Affinity chromatography in proteomics

2012 - English Afinitní chromatografie je jednou z variant chromatografie, která k separacím využívá unikátních vlastností interakce analytu a ligandu. Je totiž založena na výjimečné schopnosti biologických látek vázat specificky a reversibilně komplementární molekuly. Od doby prvních studií protein-proteinových interakcí ve druhé polovině devadesátých let minulého století zažívají afinitní separace v proteomice opravdovou renesanci. Vyvinuta byla celá řada nových afinitních matricí a experimentálních postupů založených na principech afinitní chromatografie, které posléze našly široké uplatnění ? od odstraňování nadměrně zastoupených proteinů z biologických směsí, přes profilování molekulárních cílů léčiv, až po podrobné studium posttranslačních modifikací. Tato disertační práce se věnuje roli afinitní chromatografie v moderní proteomice. Z tohoto důvodu se literární přehled zaměřuje především na použití této separační techniky v rámci proteomických experimentů. Jedinou výjimkou je první kapitola nazvaná historie proteomiky, která by měla čitateli poskytnout stručný přehled o nejdůležitějších proteomických technologiích a jejich vývoji. Ve výsledkové části jsou potom prezentovány dva autorovy nezávislé výzkumné projekty, jejichž pojítkem je právě afinitní chromatografie. Affinity chromatography is a variant of chromatography based on the ability of biological molecules (analytes) to bind certain ligands specifically and reversibly. These unique features of the analyte and ligand interaction are then utilized for the separation of analyte from a given mixture. From the first protein-protein interaction studies done in the second half of the 1990´s, affinity separations have experienced a true renaissance in proteomics. A complete parade of affinity matrices and affinity-based experimental approaches has been developed that has found numerous applications ranging from subtraction of highly abundant proteins to study of drug target profiles to large scale mapping of posttranslational modifications. This thesis deals with the role of affinity chromatography in modern proteomics. Hence, the literature review focuses on its use in the broad proteomic field. The only exception from this is the first section entitled History of proteomics, which should provide the reader an overview of the most important proteomic technologies and their development. In the results section, two independent research projects of the author utilizing affinity chromatography are presented.
