Using of flow cytometry for the detection of bacterial filtration efficiency in (waste)water treatment processes

Aim of the study was to detect filtration efficiency of several (nano)materials used for the filtration/removal of E. coli bacteria or natural bacterial communities from water samples. Filtration efficiency was evaluated by commonly used culticvation techniques as well as by flow cytometry (FCM). Although filtration efficiency of certain nanomaterials detected by cultivation was very high (>99%), it was demonstarted that sample filtration does not have to lead to significant decline of total bacterial counts measured by FCM. From this viewpoint FCM can be considered as a suitable analytical method for the assessment of wastewater treatment processess efficiency, which, compared to cultivation techniques, possess some disadvantages (difficult standardization) but also has several advantages (speed, more realistic measurement conditions). Cílem studie bylo detekovat účinnost filtrace vzorků vodních suspenzí bakteriálních kultur E. coli resp. odtoků z čistíren odpadních vod pomocí různých druhů filtračních (nano)materiálů. Účinnost filtrace byla posuzována nejen pomocí standardně používaných kultivačních technik, ale také pomocí průtokové cytometrie (FCM). Přestože účinnost filtrace některých vzorků stanovená kultivačně byla velmi vysoká (>99%), bylo zjištěno, že proces filtrace těchto vzorků zároveň nemusí nutně mít stejně výrazný vliv na změnu celkových počtů bakterií detekovaných ve filtrovaném vzorku pomocí průtokové cytometrie. Průtokovou cytometrii lze tedy chápat jako perspektivní plnohodnotnou analytickou metodu pro odhad efektivity procesů čištění odpadních vod, která má v porovnání s kultivací některé nevýhody (obtížná standardizace), ale také četné výhody (rychlost, podmínky měření odpovídající realitě).

Phosphorus Removal from Wastewater via Environmentally Friendly Technologies

Our research is recently focused on the phosphorus removal from wastewater treatment plant outlets. We chose two municipal wastewater treatment plants (with and without phosphorus removal) and tested zerovalent and hexavalent nanoiron for phosphorus removal. Ferrates showed promising results even for smaller dosages, whilst NZVI showed not satisfactorily results for full-scale application. Next work will include phosphorus fractionation and fate of different phosphorus fractions at the tertiary treatment. Increasing interest on xenobiotics, hormons, endocrine disruptors, heavy metals and PPCPs and their removal from the plant outlets will extend our testing in the future.

The surface processing and refinement of metals leads to the wastewater production that contains toxic and hazardous compounds, i.e. heavy metals and cyanides. The recent wastewater treatment technologies usually offer inadequate solutions that are either not so efficient or economic. Our environmentally friendly solution offers novel attitude based on the application of specific iron compounds. Povrchové úpravy kovů vytvářejí odpadní vodu, která obsahuje nebezpečné a toxické sloučeniny jako např. těžké kovy a kyanidy. Současné technologie čištění nabízejí řešení, která jsou buďto neúčinná nebo neekonomická. Naše ekologicky šetrné řešení tyto nevýhody smazává - je založeno na aplikaci různých valenčních stavů železa.

Greywater is the leftover water from baths, showers, hand basins and washing machines only, what implies that it is free of faeces and urine. Greywater can be recycled on-site (called white water) for flushing toilets, urinals, landscape irrigation and constructed wetlands. The heat energy reuse from greywater has been recently discussed as a hot issue. Paper deals with the new British Standard BS 8525-1(2) that introduce the issue of greywater in buildings, as well as progress in the new Czech standard preparation. A various technologocial designs are introduced including return of investment analyses. Šedou vodou nazýváme odpadní vody neobsahující fekálie a moč, které odtékají z umyvadel, van, sprch, dřezů, apod. Šedou vodu je možno po úpravě používat jako vodu provozní (tzv. bílou vodu) pro splachování záchodů, pisoárů a zalévání zahrad. Zajímavé se jeví také znovuvyužití tepelné energie obsažené v šedých vodách. Příspěvek diskutuje již existující britskou normu BS 8525-1(2), která řeší problematiku využití šedých vod v budovách a dále pak postupy přípravy české normy a návrhy různých technologických uspořádání spolu s ekonomickým zhodnocením jednotlivých technologií.

Emergence spillways are very important for phosphorus catchment balance. Our measurememnts showed that one storm rainfall brings to the recipient so much phosphorus like two months of general inlet. Therefore, it is necessary to change sampling methods and plans. Odlehčovací komory jsou důležité pro bilance fosforu v povodí. Dle našich naměřených zkušeností přináší jedna přívalová srážka do recipientu tolik fosforu jako dva měsíce standardního přítoku. Proto je nutné změnit vzorkovací metody a odběrové plány.

The presented research is focused on the state of natural regeneration (Norway spruce and Rowan), 10 years after total death of parent stands in the core untouched part Bavarian Forest National Park. The 16 permanent plots 40m x 40m are located on the southeast of Bavaria. We determined exact height, size of the last height increment, condition and species of trees. Number of saplings were counted and classified by heights. These number ranged from 53 to 17276 saplings per hectare. The spruce species composition was 96.7%. The height structure of spruce shows that almost 73% found spruce trees reached heights greater than 0.5 m. The average height increment increased with increasing height of saplings. This paper presents state of natural regeneration in mountain spruce forests in Bavarian Forest National Park without the human influence in summer 2010, 10 years after total death of parent stands.
