CA IX is a cell surface protein that exerts capacity of binding cell surface receptors, and is associated with several types of human carcinomas. Monoclonal antibody M75 recognizes specifically an epitope identified as six repeats of amino acid sequence PGEEDLP, localized in the proteoglycan like domain of CA IX. Structural study of the M75 Fab fragment in complex with its epitope peptide PGEEDLPGEEDL is presented.

Byla určena rentgenová struktura komplexu proteázy HIV-1 (PR) s fenylnorstatinovým inhibitorem Z-Pns-Phe-Glu-Glu-NH(2) při rozlišení 1,03 A, nejvyšším rozlišení, které bylo doposud uvedeno pro kterýkoli komplex HIV PR. Tento inhibitor vykazuje subnanomolární hodnoty K(i) jak pro PR divokého typu, tak pro variantu představující jednu z nejběžnějších mutací spojenou s vývojem rezistence. Struktura obsahující fenylnorstatinovou část chirality (2R,3S) vykazuje jedinečný obraz vazby vodíkových můstků na dva katalytické aspartátové zbytky. Toto vysoké rozlišení umožňuje zjistit donorové a akceptorové vztahy těchto vodíkových můstků a určit proton sdílený oběma katalytickými zbytky. Je také navržen strukturální mechanismus nenarušené inhibice mutanty proteázy Val82Ala založený na energetických výpočtech a analýzách The x-ray structure of a complex of HIV-1 protease (PR) with a phenylnorstatine inhibitor Z-Pns-Phe-Glu-Glu-NH2 has been determined at 1.03 A, the highest resolution so far reported for any HIV PR complex. The inhibiot shows subnanomolar Ki values for both the wild-type PR and the variant representing one of the most common mutations linked to resistance development. The structure displays a unique pattern of hydrogen bonding to the two catalytic aspartate residues. The high resolution permints to assess the donor/acceptor relations of this hydrogen bonding and to indicate a proton shared by the two catalytic residues. Structural mechanism for the unimpaired inhibition of the protease Val82Ala mutant is also suggested, based on energy calculations and analyses

Mammalian fertilization includes highly regulated biochemical interactions between complementary molecules located on the surfaces of both gametes as well as those present in the natural environment of the gametes. Boar seminal plasma proteins AQN, AWN spermadhesins and DQH protein that are bound to the sperm at ejaculation play important role in the reproduction process. Binding of sperm to epithelial cells, recognition of gametes and binding of sperm to glycoprotein of zona pellucida are mediated by protein-saccharide interaction, lektin-type interaction. Another type of interaction (protein-protein) participates in the formation of aggregated forms, which seem to be the natural functional forms of seminal plasma proteins. Protein aggregates form the sperm coating layers and their re-modeling in the female reproductive tract is imporatnt for the sperm capacitation and for the primary binding of spermto the ovum. Savčí reprodukce zahrnuje vysoce regulované biochemické interakce mezi komplementárními molekulami lokalizovanými na povrchu spermie a vajíčka a rovněž s látkami přítomnými v přirozeném okolí obou gamet. Proteiny kančí semenné plasmy AQN, AWN spermadhesiny a DQH protein, které jsou vázány na spermii během ejakulace, hrají důležitou roli v procesech reprodukce. Vazba spermie k buňkám oviduktu, rozpoznávání gamet a vazba spermie ke glykoproteinové síti zony pellucidy je zprostředkována protein-sacharidovou interakcí, lektinovou vazbou. Přirozenou funkční formou proteinů kančí semenné plasmy se zdají být jejich agregované formy, při kterých se uplatňuje další typ interakce (protein-protein). Proteinové agregáty tvoří povrchovou vrstvu spermie a její remodelování v samičím reprodukčním traktu je důležité během kapacitace spermie a pro primární vazbu spermie na vajíčko.

Během analýzy buněčné linie HT29 jsme připravili několik stabilních klonů, které se lišily expresí některých molekulárních markerů, např. fosforylací proteinu Src, aktivitou PKCbeta2, expresí CEA-CAM1 a odpovědí na diferenciační účinek butyrátu sodného During our analysis of HT29 cell line we established several clones, which differed in the expression of some molecular markers, e.g. phosphorylation of Src protein, PKCbeta2 activity, CEA-CAM1 expression and in the response to the differentiating effect of sodium butyrate, indicated by activity of alkaline phosphatase

Two antibodies against angiotensin receptor AT2 prepared against two short sequences recognized the receptor on Western blots of several rat and mouse tissues where AT2 is expressed. Both antibodies recognized the identical proteins; they lost this property after immunoabsorption with their antigen peptides. With one antibody, we were able to immunoprecipitate the receptor. Dvě protilátky proti angiotensinovému receptoru AT2 připravené proti dvěma krátkým sekvencím rozeznávaly receptor na Western blotech membrán různých tkání potkana a myši, v nichž se AT2 exprimuje. Obě protilátky rozeznávaly tytéž bílkoviny a po vysycení příslušným antigenním peptidem tuto schopnost ztratily. Jedna protilátka se osvědčila při imunoprecipitaci receptoru.

Male infertility due to severe azoospermia has been associated with the absence of spermatozoa in ejaculated. We tested the ejaculates (IVF latoratory Pranatal Ltd) from 30 patients with azoospermia for the presence of cells from defferent stages of spermiogenesis. Monoclonal sntibodews against intraacrosomal human sperm proteins were used in the immunofluorescence test for detection of acrosomal proteins in the acrosomal precursors and acrosomes.

The cytoskeleton of sperm head consists of resistant structural proteins of the nucleus and of perinuclear theca, which are largely rosponsible for the shaping of the nucleus. three mahjor cyteskeletal proteins, actin, tzubnulin and spectrin, are present in the head of mammalian spermatohzoa. Maoreober, the mictotubular cytoskeleton also contains other dynamic proteins, which have a morphological function within the cell and which are responsible for cell signalling processes.

The results show the significant role of CD4+ T cells, macrophages and NO produced by macophages during the rejection of the corneal allo- and xenografts. Výsledky ukazují na významnou roli CD4+ T buněk, makrofágů a makrofágy produkovaného NO v procesu rejekce alo- i xenotransplantátů rohovek.
