The first part of the thesis "Petr Zdražil - The Violin Maker from the Lesser Town of Prague" tries to give a comprehensive overview of the development of violins and the history of the violin-making. It introduces the reader to important violinmaking personalities, the tradition of Prague violinmaking, and contemporary Prague violinmakers. The second part of the work is dedicated to the life and violin-making of the Czech violin maker Petr Zdražil, whose instruments are sought after by renowned Czech and foreign violin makers. The thesis describes the specific methods and procedures of this violin maker's instruments. The work intends to expand awareness about current violin making in the Czech Republic and to introduce the reader to his outstanding personality. Information for the thesis was gathered using the book resources and using the oral history method. První část práce "Petr Zdražil - houslař malostranský" se snaží podat ucelený přehled o vývoji houslí a historii houslařského řemesla. Představuje čtenáři významné osobnosti houslařské historie, seznamuje s tradicí pražského houslařství a současnými pražskými houslaři. Druhá část práce se zaměřuje na život a houslařské umění českého houslaře Petra Zdražila, jehož nástroje jsou vyhledávány předními českými i zahraničními houslisty. V práci jsou popsány specifické metody a postupy výroby houslí tohoto houslaře. Práce si klade za cíl rozšířit povědomí o současném houslařství u nás a představit jednu z jeho výrazných osobností - houslaře Petra Zdražila. Pro získání informací byla využita zejména metoda orální historie.

Natural Killer (NK) cells can spontaneously destroy target cells, such as infected, damaged, or malignant cells. NK cell cytotoxicity is mediated by activating receptors on their surface, such as NKp80 (Natural Killer protein 80 kDa). The human receptor NKp80 stimulates cytotoxicity through its interaction with its ligand AICL (Activation Induced C-type Lectin), which is constitutively expressed by all myeloid cells. In pathological conditions, such as cancerous or damaged cells, AICL is often upregulated, resulting in the lysis of these cells by NK cells expressing NKp80. This interaction is thus a promising immunotherapeutic target for treating myeloid leukaemia. However, the structures of both proteins have remained elusive. Therefore, we have focused on the successful production of the extracellular domain of these proteins. To improve the production yield of NKp80, we introduced a series of mutations in the stalk region to study their effect on production, stability, and homodimer formation. The introduced mutations significantly increased the production yield allowing for a large-scale production with subsequent crystallisation of the protein. The crystallisation resulted in the elucidation of the hitherto unknown structure of NKp80 homodimer at a resolution of 2.9 Å. However, the large-scale... NK buňky (z angl. natural killer cells, přirozeně zabíječské buňky) buňky hrají důležitou roli při rozpoznání a eliminaci infikovaných, poškozených či nádorových buněk bez nutnosti předchozího kontaktu s nimi. Jejich cytotoxicita je zprostředkována aktivačními receptory na jejich povrchu, jako je receptor NKp80 (z angl. Natural Killer protein 80 kDa). NKp80 se váže na ligand AICL, který je konstitutivně exprimován na myeloidních buňkách. Zvýšená exprese AICL, např. na nádorových buňkách, vede k aktivaci NK buněk exprimujících receptor NKp80. Interakce NKp80:AICL je proto slibným imunoterapeutickým cílem pro léčbu myeloidní leukémie. Struktura obou proteinů dosud nebyla známá, a proto jsme se v práci zaměřili na úspěšnou produkci extracelulární domény těchto proteinů za tímto účelem. Při produkci receptoru NKp80, jsme zavedli do "stalk" oblasti sérii mutací, kde byly jednotlivé cysteiny nahrazeny seriny. Pomocí zavedených mutací jsme studovali jejich vliv na produkci, stabilitu a tvorbu homodimeru. Zavedené mutace významně zvýšily výtěžek produkce, a proto byl protein připraven ve větším množství za účelem krystalizace proteinu. The thesis "Gender in Schools with a Focus on Civic Education" deals with the issue of gender in the school environment. The theoretical part of the thesis focuses on gender identity, gender stereotypes, gender equality, and the reproduction of gender roles in schools. Special attention is also paid to the personality of teachers and their role in gender reproduction. The significance of their attitudes is analyzed in the chapter on subjective theory. The main research questions I addressed were: What is the attitude of a specific civic education teacher towards the gender topic? How does the attitude of a civic education teacher towards gender issues manifest in civic education teaching and what impact does it have on students? Therefore, the empirical part of the thesis was focused on a case study of a beginning civic education teacher, which I explored through a research probe. To gain a deeper understanding of the research problem, I employed a combination of three methods, specifically semi-structured interviews, participant observation, and focus groups. Teoretická část práce se zaměřuje na genderovou identitu, genderové stereotypy, genderovou rovnost a reprodukci genderových rolí ve školách. Zvláštní pozornost je také věnována osobnosti učitelů a učitelek a jejich roli v reprodukci genderu. Význam jejich postojů je analyzován v kapitole subjektivní teorie. Hlavní výzkumné otázky, které jsem si při psaní kladla byly: Jaký je postoj konkrétního učitele občanské výchovy k tématu genderu? Jak se postoj učitele občanské výchovy k genderovým tématům projevuje ve výuce občanské výchovy a jaký to má vliv na žáky a žákyně? Empirická část diplomové práce byla proto zaměřená na případovou studii začínajícího učitele občanské výchovy, kterou jsem zjišťovala prostřednictvím výzkumné sondy. Pro hlubší porozumění výzkumného problému jsem využila kombinaci tří metod, a to konkrétně polostrukturovaný rozhovor, zúčastněné pozorování a focus group. Transmembrane channels and transporters of the ClC protein family are present across all living organisms. They are found on the cytoplasmic and lysosomal membranes of the cells, where they participate in maintaining ion homeostasis. When dysfuncional, they lead to serious health complications. To develop treatment for these diseases, it is essential to describe transport mechanism of ClC proteins. The antiporter ClC-ec1 from E.coli is used as a model protein for the entire ClC protein family. This homodimeric protein, which transports one proton against two chloride ions, has a separate transport path in each monomer. Based on the crystal structure, it is believed that during transport the protein alternates between outward and inward-facing conformations. Conversion to the outward-facing conformation of the protein is accompanied by the protonation of three glutamates located in the transport path. To study these conditions, a QQQ mutant was designed that has these glutamates replaced by glutamines. Until now, the study of the transport mechanism of ClC-ec1 has mainly relied on studies based on X-ray crystallography. Crystallography provided static images, which did not contain sufficient information about protein dynamics. Therefore, to study transport mechanism of ClC-ec1, we chose a dynamic... Transmembránové kanály a přenášeče z ClC proteinové rodiny se vyskytují napříč všemi živými organismy. Nacházejí se na cytoplasmatických a lysosomálních membránách buněk, kde se podílí na udržování homeostázy iontů. Narušení jejich funkce vede k vážným zdravotním komplikacím. Pro vývoj efektivní léčby těchto onemocnění je potřeba objasnit transportní mechanismus ClC proteinů. Antiporter ClC-ec1 z E.coli je modelovým proteinem celé ClC proteinové rodiny. Tento homodimerní protein, který transportuje proton proti dvěma chloridovým iontům, má transportní dráhu v každé z podjednotek. Na základě krystalové struktury se přepokládá, že během transportu se protein střídavě otevírá na obě strany membrány. Otevření proteinu směrem ven je umožněno protonací tří glutamátů, které se nacházejí v transportní dráze a jsou pro transport iontů klíčové. Pro studium tohoto stavu byl navržen QQQ mutant, který má tyto glutamáty nahrazeny za glutaminy. Dosud se studium transportního mechanismu ClC-ec1 opíralo převážně o studie založené na rentgenové krystalografii. Krystalografie poskytla statické obrázky, které neobsahovaly dostatečné informace o dynamice proteinu. Hemoproteins are diverse and very important group of proteins involved in a wide range of biochemical processes. Their typical roles in living organisms include oxygen storage and distribution, enzymatic catalysis of redox reactions and electron transport in the respiratory chain. In relatively recent times, a fourth group of hemoproteins, the so-called heme sensor proteins, has been discovered. This group includes, among others, proteins that have the ability to reversibly bind heme, which in such cases acts as a signaling molecule. This reversible interaction is essential for cells in certain signalling pathways. As it was found, one of the heme sensor proteins is the p53 protein, which is an important transcription factor The first phase of this diploma thesis was focused to the preparation of p53 family proteins including some of their deletion mutants. Subsequently, the task for selected proteins was to study the influence of heme on their oligomeric state. The essential part of the thesis consists of spectrophotometric studies, which aimed to reveal potential interactions of the studied proteins with heme and to characterize the interactions. Keywords: heme, heme sensor proteins, p53 protein family, transcription factors, signal transduction Hemoproteiny jsou rozmanitou a velmi významnou skupinou proteinů podílející se na celé řadě biochemických pochodů. Mezi jejich typické role v živých organismech patří uskladňování a distribuce kyslíku, enzymová katalýza redoxních reakcí a transport elektronů v dýchacím řetězci. V relativně nedávné době byla objevena čtvrtá skupina hemoproteinů, tzv. hemové senzorové proteiny. Do této skupiny náleží mimo jiné proteiny, které disponují schopností reversibilně vázat hem, jenž takových případech plní funkci signální molekuly. Tato reversibilní interakce má pro buňky zásadní význam v některých signalizačních drahách. Jak bylo zjištěno, jedním z hemových senzorových proteinů je také významný transkripční faktor, protein p53. První fáze předkládané diplomové práce byla věnována přípravě proteinů rodiny p53 včetně některých jejich delečních mutantů. Následně bylo úkolem u vybraných proteinů prostudovat vliv hemu na jejich oligomerní stav. Zásadní část diplomové práce je tvořena spektrofotometrickými studiemi, které si kladly za cíl odhalit potenciální interakce studovaných proteinů s hemem, popřípadě dané interakce charakterizovat. Aspartic proteases (APs) are crucial for diverse cellular processes. This thesis delves into the complexities of protein expression and characterization of vacuolar aspartic endoprotease Apr1p from Candida albicans, comparing it to its Saccharomyces cerevisiae ortholog, Pep4p. Recombinant expression of Apr1p in Escherichia coli yielded the inactive proenzyme, proApr1p. Extensive refolding efforts failed to produce mature, active Apr1p, suggesting a reliance on intricate cellular machinery or specific post-translational modifications for activation. Attempts to leverage vacuolar enzymes or cell lysates for proApr1p activation were unsuccessful, potentially due to the fragility of isolated vacuoles and the complex mixture of enzymes in cell lysates. Positive results emerged when Apr1p was expressed in S. cerevisiae, where fractionated cell lysates exhibited specific proteolytic activity at acidic pH after inhibiting serine and metalloproteases proteases. The eukaryotic system can probably produce active Apr1p. However, after preliminary small-scale experiments, upscaling of Apr1p expression in S. cerevisiae will be necessary in order to obtain sufficient amount of protein for further characterization. A reciprocal gene swap experiment, exchanging PEP4 in S. cerevisiae with APR1 and vice versa,... Aspartátové proteázy (AP) mají zásadní význam pro různé buněčné procesy. Tato diplomová práce se zabývá problematikou exprese a charakterizace vakuolární aspartátové endoproteasy Apr1p z Candida albicans a srovnává ji s jejím orthologem Pep4p ze Saccharomyces cerevisiae. Rekombinantní exprese Apr1p v Escherichia coli poskytla neaktivní proenzym, proApr1p. Rozsáhlé snahy o aktivaci nevedly k maturaci Apr1p, což naznačuje závislost aktivace na posttranslačních modifikacích, jako je glykosylace a specifické interakce s jinými proteázami podobně jako u Pep4p. Pokusy o využití vakuolárních enzymů nebo buněčných lyzátů k aktivaci proApr1p byly také neúspěšné, což může být způsobeno křehkostí izolovaných vakuol a komplexní směsí enzymů v buněčných lyzátech ztěžjucií analýzu aktivace. Tyto problémy podtrhují složitou povahu aktivace proteas kdy ani různe in vitro techniky nedokážou simulovat in vivo aktivaci proteas. Slibné výsledky se však objevily u heterologní exprese proApr1p v Saccharomyces cerevisiae, kde frakcionovaný buněčný lyzát vykazovaly specifickou proteolytickou aktivitu při kyselém pH po inhibici jiných proteáz. Title: Comparison of somatognosia and postural stability between female sports aerobics and gymnasts Objectives: The aim of this thesis is to evaluate asomatognosia, constitutional hypermobility and postural stability in the studied groups (i.e. the group of sport aerobics gymnasts and the group of gymnasts) and their subsequent comparison with each other. Another aim is to determine whether there is a correlation between the different modalities studied, i.e. asomatognosia, constitutional hypermobility, postural stability, ability to evaluate sensory stimuli and body image. Methods: 86 girls and women aged 8-24 years, 41 in the sport aerobics group and 45 in the gymnastics group participated in the study. They were evaluated by the Petrie test, Body attitude test, body size tests (shoulder width test, pelvic width test, foot length test), limbs statesthesia, Beighton score and Y-balance test. Data was collected in the gyms of the participating sports clubs. It was conducted from March 2023 to November 2023. Microsoft Excel 365 and R statistical software (version 4.3.2) were used for statistical data analysis. Selected statistical tests were calculated for 95% significance level. Název: Porovnání somatognozie a posturální stability mezi sportovními aerobičkami a gymnastkami Cíle: Cílem této diplomové práce je zhodnotit somatognozii, konstituční hypermobilitu a posturální stabilitu u zkoumaných skupin (tedy skupiny sportovních aerobiček a skupiny sportovních gymnastek) a jejich následné porovnání mezi sebou. Dalším cílem je zjistit, zda existuje korelace mezi jednotlivými zkoumanými modalitami, tedy somatognozií, konstituční hypermobilitou, posturální stabilitou, schopností vyhodnocovat senzorické podněty a tělesným sebepojetím. Metody: Výzkumu se zúčastnilo 86 dívek a žen ve věkovém rozmezí 8-24 let. Z toho 41 osob ve skupině sportovních aerobiček a 45 ve skupině sportovních gymnastek. Byl u nich hodnocen test dle Petrie, Body attitude test, testy tělesných rozměrů (test šíře ramen, šíře pánve, délky chodidla), polohocit končetin, Beighton skóre a Y-balance test. Sběr dat se uskutečnil v tělocvičnách zúčastněných sportovních klubů a proběhl od března roku 2023 do listopadu roku 2023. Master's thesis titled "The Theme of War Mediated to Children in Kindergarten through Story" addresses the issue of preschool children affected by the war situation in Ukraine. The theoretical part of the thesis is based on insights discussing the inclusion of a child with a different native language in preschool education, on children from Ukraine in Czech kindergartens, and on understanding the relationships between Ukraine and Russia. The final section of the theoretical framework focuses on working with books in kindergarten and processing the theme of war through storytelling in kindergarten. Three main objectives are set for the practical part, corresponding to the primary intention of the thesis, which is to verify how to work with selected children's literature within the inclusion of children from Ukraine affected by the war situation in Ukraine. The individual objectives address perspectives on the theme of war and how to work with it in kindergarten from the viewpoint of 80 preschool teachers and three selected experts. Based on the data obtained from the questionnaire survey and individual in-depth interviews, three lessons focused on introducing the theme of war to children in kindergarten through children's publications were developed. Teoretická část práce se opírá o poznatky pojednávající o inkluzi dítěte s odlišným mateřským jazykem v předškolním vzdělávání, o dětech z Ukrajiny v české mateřské škole a pochopení vztahů mezi Ukrajinou a Ruskem. V poslední části teoretických východisek práce je věnována pozornost práci s knihou v mateřské škole a zpracování tématu války prostřednictvím příběhu v mateřské škole. Greenwashing as Unfair Competition Practice Abstract: The thesis focuses on one of the major obstacles in the fight against climate change, greenwashing. Given the difficulty in defining and the diversity of this phenomenon, this thesis aims to introduce greenwashing and examine it through the lens of unfair competition law. In particular, it examines whether greenwashing can be an unfair competition act, i.e. whether it fulfils the general unfair competition clause, whether it can possibly also fulfil one of the special unfair competition acts and whether the current unfair competition legislation is sufficient or whether it would be appropriate to create a new special unfair competition act regulating greenwashing. The first chapter of the thesis focuses on competition, defines competition law and discusses the basic concepts related to competition. All of this is important in order to subsequently highlight how competition is changing in the context of environmental protection efforts and related activities and what threats it faces. Zabývá se zejména tím, zda může být greenwashing nekalosoutěžním jednáním, tedy naplňovat generální klauzuli nekalé soutěže, zda případně může naplňovat i některou ze zvláštních skutkových podstat nekalé soutěže a zda je aktuální nekalosoutěžní právní úprava dostačující nebo by bylo vhodné vytvoření nové zvláštní skutkové podstaty nekalé soutěže upravující greenwashing. První kapitola diplomové práce se zaměřuje na hospodářskou soutěž, vymezuje právo hospodářské soutěže a zabývá se základními pojmy s hospodářskou soutěží souvisejícími. To vše je důležité pro následné poukázání na to, jak se hospodářská soutěž v souvislosti se snahami o ochranu životního prostředí a souvisejícími aktivitami mění a jakým hrozbám čelí. Etika politického vlivu na světový sport

The thesis deals with the political influence of modern sport and its ethical aspects. The author of the thesis aims to criticize actions in world sport with the help of basic ethical ideas about correct decision-making. He follows the controversial question of whether politics belong to sport and whether it is possible to separate these industries. In the first part, he presents the basics of ethics and its goals, which are supposed to help determine the correctness of actions in the world of sports. Following on from ethics, he then points out the connection between ethics and sport, where he proves the correctness of practicing sport with the help of professional literature. In the second part, he describes the process of emergence of sport as such, as well as the manifestation of the influence of world sports events by politics on specific cases and their impact on sport overall and society. At the end, the author answers the ethical questions that arise from the issue of the topic. It is based on the collection of information from professional literature and publicly available periodicals, where he gets to the result that the separation of politics and sport is not possible. Práce se zabývá politickým ovlivňováním novodobého sportu a jeho etickými aspekty. Autor práce si klade za cíl podrobit kritice jednání ve světovém sportu za pomoci základních etických myšlenek o správném rozhodování. Sleduje tak kontroverzní otázku, zdali politika patří do sportu a jestli je možné tato odvětví oddělit. V první části představuje základy etiky a její cíle, které mají pomoci určit správnost činů ve světě sportu. V návaznosti na etiku pak poukazuje na propojení etiky a sportu, kdy za pomoci odborné literatury dokazuje správnost provozování sportu. Ve druhé části popisuje průběh vzniku sportu jako takového, dále projevování ovlivňování světových sportovních událostí politikou na konkrétních případech a jejich dopady na sport celkově a společnost. Na závěr autor představuje své odpovědi na etické otázky, které vycházejí z problematiky tématu. Vychází ze sběru informací z odborné literatury a veřejně dostupných periodik, pomocí nichž dochází k výsledku nemožného oddělení politiky a sportu.
