Functional mapping and excitability testing of limbic structures in vivo

We have focused on analysis of functional connectivity and excitability testing of limbic structures in vivo. Results demonstrated that stimulation of ventral commisure activates preferentially dorsal hippocampus and medial entorhinal cortex V této práci jsme se zaměřili na analýzu funkční konektivity a excitability limbických struktur s pomocí stimulace ventrální komisury in vivo. Techniques of intracranial EEG processing in a model of temporal lobe epilepsy

To understand the role of various limbic structures in pathophysiology of temporal role epilepsy requires understanding of its functional organization. We have developed technique of recording of spontaneous and evoked acitvity in chronic model of temporal lobe epilepsy. In addition, we created a computer programme which allows sequential processing of large data sets and analysis and quantification of spontaneous epileptic activity and evoked potentials. This programme implements various techniques of seizure, HFO and interictal discharge detection Epilepsie je chronické onemocnění mozku charakterizované výskytem spontánních a opakovaných epileptických záchvatů. Porozumění dynamice, funkční organizaci a úloze jednotlivých struktur limbického systému je klíčové pro zkvalitnění její léčby. Za tímto účelem jsme vypracovali metodiku multikanálových záznamů elektrické aktivity mozku laboratorních potkanů a její zpracování. Vytvořili jsme program, který umožňuje semi-automatické zpracování a kvantifikaci jak spontánní epileptické aktivity, tak evokovaných potenciálů. Design of multichannel electrode for long-term recording of neocortical activity in vivo

The main aim of this work was to develop new multichannel electrodes for long-term monitoring of neocortical aktivity in vivo in freely moving animals. Designed electrode implant is fabricated using 3D printing technique. We have designed efficient technique of cleaning, implant assembling and surgical implantation V této práci jsme se zaměřili na vývoj multikanálové elektrody pro dlouhodobé záznamy korové aktivity in vivo u volně pohyblivých zvířat. Navržený typ elektrody využívá tisk na 3D tiskárně k výrobě těla elektrody a částí nezbytných pro zavedení jednotlivých kontaktů. Posttranslational modifications of proteins are important reactions, which significantly affect the function of proteins in the organism. In principle, they can be divided into enzymatic and non-enzymatic modifications. Non-enzymatic reactions include glycation (earlier called nonezymatic glycosylation), which plays an important role in the development of chronic complications of diabetes mellitus, uremia, in the process of aging and degeneration of the brain.This work deals with the study of glycated albumins (human serum albumin and bovine serum albumin). Methodologically we used nano-liquid chromatography coupled to Q-TOF mass spectrometer. In vitro modified proteins were cleavaged by trypsin and arising peptides were separated on C18 nano column with trap-column. Peptides and their modifications were analysed by high-resolution Q-TOF mass spectrometer MaXis with precision determination of mass below 2 ppm. We found some modifications of proteins. Besides well known carboxymethyllysine new ones were determined - create mass shift 78, 132 and 218. Origin of these modifications is discussed and possible structure is presented. Application of morphology filters to compensation of lateral illumination inhomogeneities in confocal microscopy images

Due to multiple distortion effects, confocal laser scanning microscopy (CLSM) images of even homogeneous specimen regions suffer from irregular brightness variations. In this paper, a fast correction method based on estimating a spatially variable illumination gain, and multiplying acquired CLSM images by the inverse of the estimated gain, is presented. The method does not require any special calibration (reference) images since the gain estimate is extracted from the CLSM image being corrected itself.
