PharmDr. Veronika Nováková, Ph.D. Title of Thesis Synthesis and study of subphthalocyanines axially modified by amino adamanthyl Photodynamic therapy is a promising approach for cancer treatment that relies on the administration of a photosensitizer, followed by illumination of the tumor. The generated oxidative stress subsequently activates several mechanisms of cell death. One of the novel groups of photosensitizers are subphthalocyanines with a conical π surface, which renders them less prone to aggregation and offers the possibility to tune their properties through axial substitution (aside peripheral modification). Due to the lipofilicity of the macrocycle, it is necessary to increase its water solubility. This work focused on the possibility of forming supramolecular host-guest complexes with hydrophilic macrocycles in order to increase the hydrophilicity of a subphthalocyanine. In particular, cucurbituril-7 (CB[7]) was chosen since it forms one of the most stable supramolecular interactions in water with 1-aminoadamantane, which was therefore proposed as the axial substituent on subphthalocyanine core. Vzniknutý oxidačný stres aktivuje niekoľko mechanizmov bunkovej smrti. Jednou z nových skupín fotosenzitizérov sú subftalocyaníny s kužeľovitým π povrchom, ktorý ich činí menej náchylnými k agregácii a umožňuje vedľa klasickej periférnej substitúcie aj ich axiálnu modifikáciu. Pretože je makrocyklus sám o sebe pomerne lipofilný, je nutné zvýšiť jeho rozpustnosť vo vode. Táto práca sa zaoberala možnosťou tvorby supramolekulárnych komplexov s navonok hydrofilnými makrocyklami s cieľom zvýšit hydrofilitu subftalocyanínu. Konkrétne bol vybraný kukurbituril-7 (CB[7]), pretože vytvára vo vode jednu z najstabilnejších supramolekulárnych interakcií s 1-aminoadamantánom, ktorý bol zamýšľaný ako axiálny substituent na subftalocyanínovom jadre. The Dissertation work is focused on the legal analysis of the current model of financing higher education with a significant emphasis on public universities, which is devoted a substantial part of the work. The first chapter is approximated legal status of individual types of high schools as subjects of law. The second chapter, devoted to the legislation of income of public universities, is followed by a characteristic means of providing state subsidies to the graphical representation of the distribution of subsidies for specific parts. Subsequently, one part is devoted to work and legislation funding of public universities in the Czech Republic. The fifth chapter is devoted to the review bodies performing inspection, assigned from the state budget. The sixth chapter brings the possibility of funding other types of universities. Then, after all these findings determine the impact of physical activity to body composition parameters which we measured by bioimpedance analysis. Data about physical activity we obtained by questionnaire from women before every measurement. Study attended 10 pregnant primiparous women, who have a physiological pregnancy. Study continued in the lactation. Overall we did 8 examinations - 2 examinations in period of gravidity, this were followed by examination on the day of birth, next one on day after birth and last 4 examinations in period of lactation. Body composition parameters were determined on devices, which work on basis of bioimpedance analysis. This method evaluates body composition via weak electric current, which passes through body during measurement. Different tissues show different resistance toward this weak current. Univerzita Karlova Farmaceutická fakulta v Hradci Králové Katedra biologických a lekárských vied Študent: Damiána Skoncová Vedúci diplomovej práce: PharmDr. Miroslav Kovařík, Ph.D. Názov diplomovej práce: Vplyv fyzickej aktivity na parametre zloženia tela tehotných a dojčiacich žien Cieľom mojej diplomovej práce bolo porovnať parametre zloženia tela v tehotenstve a v období dojčenia. Následne stanoviť vplyv fyzickej aktivity na parametre zloženia tela získané bioimpedančnou analýzou. Dáta o fyzickej aktivite sme získali z dotazníkov od žien pred každým meraním. Štúdie sa zúčastnilo 10 tehotných prvorodiaciach žien, u ktorých prebiehalo fyziologické tehotenstvo. Štúdia pokračovala aj v období dojčenia. Celkovo prebehlo 8 vyšetrení - 2 vyšetrenia počas gravidity, vyšetrenie v deň pôrodu, deň po pôrode a 4 vyšetrenia v laktácií. Parametre zloženia tela boli stanovené pomocou prístrojov, ktoré pracujú na podklade bioimpedančnej analýzy. Táto metóda hodnotí zloženie tela prostredníctvom slabého elektrického prúdu, ktorý telom prechádza počas merania. Rôzne tkanivá vykazujú rôzny odpor voči tomuto prúdu. Bioimpedančná analýza sa ukázala ako rýchly, neinvazívny a bezpečný nástroj pre hodnotenie zloženia tela aj u tejto špecifickej skupiny žien. Petra Matoušková, Ph.D. Title of diploma thesis: Effect of fern extracts on the SW480 cell line Cancer is one of the leading causes of death in the world. Although we have several drugs available for the treatment of cancer, none of them are completely effective and safe. The fundamental problem is toxicity, which experts try to eliminate by looking for other treatment alternatives like plants and their secondary metabolites with antiproliferative and antioxidant effects against free radicals, that can cause cancer. Our work is focused on the antioxidant, as well as the prooxidant effect of 4 species of ferns (Athyrium filix - femina, Dryopteris dilatata, Dryopteris borreri, Blechnum spicant), whose extracts of different concentrations were studied as potentially beneficial substances against oxidative stress on the SW480 cell line. We used the fluorescent probe H2DCF-DA to detect the production of reactive oxygen species (ROS) in cells. In the initial setup the potential prooxidant effect was monitored alongside with the terc- butyl hydroperoxide solution as a positive control. Petra Matoušková, Ph.D. Názov diplomovej práce: Pôsobenie extraktov z papradí na bunkovú líniu SW480 Rakovina je jednou z hlavných príčin úmrtí vo svete. Aj napriek tomu, že na trhu máme niekoľko dostupných liekov na liečbu nádorových ochorení, žiaden z nich nie je úplne účinný a bezpečný. Hlavným problémom býva toxicita, ktorú sa odborníci snažia eliminovať hľadaním iných alternatív liečby. Jednou z možností sú aj rastliny a ich sekundárne metabolity s antiproliferatívnymi a antioxidačnými účinkami vzhľadom k tomu, že karcinogenita môže súvisieť aj s voľnými radikálmi. V našom prípade sme pozornosť upriamili práve na antioxidačný, resp. prooxidačný efekt, konkrétne u papradí. V práci sa zameriavame na 4 druhy papradí (Athyrium filix - femina, Dryopteris dilatata, Dryopteris borreri, Blechnum spicant), ktorých extrakty o rôznych koncentráciách sme skúmali ako potenciálne prospešné látky v boji proti oxidačnému stresu na bunkovej línii SW480. Využívali sme metódu s fluorescenčnou sondou H2DCF-DA, ktorá sa používa na detekciu bunkovej produkcie ROS. Populační struktura a historie sahelu z pohledu metabolismu alkoholu

The Sahel is the northernmost part of sub-Saharan Africa. The Sahel is inhabited by a complex mixture of people, who differ in their ethnic background, language affiliation and lifestyle. The most important and most studied gene of ethanol metabolism, ADH1B, has been understudied in the Sahel region, mainly due to the low frequency of the variant rs1229984-T, which accelerates the first step of alcohol metabolism. Due to its non- African origin, the variant rs1229984-T represents a suitable marker for population history study. The eastern Sahel is where immigrants from Arabia have been settling and mating with African populations since the 7th century CE. To study population structure and history from the point of view of alcohol meta- bolism, we have used already known genotype data from fourteen Sahelian populations, which inhabit mainly the eastern part of the region. Due to the absence of the critical locus rs1229984 in the genotyping array, the samples have been subjected to sequen- cing. Combined data from the microarray and the sequencing constituted 26 loci of the ADH1B gene and its close periphery, which came from 318 samples. Genotypic data needed to be converted into their haplotypic form by a phasing prog- ram. To choose the more appropriate phasing program, the data have been arranged... Sahel je najsevernejšou časťou subsaharskej Afriky. Je obývaný komplexnou zmesou ľudí, ktorí sa líšia etnickým pozadím a príslušnosťou k jazykovým rodinám a životným štýlom. V oblasti sahelu sa štúdie najvýznamnejšiemu a najviac študovanému génu metabo- lizmu etanolu, ADH1B, venovali veľmi málo, najmä z dôvodu nízkeho výskytu variantu rs1229984-T, ktorý urýchľuje prvý krok metabolizmu alkoholu. Variant rs1229984-T vďaka svojmu mimoafrickému pôvodu však najmä vo východnej časti sahelu predsta- vuje vhodnú pomôcku pre štúdium populačnej histórie. Východný sahel totiž pred- stavuje miesto, kde sa od 7. st. n. l. usídľovali imigranti z Arábie a dochádzalo k ich miešaniu s africkými populáciami. Za účelom štúdia populačnej štruktúry a histórie z hľadiska metabolizmu alkoholu boli využité už známe genotypové dáta štrnástich sahelských populácií, pochádzajú- cich najmä z východnej časti regiónu. Z dôvodu absencie kritického lokusu rs1229984 v genotypizačnom čipe boli vzorky podrobené sekvenácii. Kombináciou dát z genotypi- začného čipu a sekvenácie bolo nakoniec analyzovaných 26 lokusov génu ADH1B a jeho blízkeho okolia, ktoré pochádzali z 318 vzoriek. Analýza naratívnych techník v diele Barabáš od Pära Lagerkvista a ich využitie pre vyjadrenie filozo-fie existencializmu

The bachelor's thesis deals with the narrative techniques in the novel Barabbas by Pär Lagerkvist, published in 1950. The first goal of the thesis is to analyse the narrative techniques and based on the thorough analysis the second goal is to find out if and to what extend they contribute to the overall comprehension of the novel. Specifically, how these techniques contribute to the portrayal of the characteristic traits of the philosophy of existence, which Lagerkvist provably treated and whose multiple issues he incorporated into the novel Barabbas, as a large majority of secondary literature written about the novel agree on. We are going to base the analysis of narrative situations on the theories and narrative terminology of Franz Karl Stanzel. The results of the analysis are going to be compared with various philosophies of existentialism. Anotácia Bakalárska práca sa zameriava na problematiku naratívnych techník v románe Barabáš (Barabbas) od Pära Lagerkvista vydanom roku 1950. Prvým cieľom práce je naratívne techniky analyzovať a na základe tejto dôkladnej analýzy je druhým cieľom práce zistiť, či a do akej miery sa podieľajú na celkovom vyznení románu; konkrétne ako sa tieto techniky podieľajú na stvárnení ideí filozofie exis- tencie, ktorú Lagerkvist preukázateľne recipoval a ktorej niektoré tézy do románu Barabáš vtelil, ako sa na tom zhoduje valná časť sekundárnej literatúry, ktorá je o románe napísaná. Naratívne techniky sa teda pokúsime interpretovať v rámci myšlienok filozofie existencializmu. Pri analýze rozprávač- skej situácie sa budeme opierať o teóriu a terminológiu naratológie Franza Karla Stanzela. Výsledky analýzy porovnáme s rôznymi filozofiami existencializmu.

Contribution of archaeology to the understanding of history related to the North-West region of Middle Danube (Possible interpretations of glass-beads findings beginning from the 7th up to the 10th century) Abstract The presented work, as to the type of findings (glass beads) and geographical scope has not yet been discussed in scientific literature. The analysis required a tailor made, new classification system of glass beads, as well as the modification of the existing relative-chronological periodization of early Middle Ages. The list of skeleton grave findings and places of beads was critically discussed (977 sites, 36 851 skeleton graves). More than 25370 beads were analyzed in an interdisciplinary way. Chemical analyses were implemented with three methods on selected types of glass. The results provide a link between the types of beads and their production technologies in relatively-chronological horizons of early Middle Ages. The styles of necklaces and their cultural interdependencies were identified. The results answer and clarify the question regarding cultural relationships between the Avarian Khanate, the Carolingian, Great Moravian and late Moravian cultures in the North-West region of Middle Danube and their contacts with other cultures outside this region. . Příspěvky archeologie k poznání dějin severozápadní části středního Podunají (Výpovědní možnosti nálezů skleněných korálků ze 7. až 10. století) Abstrakt Předkládané téma nebylo dosud z pohledu sledovaných nálezů (skleněné korálky) a geografického rozsahu v odborné literatuře předmětem zkoumání. Pro potřeby analýzy byl vytvořen nový klasifikační systém skleněných korálků, byla přizpůsobena a dotvořena starší relativně chronologická periodizace raně středověkého období a kriticky byl okomentován soupis nálezů kostrových pohřebišť a nálezů korálků (977 lokalit, 36851 kostrových hrobů). Interdisciplinárně bylo analyzováno víc jako 25370 korálků. Vybrané typy skel byly podrobeny chemickým analýzám, a to třemi rozdílnými metodami. Výsledkem je prokázání souvislosti typů korálků a technologie jejich výroby v relativně chronologických horizontech raného středověku. Identifikovány byly styly jednotlivých náhrdelníků a jejich kulturní vztahy. Dosažené výsledky zpřesňují nebo přinášejí odpovědi na otázky vztahů kultury avarského kaganátu, kultury karolinské, velkomoravské a povelkomoravské v severozápadní části středního Podunají a doklady kontaktů s jinými kulturními okruhy vně středodunajského prostoru. Vývoj pohledu japonské veřejnosti na teritoriální spor o Cušimu v 21. století

This master thesis deals with the perception of territorial dispute concerning Tsushima and Korean claims to this Japanese island. The impact of these claims is quite noticeable in the perception of Japanese society, as it is one of the current disputes between Japan and the Republic of Korea, and even in the 21st century provokes significant reactions among the residents of both countries. Reactions to this topic can be seen on the online forum Yahoo! Japan, where since the beginning of the year 2000 has been a visible increase of posts regarding the issue of Tsushima. For this reason, the platform was chosen as the subject of a study on the independent understanding of territorial claims to Tsushima in the public sphere of Japan. Using the published posts of this Internet community, the thesis deals with the way in which the perceptions of Korean claims to Tsushima developed during the years 2000 to 2021. The analysis uses the principles of resolving territorial disputes according to the theory of B. T. Sumner, who states the decisive factors for the assessment of territorial disputes in front of International Court of Justice. The thesis compares these legal factors with the circumstances as perceived by the online community of Yahoo! Japan, and at the same time focuses on the public factors... Diplomová práca sa venuje vnímaniu teritoriálnych sporov, ktoré sa týkajú Cušimy a kórejských nárokov na tento japonský ostrov. Dopad týchto nárokov je vo vnímaní japonskej spoločnosti značne badateľný, keďže ide o jeden z aktuálnych sporov medzi Japonskom a Kórejskou republikou, a aj v období 21. storočia vyvoláva značné reakcie medzi obyvateľmi oboch krajín. Ohlasy týkajúce sa tejto témy je možné pozorovať na online fóre Yahoo! Japan, na ktorom je od začiatku roku 2000 viditeľný nárast príspevkov ohľadom problematiky Cušimy. Z tohto dôvodu bola platforma vybraná ako predmet štúdie o nezávislom chápaní teritoriálnych nárokov na Cušimu vo verejnej sfére Japonska. Pomocou uverejnených príspevkov internetovej komunity práca sleduje spôsob, akým sa vnímanie kórejských nárokov na Cušimu vyvíjalo počas rokov 2000 až 2021. Pri analýze sú využité princípy riešenia teritoriálnych sporov podľa teórie B. T. Sumnera, ktorý uvádza rozhodujúce faktory pre posúdenie územných sporov pred Medzinárodným súdom spravodlivosti. Práca porovnáva tieto právne faktory s okolnosťami, ktoré vníma online komunita Yahoo! Japan, a zároveň sa sústreďuje na verejné faktory najčastejšie spomínané medzi užívateľmi. The aim of this study was to isolate at least one pure alkaloid from the extract of Geissospermum vellosii Alemão bark. The whole process involved bark processing, to obtain summary and alkaloid extract and subsequent column chromatography. GV-4, one of the 16 obtained fractions, was separated into 5 subfractions. The GV-4bsubfraction was used to isolate pure alkaloids, processed by preparative thin layer chromatography and crystallization of pure compound. The structure of pure compound was determined by using NMR, GC-MS analysis and optical rotation. This compound was identified as anhydropereirine and was tested its inhibitory activity against human cholinesterases, AChE and BuChE. Hlavným cieľom tejto práce bolo izolovať z extraktu z kôry Geissospermum vellosii Alemão aspoň jeden alkaloid v čistej forme. Celý proces zahŕňal spracovanie kôry, získanie sumárneho a alkaloidného extraktu a následnú stĺpcovú chromatografiu. GV-4, jedna zo 16 získaných frakcií, bola rozdelená na ďalších 5 podfrakcií. GV-4b podfrakcia bola využitá pre izoláciu čistých alkaloidov, spracovaná preparatívnou TLC a kryštalizáciou čistej látky. Štruktúra čistej látky bola stanovená pomocou NMR, GC-MS analýzy a optickej otáčavosti. Látka bola identifikovaná ako anhydropereirín a bola podrobená testovaniu inhibičnej aktivity voči ľudským cholínesterázam, AChE a BuChE. The main goal of this thesis is to simplify the beginners' experience with learning and independently solving the Rubik's cube. We provide different perspectives on how to find the solution for this puzzle. The implementation of all used components, chosen solving process for beginners, appearance of the application environment and the interactivity of different elements are explained and described in an easily understandable way. We included insights on the teaching process and analysis of its effectiveness in comparison to similar existing solutions. User has an option of adding their own solving algorithm in text format. The text contains user documentation and suggests possible improvements for future development. 1 Táto práca je vytvorená za účelom zjednodušiť pohľad začiatočníkom na výučbu skladania Rubikovej kocky. Prechádza rôznymi pohľadmi, ako vyriešiť tento hlavolam. Zameriava sa na popis implementácie samotného aplikovaného postupu skladania. Prib- ližuje výzor prostredia aplikácie a interaktivitu rôznych elementov naprieč jednotlivými úsekmi a zahŕňa popis všetkých aplikačných komponentov. Poskytuje náhľad do spra- covania vyučovacej časti a analyzuje jej efektivitu v porovnaní s existujúcimi riešeniami. Obsahuje užívateľskú dokumentáciu a návod pre pridanie vlastného algoritmu skladania v textovom formáte. Navrhuje prípadné vylepšenia do budúcnosti. 1
