The optimal conditions for field-amplified electrokinetic injection of cations followed by their separation based on the stereoselective interaction with sulfated cyclodextrins are discussed. Results show that addition of a short water plug together with a short plug free of cyclodextrins and with a buffer concentration similar to the BGE greatly increase the injection efficiency. A New evaluation of combined antitumor effects of natural and synthetic nuclear retinoid receptor ligands in human breast carcinoma cells

This work represents the study of the effects of biologically active ligands of nuclear retinoid X receptors, triorganotin compounds, on proteomic profile of MDA-MB-231 cells. We focused primarily on tributyl/phenyltin derivatives that suggest the combined antitumor effect when they are used with natural nuclear retinoid receptor ligand all-trans retinoic acid (ATRA). Proteomic strategies connected to PDQuest evaluation were applied in this study. Some specific proteins with tumor process association were chosen for more detail study. Our findings indicate that selected triorganotins derivatives resulted to significantly reduced level of studied proteins belonging to metabolic pathway or to other cellular processes as apoptosis or epithelial–mesenchymal transition. In the case of vimentin, the key protein of EMT process, the silencing was set up by triorganotin compounds and significantly enhanced by combination with ATRA. This additive effect on downregulation can result from the potential engagement of RXR/RAR heterodimer, considering co-addition of both partner ligands of mentioned heterodimer to the treatment. Characterization of upconversion nanoparticles by fluorescence spectrometry and capillary electrophoresis

Upconversion nanoparticles (UCNPs) are a novel class of luminescent tags for applications in life and material sciences. Unlike traditional fluorophores, UCNPs exhibit emission of shorter wavelength under near-infrared excitation (typically 980 nm). In this work, we have examined these unique photophysical properties by fluorescence spectrometry and capillary electrophoresis. UCNPs co-doped with Yb(III) and Er(III) were characterized using laboratory-made fluorescence spectrometer. We have exploited and evaluated two excitation sources and the dependence of the fluorescence of UCNPs on the relative excitation power. Moreover, capillary electrophoresis with laser-induced fluorescence (CE-LIF) detection was for the first time used for characterization of the nanoparticles. It was proved that CE-LIF is a valuable method to be used for investigation of upconversion luminiscence and monitoring of the interactions of UCNPs with other molecules of interest. Microfluidic devices combined with various bioanalytical and optical sensors proved\ntheir potential in chemical, biological, and biomedicine applications. At a droplet\nmicrofluidic device a fluid is divided into numerous droplets surrounded by a\ncontinuous immiscible fluid phase. Generated droplets serve then as\nvessels/microcapsules for delivering drugs, nutrients as well as for encapsulating of\nbiologically active particles and cells.\nDescribed experimental study deals with the development of a practical dropletbased\nmicrofluidic chip useful for production of monodisperse water/oil emulsion and\nfor encapsulation of thin-metal magnetic microsheets. A method for the fast and sensitive detection of dicarboxylic acids in atmospheric\naerosols was optimized. Three-years old seedlings of Picea abies were pre-treated under two different\ntemperatures (20x35 °C) and subsequently exposed to elevated concentration of CdO\nnanoparticles (CdONPs). Two-week exposure to airborne CdONPs of ecologically\nrelevant size and concentration significantly reduced photosynthetic light use efficiency\nin plants as well as contents of photosynthetic pigments and saccharides. In contrary,\namino acid content was increased under CdONPs. These changes were further\nmodulated by previous growth temperature. This work presents a new methodology for high-sensitivity analyses by capillary\nelectrophoresis (CE) with electrospray-ionization mass spectrometric (ESI-MS)\ndetection, based on electrophoretic focusing on an inverse electromigration dispersion\n(EMD) gradient. The technique is based on a robust electrolyte system providing a\ngradient of required properties, suitable for routine analyses of trace amounts of weak\nacids with pKa values between approx. 6.5 and 9. Examples analyses of several\nchlorophenols and sulfonamides are presented, with LODs around 3x10-9 M for spiked\ndrinking water without any sample pretreatment.
