Virus cleaning on sucrose gradient after polyethylene glycol (PEG) precipitation

First centrifugation removes bigger impurities. Viruses contained in supernatant will be precipitated PEG and NaCl. Precipitated viruses settled already in acceleration about 10000g. The impurities with different density will be removed on the sucrose gradient. Prvním krokem je odstranění větších nečistot, baktérií a buněk první centrifugací. poté je k supernatantu obsahujícímu virus přidán PEG a NaCl. Působením PEG a iontové síly NaCl dojde k vysrážení viru, který následně sedimentuje při centrifugaci již při zrychlení okolo 10000g. Na sacharózovém gradientu dojde k vyčištění viru od nečistot odlišné hustoty.

Five new quantitative tracer tests by using fluorescein and natrium chloride were carried out on underground tributaries of Jedovnický Creek in Rudické propadání Cave in Moravian Karst. Bylo provedeno 5 kvantitativních barvicích experimentů pomocí fluoresceinu a chloridu sodného na podzemních přítocích Jedovnického potoka v Rudickém propadání v Moravském krasu.

There are two main importance fermentation process in production swiss-type cheese.The first one - fermentation lactose by starter - proceeds during production of cheese curd, the second one - catabolism of lactate by Propionobacterium ssp during cellar. Jsou popsány 2 hlavní významy fermentačního procesu při výrobě sýrů švýcarského typu. První je fermentace mléčného cukru starterem. Zpracovává se během výroby produkcí sýřeniny. Druhý spočívá v katabolismu laktátu Propionobacterium ssp. během dozrávání ve sklepě.

Beginning of capillary isotachophoresis is in seventieth years last century. Theory of the isotachophoretic separation process has been elaborated during twentieth years, the instrumentations were developed and considerable number of applications were publicated in articles. ITP-CZE enables in the first step to inject a relatively large sample volume, to separate sample components and to remove undesirable components. In the second step a simplified and defined part of the sample is analysed. Real samples of sweeteners, human serum and urine were analysed. Kapilární izotachoforéza jako analytická technika se začala rozvíjet v sedmdesátých létech minulého století. V následujících dvaceti letech byla propracována teorie izotachoforetického separačního procesu, došlo k vývoji instrumentace a byl též publikován značný počet aplikací v odborných časopisech. ITP-CZE umožňuje v prvním kroku nadávkovat relativně velké množství vzorku, separovat vzorek a odstranit nežádoucí složky vzorku. V druhém kroku je analyzována jednoduchá a definovaná část vzroku. Byly analyzovány reálné vzorky umělých sladidel, lidského séra a moči.
