se soustředí na vývoj nové analytické metody založené na fotochemickém generování těkavých specií rhodia jako techniky účinného vnášení vzorku pro hmotnostní spektrometrii s indukčně vázaným plazmatem. Nejprve byla provedena optimalizace parametrů, které mají vliv na účinnost fotochemického generování, tzn. průtoková rychlost nosného plynu (Ar), koncentrace kyseliny mravenčí v reakčním médiu, doba ozařování a v neposlední řadě vliv přítomnosti reakčních modifikátorů, především iontů přechodných kovů. Jako optimální byla vybrána kombinace Cu2+ a Co2+ iontů přidávaných do standardu/vzorku, která zvýšila účinnost generování více než 60krát. V průběhu práce byl také zjištěn závažný vliv doby setrvání těkavé specie v reakčním médiu po výstupu z ozařované části fotoreaktoru. Modifikací části aparatury pak bylo dosaženo přibližně dvojnásobného zvýšení účinnosti generování. Obdobné zvýšení poskytoval i přídavek dusičnanových aniontů do reakčního média. Za zvolených optimálních podmínek byla stanovena mez detekce (3σ, n = 11) na 13 pg dm-3, mez stanovitelnosti (10σ, n = 11) na 42 pg dm-3 a účinnost generování na 13,5 ± 0,1 %. This thesis presents options how to acquaint preschool-aged children with etiquette by the work with a book. The theoretical part of this thesis defines the basic terms which are related to etiquette and are crucial to the topic of thesis. It deals with socialization, social and moral development of preschool-aged children. This part gains an insight into etiquette. It sums up the findings about children's literature and takes a deeper view on stories with educational content. There is also a closer view on information how to work with a book in nursery school by the methods of Reading and Writing for Critical Thinking and Three Phase Model of Learning. There were determined five partial goals in the practical part. First of all it was being found out if and in what way the nursery teachers work with children with a topic of the formal rules of correct or polite behaviour in society. To achieve this goal was made the questionnaire for nursery teachers. On the basis of the outcome of this questionnaire were evaluated five most common difficulties of children's etiquette in the nursery (relationships, observing the rules, table manners, manipulation with things and relationship with authorities). Diplomová práce předkládá možnosti, jak prostřednictvím práce s knihou seznamovat děti předškolního věku se společenským chováním. Teoretická část práce definuje základní pojmy, které jsou spojovány s etiketou a jsou klíčové pro téma práce. Dále se zabývá socializací, sociálním vývojem a morálním vývojem dítěte předškolního věku. Přibližuje také pojem společenské chování. Dále shrnuje poznatky o literatuře pro děti, více do hloubky se věnuje příběhům s výchovným obsahem. Jsou zde také přiblíženy informace, jak pracovat s knihou v mateřské škole prostřednictvím metod programu Čtením a psaním ke kritickému myšlení a třífázového modelu učení. V praktické části bylo stanoveno pět dílčích cílů. Nejprve bylo zjišťováno, zda a jakým způsobem pedagogové mateřských škol pracují u dětí s tématem slušného chování. Pro dosažení tohoto cíle byl sestaven a distribuován dotazník pro pedagogy mateřských škol, na jehož základě bylo diagnostikováno pět nejčastějších problémů v oblasti slušného chování u dětí v předškolním věku (konkrétně jde o oblasti vztahy mezi dětmi, dodržování pravidel, stolování, zacházení s věcmi a vztah k autoritě). Title: The Affinity of Thoracic Outlet Syndrome and Chronic Finger Pain in Climbers Objectives: To determine possible affinity between thoracic outlet syndrome (TOS) and chronic finger pain in climbers by comparing the occurrence of TOS among climbers with chronic finger pain, climbers without chronic finger pain, and non-climbers. Methods: Sixty-six participants of both genders aged 18-40 were included in the study. Based on the initial questionnaire, they were divided into three groups: climbers with chronic finger pain, climbers without chronic finger pain, and non-climbers. The homogeneity of the groups based on quantitative characteristics was assessed using an ANOVA test. All study participants were examined using the same examination protocol for the presence of Thoracic Outlet Syndrome (TOS). The examination protocol consisted of three questions about clinical symptoms, five examination tests, and palpation of the scalene and pectoralis minor muscles. A positive finding of TOS was defined as a positive result of 60 % or more of the examination protocol tests. The frequency of positive TOS findings was subsequently compared between the group of climbers with chronic finger pain, climbers without chronic finger pain, and non- climbers. Název: Afinita thoracic outlet syndromu a chronické bolesti prstů u lezců Cíle: Ověření možné afinity thoracic outlet syndromu (TOS) a chronické bolesti prstů u lezců na základě porovnání četnosti výskytu thoracic outlet syndromu u skupiny lezců s chronickými bolestmi prstu, lezů bez chronické bolesti prstu a u nelezců. Metody: Do studie bylo zařazeno 66 probandů obou pohlaví ve věku 18-40 let. Dle vstupního dotazníku byli rozděleni do tří skupin: skupiny lezců s chronickou bolestí prstu, skupiny lezců bez chronické bolesti prstu a skupiny nelezců. Homogenita skupin na základě kvantitativních znaků byla posouzena pomocí ANOVA testu. Všichni účastníci studie byli vyšetřeni dle stejného vyšetřovacího protokolu na přítomnost TOS. Vyšetřovací protokol se skládal ze tří otázek na klinické příznaky, pěti vyšetřovacích testů a palpace mm. scaleni a mm. pectorales minor. Za pozitivní nález TOS byl stanoven pozitivní výsledek 60 % a více testů vyšetřovacího protokolu. This diploma thesis discusses the influence of the social climate of the teaching staff on the work performance and motivation of teachers in kindergartens. The theoretical part of the work is dedicated to the definition of teaching staff, the work duties they must perform and also their motivation. The role of kindergarten principals and their influence on the climate of the teaching staff and the motivation of pedagogues is also described. The theoretical part of the thesis explains the differences between the school climate, the classroom climate and the social climate of the teaching staff. The empirical part of the work presents the research results. The study focused on investigating the social climate of teaching staff in kindergartens and its influence on teacher's motivation and satisfaction. In this approach a combination of quantitative and qualitative methods was used to identify factors influencing the climate of the teaching staff and their influence on the work and motivation of teachers. Tato diplomová práce pojednává o vlivu sociálního klimatu pedagogického sboru na pracovní výkon a motivaci učitelů v mateřských školách. Teoretická část práce se věnuje definici pedagogických pracovníků, pracovními povinnostmi, které musejí zastávat a také jejich motivaci. Popsána je také role role ředitelů mateřských škol a jejich vliv na klima pedagogického sboru a motivaci pedagogů. Teoretická část práce vysvětluje rozdíly mezi školním klimatem, klimatem školní třídy a sociálním klimatem pedagogického sboru. Empirická část práce prezentuje výsledky výzkumu. Studie se zamĕřila na zkoumání sociálního klimatu pedagogických pracovníků v mateřských školách a jeho vlivu na motivaci a spokojenost učitelů. The present thesis deals with the influence of body composition on physical fitness. The thesis deals with the issue of increasing prevalence of obesity in children and adolescents. The aim of the thesis is to assess physical fitness in older school-aged children using a validated test battery and to determine the association of physical fitness in these children with their body composition. A total of 52 pupils aged 12-14 years were tested, of whom 34 were girls and 18 were boys. We used a modified Assessing levels of physical activity (ALPHA) test battery to determine the physical fitness of the children and adolescents. We also used a questionnaire survey to determine anthropometric parameters relevant to body composition. For the actual measurement, we used three skinfold measurements, dynamometry, Unifittest (6-60). F-test was used to calculate statistical significance of differences and Cohen's d was calculated to calculate substantive significance. The results showed that girls had higher amount of subcutaneous fat than boys. There was no difference in body weight and body height between girls and boys. PYedkládaná diplomová práce se zabývá vlivem tlesného slo ení na tlesnou zdatnost. Práce se zabývá problematikou zvyaující se prevalence obezity u dtí a dospívajících. Cílem práce je zhodnotit tlesnou zdatnost u dtí staraího akolního vku pomocí validizované testové baterie a zjistit souvislosti tlesné zdatnosti tchto dtí s jejich tlesným slo ením. Celkem bylo testováno 52 áko ve vku 12-14 let, z toho bylo 34 dívek a 18 chlapco. Pro zjiatní tlesné zdatnosti dtí a dospívajících jsme vyu ili modifikovanou testovou baterii Assessing levels of physical activity (ALPHA). 1 Abstract The continuous advance in industry, general intensification of processes and urbanisation inevitably leads to production of new substances each year, collectively referred to as emerging pollutants (EPs). As the mass production of EPs increases, the demand for wastewater treatment plants also dramatically increases. The inadequacy of conventional tretament plants in removing EPs, such as pharmaceuticals, pesticides, personal care products, and others, results in the presence of these substances in surface and groundwater at significant concentrations. The often negative or unclear impact of EPs to the environment furhter emphasizes the urgency of this issue and requires a corresponding response. Photocatalysis belongs to the co-called advanced oxidation processes, key techniques for ensuring effective wastewater treatment. This thesis is focused on the photocatalytic degradation of tetracycline, trimethoprim and sulfamethoxazole, three common antibiotics frequently found in wastewater. Graphitic carbon nitride, a promising material in research of fotocatalytic degradation, was used as a photocatalyst and was activated by visible light emitted from a mercury xenon lamp. Rhodamine B was... 1 Abstrakt Neustálý pokrok v průmyslu, obecné zintenzivňování procesů a urbanizace nevyhnutelně vede každým rokem k produkci nových látek, souhrnně označovaných jako tzv. emerging pollutants (EPs). S narůstající masovou produkcí EPs se rovněž dramaticky zvyšují nároky na čisticí technologie v čistírnách odpadních vod. Nedostatečnost běžných čistíren v odstraňování EPs, jako jsou farmaceutika, pesticidy či produkty osobní péče, má za následek přítomnost těchto látek v povrchových a podzemních vodách v nezanedbatelných koncentracích. Společně s negativním či nejasným dopadem EPs na životní prostředí je tak umocněna naléhavost tohoto problému a vyžaduje odpovídající reakci. Fotokatalýza patří mezi tzv. pokročilé oxidační procesy, klíčové techniky pro zajištění účinného čištění odpadních vod. Tato práce se věnuje studiu fotokatalytické degradace tetracyklinu, trimethoprimu a sulfamethoxazolu, tří běžných antibiotik často se vyskytujících v odpadních vodách. The aim of the master's thesis is a production and a realization of an educational programme in a cooperation with the National theatre for students at high schools. The theoretical part of the thesis is connected on published books of methodology of education of culture heritage and alternative ways of an interpretation. Thanks to these, the thesis is looking for a possibility of a public space for an education. In the second part is the thesis focused on a historical progression of the Czech professional theatre in Prague during years 1743-1892. Based this knowledge is realizing the educational programme in a public space of the Old Town and the New Town in the vicinity of the historical building of The National Theatre. The educational programme is using methodology descripted in the theoretical part which means a geocaching, an audio guide and a drama education. The practical part of the thesis is finished by reflection of the programme. KEYWORDS National theatre, escape game, audio guide, drama education, public space Cílem diplomové práce je tvorba a následná realizace edukačního programu pro střední školy ve spolupráci s Národním divadlem. Teoretická část diplomové práce navazuje na již vydané metodiky edukace kulturního dědictví a možnosti netradiční interpretace, pomocí kterých nalézá možnosti využití veřejného prostoru pro edukační činnost namísto uzavřeného prostoru kulturních institucí. Dále se teoretická část práce zabývá historickým vývojem české profesionální divadelní scény v Praze mezi léty 1743-1892. Na základě těchto poznatků je realizován edukační program ve veřejném prostoru Starého a Nového města pražského v okolí historické budovy Národního divadla. Edukační program využívá metody popsané v teoretické části práce, tedy geocaching, audioguide a dramatickou hru. This diploma thesis deals with summarization of information about Emilia Fryšová, a woman whose most important life roles included being an educator, ethnographer, and publicist. The first step in my processing for this work has been to research of given topic, then I have categorized discovered information based on its contents, I compared the information, and lastly analysed it. During the writing process, I have been verifying new leads and discovering today's long-forgotten facts. As I have already mentioned, besides teaching Fryšová had a close relation to ethnography, which primarily included cognizance of the South Bohemian Blata area, which she came to like, when she started to live and work in Soběslav. For a better context of this topic, I have decided to dedicate the first two chapters to themes that are closely connected to Fryšová, such as the life of teachers and the beginnings of ethnography in Bohemia. The main chapters of my thesis focus on detailed information about Fryšová. The third chapter is oriented towards her life as a whole. In the fourth chapter, I more deeply examine her association work and the activities as well as places and people linked to it. The fifth chapter contains the description of the (mainly) ethnographic collection, of which she was a proud owner, and it... Tato diplomová práce se zabývá shrnutím informací o Emilii Fryšové, ženě, mezi jejíž nejdůležitější životní role bych zařadila roli učitelky, etnografky a publicistky. Pro zpracování práce jsem nejprve provedla rešerši k danému tématu, následně jsem zjištěné informace roztřídila podle jejich obsahu, poté jsem je mezi sebou komparovala, a nakonec jsem je analyzovala. Během psaní jsem prověřovala případné nové stopy a odhalovala jsem tak dnes už dávno zapomenuté skutečnosti. Jak je již výše uvedeno, kromě učitelství byla Fryšové blízká hlavně etnografie, v jejímž rámci se zaměřovala především na poznání oblasti jihočeských Blat, které si oblíbila, když začala žít a pracovat v Soběslavi. Pro pochopení kontextu jsem se v prvních dvou kapitolách nejprve rozhodla přiblížit témata, která s Fryšovou v mnohém souvisí, a to život učitelek a počátky etnografie v českých zemích. Hlavní kapitoly práce jsou zaměřené na detailní poznání Fryšové. V třetí kapitole se věnuji jejímu životu jako celku. Ve čtvrté kapitole hlouběji rozebírám její spolkovou činnost, konkrétní aktivity s tím spojené, místa a osobnosti s tím související. V páté kapitole předkládám popis (především) etnografické sbírky, jejíž hrdou majitelkou byla, a s tím spojuji její účast na dobových výstavách a její zapojení do sbírkotvorné činnosti...
