We demonstrated that the mitogen-activated protein kinases (MAPKs) play a crucial role in cancer cell death and survival, however their role seems to be indirect. We confirmed the previously published results showing that different intracellular processes are underway in cancer cells treated with Dox.HCl compared to those incubated with HPMA conjugates Prokázali jsme, že mitogenem aktivované signalizační dráhy (MAPKs) hrají důležitou roli při smrti a proliferaci buněk, ačkoli je jejich role nepřímá. Potvrdili jsme předchozí výsledky, které ukazují, intracelulární procesy buněk inkubovaných s volným doxorubicinem se liší od procesů buněk inkubovaných s HPMA konjugáty

Studies of structure and function of promoters in C. glutamicum were used in graded expression of genes involved in biosynthetic pathway of valine (ilvBNC, ilvD, ilvE) and in competing or branching-off pathways (ilvA, leuA). Combination of particular promoter mutants resulted in tuning the whole pathway to overall higher flux of the intermediates through the pathway and in high production of valine Byla studována struktura a funkce promotorů v C. glutamicum a výsledky využity k regulaci exprese genů biosyntézy valinu (ilvBNC, ilvD, ilvE) a genů konkurenčních nebo odvětvujících se biosyntetických drah (ilvA, leuA). Kombinace mutací v určitých promotorech vedly k modulaci celé biosyntetické dráhy s následným zvýšením toku meziproduktů a zvýšenou produkcí valinu

Promoters of the amino acid-producing bacterium Corynebacterium glutamicum regulated differently in various phases of growth have been searched for. It was found that activity of promoter of the sigE gene coding for alternative sigma factor of RNA polymerase was strongly increased when the culture entered the stationary phase of growth Byly vyhledávány promotory producenta aminokyselin Corynebacterium glutamicum různě regulované v různých fázích růstu. Bylo zjištěno, že aktivita promotoru genu sigE, kódujícího alternativní sigma-faktor RNA polymerasy, byla silně zvýšena ve stacionární fázi růstu

Lectin-like receptors can modulate the cytotoxic, regulatory and dendritic cell responses by their interactions. GN8 multivalent glycoconjugates are able to internalize inside and activate NKR-P1 positive cells inducing production of IFN-gamma and expression of CD69. Polymeric conjugate based on HPMA containing cytotoxic drug (doxorubicin) and human “non-specific” immunoglobulin has a significant anti-tumour effect and does not impair immune functions of the host. C57BL/6 mice with EL-4 lymphoma, when completely cured with the conjugate, were protected against the same tumour due to a specific anti-tumour immune reaction Polymerní konjugát na bázi HPMA s doxorubicinem a normálním lidským imunoglobulinem má významný protinádorový účinek. U myší C57BL/6 může úplně vyléčit syngenní lymfom EL-4. Léčba pravděpodobné přispívá ke stimulaci imunokompetentních buněk, takže vyléčení jedinci jsou po léčbě chráněni proti nádoru specifickou imunitní reakcí
