Flow cytogenetics is an attractive tool that facilitates genome mapping. Flow cytometric analysis permits classification of isolated chromosomes according to their DNA content As thousands of chromosomes are analysed in each sample, quantitative detection of structural and numerical chromosome changes is possible. Any chromosome that can be discriminated can also be sorted in large quantities and high purities for subsequent molecular analysis. Unfortunately, only the smallest chromosome 5(1H) can be discriminated and sorted in barley. This is due to similarity in relative DNA content among the chromosomes 1 - 4,6 and 7 (2H - 7H). Cytogenetic stocks facilitate sorting of those translocation chromosomes that differ in DNA content Even if the sorted translocation chromosomes were found useful for mapping DNA sequences to subchromosomal regions, their use remained limited. Here we describe a novel approach that facilitates sorting of any of the 14 barley chromosome arms. The strategy is based on using wheat-barley addition lines carrying barley chromosome arms (telocentric chromosomes). Furthermore, we will be able to sort subarm chromosomal segments that are generated in the wheat-barley chromosome addition lines by the gametocidal system. These advances make flow cytogenetics an attractive tool that can simplify barley genome mapping. A possibility to purify large quantities of individual chromosome arms and their segments opens avenues for targeted isolation of low-copy "genie" sequences, preparation of specific probes for screening EST arrays, HAPPY mapping, and construction of chromosome arm-specific BAG libraries. Analýza pomocí průtokového cytometru umožňuje klasifikaci izolovaných chromozómů podle obsahu DNA. Každý chromozóm, který může být pomocí průtokového cytometru identifikován, může být zároveň velkou rychlostí tříděn z populace všech chromozómů. U ječmene však může být tříděn pouze nejmenší chromozóm 1H. Nová strategie je založena na použití adičních linií pšenice-ječmen nesoucích v genomu pšenice chromozómová ramena ječmene ve formě telocentrických chromozómů. Tato strategie umožňuje třídění jakéhokoliv ze 14 chromozómových ramen ječmene.

Brassinolides are known to stimulate plant growth and to posses antistresiTactivities in plants. This work was aimed at exploring possible beneficial effects of 24-epibrassinolide on cultured mammalian cells. A mouse hybridoma, i.e. a hybrid ymphocyte cell line, was cultured either in standard serum-free medium, or in diluted medium in which the cell grew inder nutritional stress. Steady-state parameters of semicon-finuous cultures conducted at 24-epibrassinolide concentra-ions from 10~16 M to 1CT9 M were evaluated. Typical effects of he agent that were observed both in standard and in diluted i [media were suppression of intracellular antibody level, increa- / ,ed value of mitochondrial membrane potential, higher frac- / ion of cells in the G(/Gl phase and lower fraction of cells in / he S phase. Alleviation of nutritional stress was observed in j (cultures conducted in diluted media. Viable cell density was j ignificantly higher at 24-epibrassinolide concentrations 10~13[ knd 10~12 M. Results of this exploratory study show that thej plant hormone 24-epibrassinolide may induce alterations of he cell division mechanism of the energetic metabolism, and of secreted protein synthesis in a mammalian cell line.

Stolní skenery mohou být využity pro dokumentaci biologických, a zvláště botanických, plochých objektů v laboratoři stejně dobře, jako (digitální) fotodokumentace. Lépe než herbářové položky zachycují tvar a barevnost, oproti fotografické dokumentaci jsou většinou rychlejší a levnější. Drobné trojrozměrné předměty (plody, semena) lze dokumentovat také. Obvyklé rozlišení 600 dpi je již vyšší než rozlišení lidského oka a může nahradit použití lupy při pozorování objektu. Table scanners may be used for documentation of biological, and especially flat botanical, objects in laboratory. In comparison with (digital) photography is picture from scanner mostly cheaper and quicker. In comparison with classical botany herbaria picture better catch the shape and colour of fresh biological object. Small 3D objects (seeds, fruits) may be also scanned without problems. Resolution 600 dpi is higher than that of human eye. Scanned picture may be used instead of using small magnifying glass.
