Eye-tracking jako metoda pro sledování strategií žáků při práci s periodickou tabulkou prvků

This method allows capturing certain procedures of the respondents from their eye movements. We can subsequently derive certain specific strategies from the procedures, and subsequently adapt the teaching materials to themThe thesis focuses on the detection of different strategies of pupils when working with the periodic table with the help of an eye-tracking device. The work uses a pre-prepared test in which the students determined the specific location of specific elements in a table with displayed element symbols and proton numbers and subsequently the location of other specific elements in a blind periodic table. Subsequently, the strategy of pupils with higher self-confidence was examined and compared with the strategy of pupils with lower self-confidence. Success in finding individual elements in the test was also compared. A typical strategy, which was observed in most respondents, is to determine the location where the element is most likely to be located and then search in a sufficient area for signs that could belong to the searched element. Výukové metody se každým rokem snaží zdokonalovat a moderní technologie tomuto pokroku dopomáhají. Jednou z těchto technologie je i metoda eye-trackingu. Tato metoda dovoluje zachycovat určité postupy respondentů z pohybu jejich očí. Z postupů můžeme následně vyvozovat určité konkrétní strategie a následně na ně přizpůsobovat výukové materiály Práce se zaměřuje na zjišťování různých strategií žáků při práci s periodickou tabulkou s pomocí eye-trackingového zařízení. Práce využívá předpřipravený test, ve kterém žáci určovali konkrétní lokality specifických prvků v tabulce se zobrazenými značkami prvků a protonovými čísly a následně lokalizace jiných specifických prvků ve slepé periodické tabulce. Následně byla zkoumána strategie u žáků s vyšší sebedůvěrou a porovnána se strategií žáků s nižší sebedůvěrou. Byla porovnána i úspěšnost při hledání jednotlivých prvků v testu. Typickou strategii, která byla pozorována u většiny respondentů, spočívala v určení lokality, kde se prvek mohl s největší pravděpodobností nacházet a dále pak hledání v omezené oblasti značky, která by mohla patřit hledanému prvku. Tato skutečnost neplatila pouze v případě hledání prvku antimon, který respondenti mohli špatně určit kvůli zmatení vycházející z jeho českému názvu. 2024 - Czech The bachelor's thesis focuses on the teaching of laboratory methods at primary and secondary schools and its support with educational videos. A comparison of framework education programs and school education programs revealed a too general concept and little emphasis on laboratory teaching and the development of laboratory skills in primary and secondary education. A questionnaire survey among chemistry teachers helped us to define the basic laboratory methods taught. Information from the questionnaire survey was used in the creation of videos, specifically: work with kahan, filtration and crystallization. The created videos were subjected to feedback using a second questionnaire survey, on the basis of which the final editing of the videos took place. Bakalářská práce se zaměřuje na výuku laboratorních metod na ZŠ a SŠ a její podporu výukovými videi. Z porovnání rámcových vzdělávací programů a školních vzdělávacích programů vyplynulo příliš obecné pojetí a malý důraz na laboratorní výuku a rozvoj laboratorních dovedností v základním i středním školství. Dotazníkové šetření mezi vyučujícími chemie nám pomohlo definovat základní vyučované laboratorní metody. Informace z dotazníkového šetření, byly použity při tvorbě videí, konkrétně: práce s kahanem, filtrace a krystalizace. Vytvořená videa byla podrobena zpětné vazbě pomocí druhého dotazníkového šetření, na základě kterého došlo k finální úpravě videí. 2024 - Czech The topic of this bachelor thesis is Domestic violence against the elderly. I was led to work on this topic by my interest stemming from my experience with a case of domestic violence against an elderly woman from the time when I worked as a carer in a home care service center. This experience awakened in me the subject for this bachelor's thesis and the interest to deal more with this issue in the future, within the profession as a social worker. The thesis presents the issue of domestic violence against the elderly and research into the methods and quality of assistance to victims by providers of field and residential care service. The bachelor's thesis consists of two parts, theoretical and practical. In the theoretical part I define the terms senior and old age, domestic violence, perpetrator of domestic violence, consequences of domestic violence and possibilities for help and prevention. The practical part of this thesis includes qualitative research, focused on the method and quality of assistance to victims of domestic violence among the elderly provided by field and residential care service facilities. Exprese a vazebné vlastnosti lidského galektinu-4

2024 - Czech Galectin-4, a member of the galectin family known for its affinity to β-galactosides, belongs to tandem-repeat galectins and consists of two distinct subunits connected with a peptide linker. Galectins play a key role in various biological processes and are involved in the pathogenesis of many diseases. Colorectal cancer (CRC) tissue shows a lower expression of galectin-4 compared to healthy colon tissues, and the lower expression of galectin-4 promotes tumor progression and transport. Galectin-4 was found to inhibit tumorigenesis of CRC cells via Wnt/β-catenin and IL-6/NF-κB/STAT3 signaling pathways, which play a key role in CRC development. In inflammatory bowel diseases such as Crohn's disease and ulcerative colitis, galectin-4 interacts with CD4+ T lymphocytes, influencing their behavior in the immunological synapsis. Furthermore, galectin-4, primarily expressed in the gastrointestinal tract, participates in the apical transport in epithelial cells, promotes the healing of intestinal epithelial wounds, participates in the adaptive immune response and has multiple roles in the central nervous system. Studying inhibitors of galectin-4 helps to understand its specific roles in these processes, and thus to identify new potential therapeutic targets. Galektin-4, člen rodiny galektinů typický svou afinitou k β-galaktosidům, patří mezi tandemové galektiny a skládá se ze dvou odlišných podjednotek spojených peptidovým linkerem. Galektiny hrají klíčovou roli v různých biologických procesech a jsou zapojeny do patogeneze mnoha onemocnění. Tkáně s kolorektálním karcinomem (CRC) vykazují nižší expresi galektinu-4 ve srovnání se zdravými tkáněmi tlustého střeva a právě nižší exprese galektinu-4 podporuje progresi nádoru a metastáz. Bylo zjištěno, že galektin-4 inhibuje tumorigenezi buněk CRC prostřednictvím signálních drah Wnt/β-katenin a IL-6/NF- κB/STAT3, které hrají klíčovou roli ve vývoji CRC. V zánětlivých střevních onemocněních, jako je Crohnova nemoc a ulcerózní kolitida, interaguje galektin-4 s CD4+ T lymfocyty, což ovlivňuje jejich chování v imunologické synapsi. 2024 - German The subject of this bachelor's thesis is an annotated translation of selected chapters from the book Jóga mezi e-maily by Helena Nehasilová. In this book, the author deals with the topic of a healthy lifestyle in the office with the help of yoga practice. The thesis is divided into a practical and a theoretical part: the first part is a translation of the original text from Czech into German and the second part is a commentary on the translation. The commentary consists of a translation analysis of the source text based on Christiane Nord's method, translation methods, translation difficulties and possible solutions. Studium proteinu oligandrinu sekretovaného oomycetou Pythium oligandrum

2024 - Czech Oomycete Pythium oligandrum acts as a mycoparasite of pathogenic fungi, bacteria and oomycetes in soil due to the production of a number of hydrolytic enzymes for the degradation of polysaccharides and proteins in the cell wall of the host. At the same time, P. oligandrum can interact with plant roots through specific elicitors and stimulate plant defense and growth. Thus, P. oligandrum is a successfully used environmentally friendly biological control agent of plants. Among the secreted elicitors P. oligandrum to the low molecular weight proteins belong oligandrins (~10 kDa), which have a characteristic structure and a conserved sequence among the group of so-called elicitins, and at the same time are not homologous to plant proteins. In the framework of this bachelor thesis, was analyzed the production of oligandrin in various types of growing media after cultivation of P. oligandrum. The total proteolytic activity and the content of phenolic substances as other possible elicitors of plant defense reactions were also observed in culture media. Further, this work focused on the possibility of recombinant oligandrin expression in E. coli and its subsequent purification. Key words: Pythium oligandrum, oligandrin, elicitors, cultivation, recombinant expression [IN CZECH] Oomyceta Pythium oligandrum působí jako mykoparazit patogenních hub, bakterií i oomycet v půdě díky produkci řady hydrolytických enzymů pro degradaci polysacharidů i proteinů v buněčné stěně hostitele. Zároveň také P. oligandrum dokáže interagovat s kořeny rostlin prostřednictvím specifických elicitorů a stimulovat obranyschopnost a růst rostlin. P. oligandrum je tedy úspěšně používaným ekologicky šetrným prostředkem biologické ochrany rostlin. Mezi sekretované elicitory P. oligandrum patří nízkomolekulární proteiny oligandriny (~10 kDa), které mají charakteristickou strukturu a konzervovanou sekvenci mezi skupinou tzv. elicitinů, a současně nejsou homologní k rostlinným proteinům. V rámci této bakalářské práce byla analyzována produkce oligandrinu v různých typech pěstebních médií po kultivaci P. oligandrum. V kultivačních médiích byla také sledována celková proteolytická aktivita a obsah fenolických látek jako dalších možných elicitorů obranných reakcí rostlin. Dále se tato práce soustředila na možnost rekombinantní exprese oligandrinu v buňkách E. coli a jeho následnou purifikaci. Mitochondriální biomarkery u Huntingtonovy nemoci

2024 - Czech Huntington' s disease (HD) is a severe hereditary neurodegenerative disorder. The cause of this disease is a mutation of the gene that codes the protein huntingtin (htt). The mutation manifests itself as an abnormal extension of CAG triplets. In this way, a polyglutamine chain is formed in the htt molecule, which causes its fragmentation and aggregation. The result is a misfolded protein and this results in damage of neurons, especially in the area of striatum. The biochemical basis of neuronal death has not yet been clarified. Today, there is no effective treatment for HD. Htt participates in many different cellular processes, for example it participates in the regulation of transport events in the cell, participates in the transcription of some genes and also controls apoptosis. Mutated htt can be the cause of mitochondrial dysfunction. The consequence of the action of mutated htt is the occurrence of oxidative stress and subsequently the onset of the neurodegeneration process, but the exact state of mitochondria during the development of the HD has not yet been carefully investigated. Previous studies have studied mitochondrial disorders not only in the brain, but also in other extraneuronal tissues. The aim of the bachelor's thesis was to optimize the preparation of a sample from buccal smear... Huntingtonova nemoc (HN) je dědičné závažné neurodegenerativní onemocnění. Příčinou této choroby je mutace genu, který kóduje protein huntingtin (htt). Mutace se projevuje abnormálním prodloužením CAG tripletů. V molekule htt tak vzniká polyglutaminový řetězec, který způsobuje jeho fragmentaci a agregaci. Výsledkem je nesprávně sbalený protein a to má za následek poškození neuronů, především v oblasti striata. Biochemická podstata odumírání neuronů není dosud objasněna. Dodnes neexistuje žádná efektivní léčba HN. Htt se podílí se na mnoha různých buněčných procesech, např. se účastní regulace transportních dějů v buňce, podílí se na transkripci některých genů a kontroluje také apoptózu. Mutovaný htt může být příčinou poruch mitochondrií. Důsledkem působení mutovaného htt je vznik oxidačního stresu a následně nástup procesu neurodegenerace, ale přesná úloha mitochondrií během rozvoje nemoci nebyla dosud pečlivě prozkoumána Předchozí studie prokázaly poruchy mitochondrií nejen v mozku, ale i v dalších extraneuronálních tkáních. 2024 - Czech This bachelor thesis deals with the internal and external variation in speaker's assessments based on speech performance. Using the material of two poems ("Portrét" by Karel Toman and "Píseň zhýralého jinocha" by František Gellner), the variability in the evaluation of the recordings of 32 speakers (16 women and 16 men) was studied. Their performance was rated by 30 listeners on a seven-point scale based on their impression of the overall recitation. The general nature of the perception test instructions allowed us to observe which factors may play an important role in the perception of the overall speech performance. The results point, among other things, to the possible influence of the text of the poems on respondents' assessments. This study serves as a basis for further in-depth research in the area of speech assessment. Na materiálu dvou básní (Portrét Karla Tomana a Píseň zhýralého jinocha Františka Gellnera) byla sledována variabilita v hodnocení nahrávek 32 mluvčích (16 žen a 16 mužů). Jejich výstup hodnotilo 30 posluchačů na sedmibodové škále na základě svého dojmu z celkové recitace. Obecný charakter motivačního textu nám umožnil sledovat, jaké faktory mohou hrát důležitou roli při vnímání celkového mluvního projevu. Výsledky mimo jiné poukazují na možný vliv textu básní na hodnocení respondentů. Tato studie slouží jako podklad pro další hlubší výzkum v oblasti hodnocení mluvního projevu. Spisovatelka a její kritici. České reflexe tweetů J. K. Rowlingové o trans lidech

2024 - Czech This bachelor thesis focuses on the presentation of the conflict between the writer J.K. Rowling and her critics on five of the most widely read Czech online news portals, namely Novinky.cz, Lidovky.cz, Aktualne.cz, Echo24.cz, and iDNES.cz. The thesis examines the textual strategies used to communicate about trans individuals and the controversy surrounding them. The thesis employs methods derived from ethnomethodology, specifically Membership Categorization Analysis (MCA) and Media Dialogical Network (MDN). Using the MCA method, it was found that by categorizing the writer and her critics into collections of membership categories, a certain image of the situation was conveyed. The controversy was portrayed as interpersonal drama among celebrities, damage to J.K. Rowling's career, cancel culture, injecting politics into the apolitical sphere of art and culture, but only rarely as the struggle of trans people for their rights. In the analysis of texts using the MDN method, it was found that the portals, in developing the dialogic network, focus primarily on how the specific case harmed the life and career of Rowling and other artists. In the news, trans people were thus pushed into the background, with their lives and rights being only minimally addressed. Trans people in the texts become mere... Tato bakalářská práce se věnuje prezentaci konfliktu mezi spisovatelkou J. K. Rowlingovou a jejími kritiky na pěti z nejčtenějších českých internetových zpravodajských serverů, tj. na severech Novinky.cz, Lidovky.cz, Aktualne.cz, Echo24.cz a iDNES.cz. Práce se zabývá tím, jaké textové strategie jsou používány ke komunikaci o trans lidech a kontroverzi, která se jich týká. Používá metody vycházející z entometodologie, konkrétně členskou kategorizační analýzu (ČKA) a dialogickou mediální síť (DMS). Pomocí metody ČKA bylo zjištěno, že díky řazení spisovatelky a její kritiků do souborů členských kategorií byl předáván určitý obraz situace. Kontroverze byla zobrazována jako mezilidské drama mezi celebritami, újma na kariéře spisovatelky J. K. Rowlingové, cancelling, vnášení politiky do apolitické sféry umění a kultury, ale pouze výjimečně jako boj trans lidí za jejich práva. Při analýze textů metodou DMS bylo zjištěno, že servery se při rozvíjení dialogické sítě soustředí především na to, jak daná kauza ublížila životu a kariéře Rowlingové a jiným umělcům. Ve zprávách tak byli odsunuti do pozadí samotní trans lidé, jejichž životy a práva jsou tematizována pouze minimálně.
