We also calculated the electron optical properties of NiCol column with hexapole corrector of spherical aberration. V rámci smluvního výzkum byla navržená nová generace systému pro detekci sekundárních elektronů v multibeam skenovacím elektronovém mikroskopu. The contract research was focused on the development of multi-axis measuring systems using the most accurate method of measurement based on the principle of laser radiation interference. The basic reference is a laser source with a wavelength of 633 nm. Measuring systems are intended for devices requiring measurement accuracy in the order of nanometers. Smluvní výzkum byl zaměřen na vývoj víceosých odměřovacích systémů využívajících nejpřesnější způsob měření na principu interference laserového záření. Základní referencí je laserový zdroj o vlnové délce 633 nm. Odměřovací systémy jsou určeny do zařízení vyžadující přesnost měření v řádech nanometrů. Laser ablation of calcium and aluminium atoms as a particle source for ion trap

Laser ablation is an effective method for generating particles from a variety of materials, including those with high evaporation temperatures such as aluminum. The use of high-energy laser pulses enables controlled evaporation of the material and the creation of an ablation crater. Successful results have been obtained in a pilot setup for the generation of neutral calcium and aluminium atoms by laser ablation. The method is being compared with thermal resistance oven in order to optimize the prosec generation of these particles. Subsequent spectroscopic measurements on the generated atoms are performed to optimize the absorption and emission of photons for subsequent ionization in the ion trap. A study of the transitions of neutral Ca and Al atoms shows that laser ablation provides a more efficient generation of atoms with the possibility of greater control over the process. Laserová ablace je účinnou metodou pro generování částic z různých materiálů, včetně těch s vysokou teplotou vypařování, jako je hliník. Využití vysokoenergetických laserových pulsů umožňuje kontrolované odpaření materiálu a vytvoření ablačního kráteru. V pilotní sestavě pro generování neutrálních atomů vápníku a hliníku pomocí laserové ablace byly dosaženy úspěšné výsledky. Metoda je porovnávána s tepelnou píckou s cílem optimalizovat proces generování těchto částic. Následná spektroskopická měření na generovaných atomech jsou prováděna s cílem optimalizace absorpce a emise fotonů pro následnou ionizaci v iontové pasti. Studie přechodů neutrálních atomů Ca a Al ukazuje, že laserová ablace poskytuje efektivnější generaci atomů s možností větší kontroly nad tímto procesem. Comparison of finite element simulation of tungsten nanoindentation with Berkovich and conical indenter

Finite Element Method (FEM) was used to study the difference between using Berkovich and conical indenters in nanoindentation test of tungsten specimen. The study was aimed to determine the\nmodulus of elasticity of a tested specimen and how it changes with different indenter. The FEM analysis also revealed that the modulus of elasticity was sensitive to the dimensions of both specimen and the indenter for which were made several numerical calculations with different dimensions. Analysis of secondary metabolites from Streptomyces by Raman spectroscopy in vivo

The work deals with the study of Raman spectra of secondary metabolites of bacteria of the genus Streptomyces, especially polyene antifungal agents. Purified isolates and commercial standards of these compounds were used to obtain reference spectra from which spectral markers characterizing specific details of the chemical structure were derived. Verification of the observed markers was performed using in silico Raman spectra predictions. Subsequently, these findings were used for the spectroscopic characterization of the composition of secondary metabolites in vivo, directly in bacteria growing on a solid medium. The developed methodologies will help to optimize the conditions for the biotechnological production of antibiotics. Práce se zabývá studiem Ramanových spekter sekundárních metabolitů bakterií rodu Streptomyces, zejména polyenních antifungálních látek. Purifikované izoláty a komerční standardy těchto sloučenin byly využity k pořízení referenčních spekter, ze kterých byly odvozeny spektrální markery charakterizující konkrétní detaily chemické struktury. Verifikace pozorovaných markerů byla provedena pomocí predikcí Ramanových spekter in silico. Následně byly tyto poznatky využity ke spektroskopické charakterizaci složení sekundárních metabolitů in vivo, přímo v bakteriích rostoucích na pevném médiu. Vypracované metodiky pomohou optimalizovat podmínky pro biotechnologickou produkci antibiotik. Ambiguity in Stochastic Optimization Problems with Nonlinear Dependence on a Probability Measure via Wasserstein Metric

Many economic and financial applications lead to deterministic optimization problems depending on a probability measure. It happens very often (in applications) that these problems have to be solved on the data base. Point estimates of an optimal value and estimates of an optimal solutionset can be obtained by this approach. A consistency, a rate of convergence and normal properties, of these estimates, have been discussed (many times) not only under assumptions of independent data corresponding to the distributions with light tails, but also for weak dependent data and the distributions with heavy tails. However, it is also possible to estimate (on the data base) a confidence intervals and bounds for the optimal value and the optimal solutions. To analyze this approach we focus on a special case of static problems depending nonlineary on the probability measure. Stability results based on the Wasserstein metric and the Valander approach will be employed for the above mentioned analysis. System of frequency standards at the ISI

The contribution presents a system of frequency standards developed at the Institute of Scientific Instruments of the Czech Academy of Sciences. It discusses the need for highly coherent optical frequency references for demanding experiments in spectroscopy with cooled atoms. The system comprises interconnected standards operating in the radiofrequency and optical parts of the electromagnetic spectrum, including an active hydrogen maser, a fiber laser locked to a high-finesse optical resonator, and an optical frequency comb. The relative frequency stability of these standards is analyzed, highlighting their suitability for different integration times and applications. The article also describes the experimental setup for comparing the stability of the frequency standards at the Institute of Scientific Instruments with those at the BEV laboratory in Vienna using a stabilized phase-coherent optical fiber link. The results demonstrate the excellent stability of the frequency standards reaching the -15th order of magnitude, essential for spectroscopy and ion trapping experiments. Příspěvek představuje systém frekvenčních normálů vyvinutý na Ústavu přístrojové techniky AVČR. Diskutuje potřebu vysoce koherentních optických frekvenčních referencí pro náročné experimenty v spektroskopii se zchlazenými atomy. Systém zahrnuje propojené normály pracující v rádiové a optické části elektromagnetického spektra, včetně aktivního vodíkového maseru, vláknového laseru uzamčeného na vysoce jakostní optický rezonátor a optického frekvenčního hřebene. Relativní frekvenční stabilita těchto normálů je analyzována s důrazem na jejich vhodnost pro různé integrační časy a aplikace. Dále je popsáno experimentální uspořádání pro porovnání stability frekvenčních normálů na ÚPT s těmi v laboratoři BEV ve Vídni pomocí stabilizované fázově koherentní optické vláknové linky. Výsledky ukazují vynikající relativní stabilitu frekvenčních normálů dosahující -15. řádu, která je nezbytná pro aplikace ve spektroskopii a experimenty se zachycenými zchlazenými ionty.
