Currently, the nursery production of plant material emphasizes the production of healthy, virus-free material. For this reason, it is necessary to have reference plant materials in which the virus is present. Some types of viruses are difficult to maintain outside the plant host. Because of the potential transmission to outdoor plantings, it is necessary to keep infected plants in special cultivation areas, and net-houses, which brings considerable financial and labour costs. Therefore, a methodology was developed in which the virus status in plant material is analysed, the infected material can be stored using cryopreservation in liquid nitrogen and then regenerated in vitro, and relevant data in GeneEver cryobox can be stored.

The goal of the preservation procedure is to practically describe the technological process of both the production of casein sealant itself and the method of application to damaged parts of the wood material, as well as the related preparation related to the cleaning of the damaged area from the remains of non-cohesive wood material, including its consolidation. The procedure primarily respects the protection of cultural heritage, especially the need to minimize interference with the original wood material. The applicability of this technique is very broad. There are many possibilities as to why and how to apply it. Cílem památkového postupu je prakticky popsat technologický proces jak samotné výroby kaseinového tmelu a způsob aplikace do poškozených částí dřevní hmoty, tak související přípravu týkající se čištění poškozeného místa od zbytků nesoudržné dřevní hmoty, včetně její konsolidace. Postup primárně respektuje ochranu kulturního dědictví, zejména potřebu minimalizace zásahu do originální dřevní hmoty. Použitelnost této techniky je velmi široká. Existuje mnoho možností, proč a jakým způsobem ji aplikovat.

Monument procedure - Measuring during repairs of carpentry structures

In this text, the author presents several measurement procedures intended for the repair of valuable carpentry structures. This is not a comletely exhaustive overview of individual options or a comparison of different methods. However, the selected examples summarize the most interesting experiences from twenty years of practice and experiments. For carpenters with varying degrees of expirience, the text tries to offer a number of inspiring insights thet can be used in a creative way even in conditions thet are quite different. Imporved measurement without unnecessary excesses decisively pushes the current limits of technical possibilities, thereby opening th way to more advanced repairs. V tomto textu autor představuje několik postupů vyměřování určených pro opravy hodnotných tesařských konstrukcí. Nejedná se o zcela vyčerpávající přehled jednotlivých možností či porovnání různých metod. Vybrané příklady však shrnují nejzajímavější zkušenosti z dvacetileté praxe a provedených experimentů. Tesařům s různou mírou zkušenosti se text snaží nabídnout řadu inspirativních postřehů, které lze tvůrčím způsobem využít i v podmínkách značně odlišných. Zdokonalené vyměřování bez zbytečných nadměrků totiž rozhodujícím způsobem posouvá stávající hranice technických možností, čímž otevírá cestu k vyspělejšímu provedení oprav.

Památkový postup definuje využití netoxické metody likvidace všech vývojových stádií biologických škůdců - hmyzu, který poškozuje historické předměty z organických materiálů prostřednictvím ošetření v přesné termokomoře za určitých tepelných a vhlkostních podmínek.

The content of the presented methodological procedure is guidance on a suitable and reliable method of laboratory analysis of rocks and crushed aggregate for the presence of naturally occurring asbestos. The created methodology follows on from the Methodological guide for sampling to determine the presence of asbestos fibers in rocks, with which it is closely linked. Together, they represent the first methodological documents dealing with the issue of asbestos in rocks and aggregates within the Czech Republic. Náplní předkládaného metodického postupu je návod vhodného a hodnověrného způsobu laboratorní analýzy hornin a drceného kameniva na přítomnost přirozeně se vyskytujícího azbestu. Vytvořená metodika navazuje na Metodický návod pro odběr vzorků pro stanovení přítomnosti azbestových vláken v horninách, se kterým je úzce propojena. Společně představují první metodické dokumenty, které se v rámci České republiky zabývají problematikou azbestů v horninách a v kamenivu.

Modeling the impacts of combined climate events on the growth, development and yield parameters of field thermophilic vegetables and oilseed rape in a decision support system in the field of agrotechnology transfer - DSSAT. This book will introduce you to the latest information technologies and their applications in agriculture, including decision support systems, crop simulation models and other computer tools. It should also help you to solve problems at the regional level on issues related to crop production and other issues related to agriculture, natural resource management and cropping systems and food security, as well as issues related to climate change and variability. Elemental analysis of vehical glass using neutron activation analysis and X-ray fluorescence analysis

Glass microfragments provide valuable information about certain vehicle crimes. The methodology describes application of X-ray fluorescence analysis (XRF) and neutron activation analysis (NAA) of elemental admixtures in vehicle glass fragments. XRF allows rapid and non-destructive test of the sample and helps in selection of protocols for detailed inspections by additional methods. NAA is time demanding, however, has potential to provide robust results for law enforcement agencies. Both methods are described together with protocols of analysis and their variants. The methodology includes results for 80+ vehicle glass samples. Mikrofragmenty skel nesou cenné informace o některých druzích zločinů spojených s vozidly. Metodika popisuje využití Rentgenové fluorescenční analýzy (RFA) a neutronové aktivační analýzy (NAA) pro stanovení prvkových přímesí ve střepech automobilových skel. RFA umožňuje rychlé nedestruktivní testování vzorku, které umožní výběr analytických postupů pro další analýzu. Metoda NAA je časově náročnější, má potenciál poskytovat robustní výsledky pro orgány činné v trestním řízení. Metodika zahrnuje popis obou metod a jejich uplatnění při analýze fragmentů skel. Součástí metodiky jsou výsledky analýzy více než 80 vzorků automobilových skel.

The early warning system against negative weather effects offers a daily updated description of abiotic and selected biotic risks threatening field production at the cadastral level. It includes their 9-day forecast. The methodology is primarily intended for agronomists,plant doctors and agricultural managers, as well as scientists, agricultural consultants and representatives of public administration. Systém včasné výstrahy před negativními vlivy počasí nabízí na katastrální úrovni denně aktualizovaný popis abiotických a vybraných biotických rizik ohrožující polní produkci. Jeho součástí je jejich 9denní předpověď. Metodika je určena především agronomům, rostlinolékařům a zemědělským managerům, stejně jako vědeckým pracovníkům, zemědělským poradcům a zástupcům veřejné správy.

The material compiles conservation measures and methods for the care of endangered butterflies of open forests, protected by the EU Habitat Directive in the Czech Republic and under climate changes. These species are Euphydryas maturna, Lopinga achine, Parnassius mnemosyne, Coenonympha hero, and partially also Zerynthia polyxena. It results from predictions of vegetational and climatic changes of forest habitats, and outlooks of forest and water management conditions. Metodika, shrnující zásady péče o populace hnědáska osikového (Euphydryas maturna), okáče jílkového (Lopinga achine), jasoně dymnivkového (Parnassius mnemosyne), okáče hnědého (Coenonympha hero) a částečně pestrokřídlece podražcového (Zerynthia polyxena), druhů světlých lesů chráněných Směrnicí o stanovištích EU, v podmínkách měnícího se klimatu. Vychází z predikcí vegetačních a mezoklimatických změn lesních biotopů, výhledů lesního hospodářství v ČR a vodohospodářských poměrů.

The methodology describes the issue of ex-situ protection of endangered plant species, especially through cultivation. It evaluates their risks and the possibilities of their elimination. It also sets out the rules for the ex-situ cultivation of endangered plant species in private gardens and demonstrates its process using the example of Minuartia smejkalii. Metodika popisuje problematiku ex-situ ochrany ohrožených druhů rostlin, zejména kultivací. Hodnotí jejich rizika a možnosti jejich eliminace. Dále stanovuje pravidla pro ex-situ kultivace ohrožených druhů rostlin v soukromých zahradách a na příkladu kuřičky hadcové demonstruje její průběh.
