It is possible to detect alcohol in human body only in a relatively short time after ethanol consumption. Ethylglucuronide as an alcohol metabolite, can be determined in body fluids, tissues and hair even after several days. ITP-CZE enables in the first step to inject a relatively large sample volume, to separate sample components and to remove undesirable components. In the second step a simplified and defined part of the sample is analysed Detekce alkoholu v těle ve formě etanolu je možná jen v relativně krátkém čase. Etylglukuronid může být stanoven v tělních tekutinách, tkáních a vlasech po několika dnech. ITP-CZE umožňuje v prvním kroku nadávkovat větší objem vzorku, rozseparovat složky vzorku a odstranit nežádoucí složky. V druhém kroku je analyzována zjednodušená a definovaná část vzorku

The principles of capillary zone electrphoresis and isotachophoresis, their instrumentation and on-line connection are described, and the possibilities of choice of background, resp. leading and terminating electrolyte are discussed. Jsou popsány principy kapilární zónové elektroforézy a izotachoforézy, jejich instrumentace a on-line spojení a diskutovány možnosti volby základního, event. vedoucího a koncového elektrolytu.

Application of the generalized dispersion theory for an exact solution of migration velocity and zone broadening in free-flow electrophoresis

The aim of the work was to find the optimum dimensions and separation conditions for maximum production of compounds of given purity at the basis of exact analysis of transport processes. The methods of solution of partial diferential equations for the calculation of dispersion coefficients as functions of time, separation cell dimensions, separation medium flow-rate and parameters of separated compounds were demonstrated. Cílem práce bylo nalézt optimální rozměry a separační podmínky pro maximální produkci látek o dané čistotě na základě exaktní analýzy transportních procesů. Byly demonstrovány metody řešení parciálních diferenciálních rovnic pro výpočet koeficientů disperze jako funkcí času, rozměrů separační komory, průtoku separačního media a parametrů separovaných látek.

Capillary zone electrophoresis proved to be a very useful method for analyses of many kinds of samples. The most often used background electrolyte system is borate buffer containing usually sodium, ammonium or Tris. Using of borate buffer we have proven, that not only coion but also counterion can be used for optimization of the separation. Experiments and calculations explaining behavior of analytes in sodium and ammonium system were verified by computer simulation using programme Simul 4.0. Kapilární zónová elektroforéza je analytická metoda oblíbená v řadě rutinních analýz. Jedním z nejvíce používaných elektrolytových systémů je tzv. borátový pufr, obsahující nejčastěji sodík, amonium nebo Tris. Na příkladu borátového elektrolytu je demonstrováno, že nejen coion ale i protion použitého elektrolytu může být využit k optimalizaci separace. Experimenty a výpočty vysvětlující chování analytů v sodném a amonném systému byly ověřeny počítačovými simulacemi využívajícími program Simul 4.0.

The determination of ammonia by means of the contactless conductivity detector is presented. For preconcentration of atmospheric amonia, a cylindrical wet effluent diffusion denuder was used. Je prezentování stanovení stopových koncentrací amoniaku ve vzduchu pomocí bezkontaktního vodivostního detektoru. Pro předkoncentrování atmosférického amoniaku byl použit cylindrický difúzní denuder se stékajícím filmem absorpční kapaliny.

A new method for continuous determination of formaldehyde in air based on preconcentration of formaldehyde in the cylindrical wet effluent diffusion denuder and subsequent chemiluminescence detection of formaldehyde in the denuder concentrate is presented. Je prezentována nová metoda pro kontinuální stanovení formaldehydu ve vzduchu, založená na prekoncentraci formaldehydu v cylindrickém difúzním denuderu se stékajícím filmem absorpční kapaliny a následné chemiluminiscenční detekci formaldehydu ve vzniklém koncentrátu.
