Activation of the protein kinase B and glycogen synthase kinase-3 signalling pathway during transient differentiation of human colon cancer HT-29 cells

Using synthetic peptides which reproduce phosphoacceptor sites in the molecule of GSK-3, PKB an eIF2B, we found that phosphorylation and activity of PKB increased very early during in vitro differentiation induced by sodium butyrate. This was inhibited by wortmannin, the selective inhibitor of phosphoinositol-3 kinase, indicating that the control of PI3 kinase/PKB/GSK-3 signalling pathway is the early event of transient differentiation of the human colon cancer cells HT-29.

Inherited predisposition to hyperproduction of IgE against common environmental allergens. It is a major risk factor for clinical expression of allergic diseases.The prevalence of atopic disorders is rising worldwide, but the reason for this is unknown. Moreover, the allergens triggering and supporting the development of atopy vary in different areas. The reason for this most likely includes diversity of allergens prevalence in different environment. Therefore, determination of major allergens is important when performing diagnostic testing and selecting immunotherapy. It also could provide important clues to the etiology of atopic disorders in a certain geographical area. Atopie je dědičná náchylnost k nadprodukci IgE v reakci na běžné alergeny životního prostředí. Atopie je hlavním rizikovým faktorem pro rozvoj klinických příznaků alergického onemocnění. V celém světě dochází k nárustu atopických onemocnění, ale důvod, proč dochází k tomuto nárustu není znám. Ačkoli bylo zformulováno několik hypotéz, uvažujících příčiny tohoto nárustu jako je hygiena, dieta smog, tabákový kouř, etiopatologie onemocnění je neznámá. Pravděpodobně ke vzniku onemocnění přispívá velké množství změn v životním stylu. Určení hlavních alergenů je důležité pro vytvoření vhodných diagnostických testů a pro cílenou imunoterapii. To může přispět i k objasnění etiologie atopického onemocnění v různých zeměpisných oblastech.
