The methodology utilizes a portable liquid chromatograph for rapid detection and identification of selected chemical warfare agent simulants and degradation products. It is primarily designated for SÚJCHBO, v. v. i. to control compliance with the law 19/1997 Sb. and for research activity. Metodika využívá přenosný kapalinový chromatograf pro rychlou detekci a kvantifikaci rozkladných produktů BChL a simulantů BChL. Je určena zejména pro potřeby SÚJCHBO, v. v. i. v rámci činností v oblasti VaV a zkušebnictví a při kontrolní a dozorové činnosti v oblasti dodržování Zákona č. 19/1997 Sb. Oxidative Potential of PM1 and PM2.5 Urban Aerosol and Associated Elements in Three Simulated Lung Fluids

Mass concentration of particulate matter (PM) has been used in several epidemiologic studies as an indicatior conenecting PM concentrations with human health effects (Ostro, 1993). However, mass concentration of PM doesn´t consider the different compositions and toxicological effects of its components. Majority of PM mass constitutes low-toxicity components, while minority of trace components may have high toxicological activity (Tonne, 2012). Oxidative potential (OP), defined as the ability of PM to induce oxidative stress, is in recent years recognized as one of the main biological mechanisms considered to be contributing to negative impacts from air pollution exposure. Oxidative stress is caused through the capability of PM to generate reactive oxygen species (ROS) within the lung, which leads to pro-inflammatory responses that can ultimately results in cell apoptosis (Borlaza, 2021). Development of divergent-flow isoelectric focusing chip for separation of biological agents

Isoelectric focusing (IEF) is an efficient method for separation of different bacterium species. Divergent flow format of IEF (DF-IEF) enables continual separation of large volumes of samples and fractionation of the separated analytes for later analysis. The mentioned properties enabled development of DF-IEF chip for separation of H2O2 inactivated biological agents (B-agents). The chip was designed to perform separation in vertical alignment which solved issues connected with sedimentation of bacterial cells. The separation method was optimized using colored markers of isoelectric point and four different b-agents were separated using the DF-IEF chip. The obtained fractions were subsequently analyzed with matrix assisted laser desorption/ionization mass spectrometry (MALDI-MS). The mass spectra proved that the developed DF-IEF chip is an effective tool for separation of inactivated B-agents. Izoelektrická fokusace (IEF) je účinnou metodou schopnou separovat jednotlivé druhy bakterií. IEF v rozbíhavém toku (DF-IEF) umožnuje kontinuálně separovat větší objem vzorku a separované analyty jímat do frakcí k pozdější analýze. Těchto vlastností bylo využito při vývoji DF-IEF čipu pro separaci biologických agens (B-agens) inaktivovaných parami H2O2. Čip byl navržen pro separaci ve vertikální poloze, která zabránila sedimentaci běžně se vyskytující u bakteriálních vzorků a znemožňující účinnou separaci. Po optimalizaci separační metody za využití barevných markerů izoelektrického bodu byla provedena separace čtyř druhů B-agens. Všechny získané frakce byly dále analyzovány pomocí hmotnostní spektrometrie s matricí asistovanou laserovou desorpcí/ionizací (MALDI-MS). Hmotnostní spektra prokázala účinnost vyvinutého DF-IEF čipu pro separaci inaktivovaných b-agens. Keywords: isoelectric focusing; B-agents; chip Available in digital repository of the ASCR

Elements' bioaccessibility and oxidative potential of urban aerosol

Atmospheric aerosols (Particulate Matter, PM) have significant impact on health and environment. The ability of aerosol to produce reactive oxygen species (ROS) is one of the widely proposed mechanisms related to the negative effects of aerosol on human health. Oxidative stress, caused by the production of ROS, is associated with several cardiovascular and respiratory diseases.\nThe capacity of aerosol to produce ROS is also called oxidative potential (OP). Although ROS generation capacity of PM exhibits a good correlation with PM mass concentration, it may depend more on physico-chemical properties of PM. Elements bound to PM play a significant role in contributing to the OP of aerosol. Exposure to elements was linked with several health issues (e.g. respiratory and cardiovascular diseases, asthma, inflammatory effects, lung cancer). Atmosférické aerosoly (ParticulateMatter, PM) mají významný dopad na zdraví a životní prostředí. Schopnost aerosolů produkovat reaktivní formy kyslíku (ROS) je jedním z často navrhovaných mechanismů souvisejících s negativními účinky aerosolu na lidské zdraví. Oxidační stres, který je způsobený zvýšenou koncentrací ROS, je spojován s celou řadou kardiovaskulárních a respiračních onemocnění. Schopnost aerosolu produkovat ROS je nazývána oxidativní potenciál (OP). Přestože schopnost PM tvořit ROS je dobře korelována s hmotnostní koncentrací PM, může více\nzáviset na fyzikálně-chemických vlastnostech PM. Bylo prokázáno, že prvky vázané na PM významně přispívají k OP aerosolu. Expozice prvkům bývá spojena s několika zdravotními problémy (např. respirační a kardiovaskulární onemocnění, astma, zánětlivé procesy, rakovina plic).
