This bachelor thesis is focused on the design of an external type of UHF sensor, which will be placed in the gap of the transformer vessel. First, the waveguide-to-coax adapter is simulated in Ansys HFSS software. Based on the piece of knowledge, the sensor was constructed. The sensor uses two types of waveguide couplers for measuring, monopole probe and loop coupling. The simulated and constructed sensor is then compared for functionality and sensitivity. Tato bakalářská práce se zabývá návrhem externího typu UHF sensoru, který bude umístněný na štěrbině transformátorové nádoby. V prvním kroku byl vlnovod-koax adaptér simulován v programu Ansys HFSS. Na základě poznatků byl zkonstruován sensor. Sensor využívá pro měření dva typy buzení vlnovodu, buzení proudovou sondou a buzení magnetickou smyčkou. Simulovaný a vyrobený sensor je porovnán z hlediska funčnosti a citlivosti.

The proposal consists of three separate buildings connected with a pedestrian path. Two buildings fill the structure of the block and one main library building is placed in between them. Form of the plan of the main building is based on the tweening of the two edge buildings and is reflecting their geometrical relation. Main idea of creating a barrier free pedestrian path across the whole site and thus connecting the two bordering streets is realized by a series of communications such as ramps, stairs and elevator. Program of the site and its buildings is symmetrically spreading from the middle, where the main library building stands . Emphasizing the spread of the knowledge from the library through culture into the city and allowing regular pedestrians to experience it. Two edge buildings serve as basic structures providing shelter and infrastructure for operating different programs and facilities such as cafe, theater, performing events, exhibitions, lectures etc. Main library building is an enclosed building with different spaces for studying, working, absorbing information and community meetings. Návrh sestává ze tří samostatných propojených budov se stezkou pro pěší. Strukturu budovy vyplňují dvě budovy bloku a mezi nimi je umístěna jedna hlavní budova knihovny. Forma plánu hlavní budovy je založena na doplnění dvou okrajových budov a odráží jejich geometrické uspořádání vztah. Hlavní myšlenka vytvoření bezbariérového chodce cesta přes celý areál a tím spojení obou sousedících ulice je realizována řadou komunikací jako např rampy, schodiště a výtah. Program lokality a jejích budov se symetricky šíří od středu, kde je hlavní knihovna stavební stojí. Důraz na šíření znalostí od knihovny přes kulturu do města a povol běžní chodci, aby to zažili. Slouží dvě okrajové budovy jako základní struktury poskytující úkryt a infrastrukturu pro provoz různé programy a zařízení, jako je kavárna, divadlo, pořádání akcí, výstav, přednášek atd. Hlavní budova knihovny je uzavřená budova s různými prostory pro studium, práci, vstřebávání informací a komunitní setkání.

This work focuses on designing a pay-per-click advertising campaign through Facebook Ads for the young outdoor brand Flipky., of which I am a co-founder. The work is divided into three parts: theoretical, analytical and proposal. The theoretical part aims to provide a brief overview of the necessary information for understanding marketing, paid advertisements, specifically focusing on marketing on Facebook. The second analytical part focuses on analyzing the current state and identifying opportunities for improvement of marketing for Flipky. The final proposal part takes into account findings of the previous sections and proposes a solution for the summer campaign 2023 for the brand Flipky. Táto práca sa zameriava na navrhnutie reklamnej kampane typu pay-per-click prostredníctvom reklám na Facebooku pre mladú outdoorovú značku Flipky., ktorej som spoluzakladateľom. Práca je rozdelená na tri časti: teoretickú, analytickú a návrhovú. Teoretická časť má za cieľ poskytnúť stručný prehľad nevyhnutných informácií pre pochopenie marketingu, platených reklám, s osobitným zameraním na marketing na Facebooku. Druhá analytická časť sa zameriava na analýzu súčasného stavu a identifikáciu príležitostí na zlepšenie marketingu pre značku Flipky. Posledná návrhová časť zohľadňuje predchádzajúce časti a navrhuje riešenie pre letnú kampaň 2023 pre značku Flipky.

The aim of this bachelor's thesis is to analyze the Čistírna u Kostelíčka organization from the marketing point of view to propose a concept to change promotion of the organization in business-to-business market. In the theoretical part the definitions of services, marketing and frameworks used, including the 7 Ps marketing mix, microenvironment, and marketing research, to analyze the current situation of the organization are defined. The analytical part is focused on analyzing the current situation of the organization by using the frameworks defined in the theoretical part and identifying the main factors used for the proposal suggestions part. The proposal suggestions part identifies changes in promotion of the services that the company offers, and their financial costs and time schedule the changes should be done in. Cílem této práce je analyzovat firmu Čistírna u Kostelíčka z marketingového pohledu tím způsobem, aby mohl být navržen koncept pro změnu propagace firmy na B2B trhu. Teoretická část obsahuje definici služeb, marketingu a rámců použitých pro analýzu současného stavu organizace, kde jsou zmíněny marketingový mix služeb 7P, mikroprostředí a marketingový výzkum. Analýza současného stavu využívá nástroje popsané v teoretické části a je zaměřena na rozpoznání hlavních faktorů, na základě kterých je vytvořena návrhová část. V návrhové části jsou identifikovány návrhy na změnu propagace služeb, které organizace nabízí a jejich finanční náklady a také časový plán ve kterém by měly být změny provedeny.

Řízená evoluce enzymů na mikrofluidním čipu

This thesis focuses on the directed evolution of enzymes using Fluorescence–Activated Droplet Sorting (FADS) on a microfluidic chip. A new FADS system was constructed, validated for high-throughput optical detection of microfluidic droplets, and coupled with real-time dielectrophoretic sorting. The system was benchmarked using two different populations (DhaA31, ancHLD-RLuc) of Haloalkane dehalogenases (HLD) expressed in Escherichia coli cells and one population of empty pIDR9 vector in a ratio of 5:45:50, mimicking the protein library. The collected and analyzed sorting data show that the droplet intensities were recorded in the same proportion as the theoretical ratio. The library of HLD mutants with insertions and deletions was then sorted using the system to obtain 1 % of the variants with the highest activity and the results are reported. The FADS data analyzing system was programmed in LabVIEW bundled with Python, with functions for initial analysis, prediction of threshold voltage, validation and graphical interpretation of data, and final reporting of processes. Tato práce se zaměřuje na řízenou evoluci enzymů pomocí fluorescenčně řízeného kapénkového třídení (ang. zkratka FADS) na mikrofluidním čipu. Byl sestaven nový třídicí systém, který byl validován pro vysoce výkonnou optickou detekci mikrofluidních kapének spojenou s dielektrickým tříděním. Systém byl testován pomocí dvou různých (DhaA31, ancHLD-RLuc) populací haloalkan dehalogenas (HLD) exprimovaných v buňkách Escherichia coli a jedné populace prázdného vektoru pIDR9 v poměru 5:45:50, což imituje proteinovou knihovnu. Naměřená a analyzovaná data z experimentu ukazují, že intenzity kapek ve validačním experimentu byly zaznamenány v stejném poměru jako teoretický poměr. Poté byla knihovna HLD mutantů s inzercemi a delecemi tříděna pomocí FADS, za účelem získaní 1 % nejaktivnějších variant. Program pro analýzu dat pro FADS byl programován v prostředí LabVIEW s využítím jazyka Python, s funkcemi pro počáteční analýzu, predikci prahového napětí, validaci a grafickou interpretaci dat a konečnou zprávu o experimentu.

Cars are getting more and more complicated as both technology and government regulations evolve. This fact is of concern for individual owners as well as fleet operators as they are getting much more expensive to maintain. To reduce maintenance costs and downtime to a minimum, owners should have an easy-to-use monitoring tool that would notify them about potential problems and let them fix them before they cause more damage. This thesis aims to create a device that would be mounted inside the car and would monitor and report important engine parameters to an external server for further processing. The theoretical part of this thesis describes current OBD II regulations and communication protocols used in the majority of vehicles currently on the road and discusses currently available low power wide networks (LPWAN) and global navigation satellite systems (GNSS). The practical part describes the design of an OBD II monitoring device with support for the most commonly used protocols in Europe, NB-IoT network, and a positioning system. The last chapter analyses the cost of a single device and its reduction for potential commercial use. Automobily sa s vývojom technológie a vládnych nariadení stávajú viac a viac komplikované. To sa však stáva problémom nie lel pre individuálnych majiteľov vozidiel ale aj pre prevádzkovateľov flotíl, kde sa im zvyšujú náklady na údržbu a prestoje. Aby majitelia vozidiel mohli znížiť svoje náklady na minimum, mali by mať nainštalované jednoduchej monitorovacie zariadenie, ktoré ich môže upozornit na potenciálne problémy a predísť tak oveľa nákladnejším opravám. Cieľom tejto práce je vytvoriť zariadenie, ktoré sa umiestni pevne vo vozidle a bude monitorovať a odosielať doležíté motorické parametre na vzdialený server pre ďalšie spracovanie. V teoretickej časti práce sú opísané aktuálne OBD nariadenia, používané komunikačné protokoly v súčasných autách, dostupné nízkoenergetické siete pre rozsiahle územie (LPWAN) a navigačné systémy (GNSS). Praktická časť sa zaoberá návrhom OBD II ˙monitorovacieho zariadenia, ktoré podporuje najpoužívanejšie komunikačné protokoly v Európe, NB-IoT sieť a systém na určenie polohy. Posledná kapitola analyzuje výslednú cenu zariadenia a opisuje jej prípadnú optimalizáciu pre komerčné použitie.

The aim of this work is to analyze various search engines with the intention to select the most suitable for the creation of a tool that should be able to perform security testing. Comparisons were made between Shodan, ZoomEye, Nexpose, Censys and BinaryEdge. These search engines were compared according to different criteria, namely their perfomance and publicly available information. Shodan and ZoomEye were chosen as the most suitable ones and as a result, web application was created. This application is able to retrieve search results from engines' servers using their API's and is able to store these results in a database. The thesis includes a description of a variety of industrial and energy protocols, as well as industrial and energy networks and their security features. Moreover, a few of the most well-known attacks on these networks were described. The last goal of the work is to test and evaluate the implemented tool on at least two devices. Cílem této práce je analyzovat různé vyhledávače a vybrat ty nejvhodnější pro vytvoření nástroje k testování průmyslových a energetických zařízení z pohledu bezpečnosti. Bylo provedeno porovnání mezi Shodan, ZoomEye, Nexpose, Censys a BinaryEdge. Tyto vyhledávače byly porovnávány podle různých kritérií, například výkonnosti a jejich dalších veřejně dostupných informací. Jako nejvhodnější byly vybrány Shodan a ZoomEye a pomocí těchto vyhledavačů byla vytvořena webová aplikace. Tato aplikace je schopna načítat výsledky vyhledávání ze serverů pomocí jejich rozhraní API a je schopna je ukládat do databáze. Součástí práce je popis průmyslových a energetických protokolů, sítí a jejich bezpečnostních prvků. V rámci práce byl také popis několika nejznámějších útoků na tyto sítě. Posledním cílem práce je provést testování a evaluaci realizovaného nástroje na minimálně dvou zařízeních.

This work provides a general overview of the properties and possibilities of underwater wireless communication and its configurations. It also contains information about the refractive index in general and in the aquatic medium, its determination for certain environments, and the effect on underwater wireless communication. Finally, there is an overview of the structure and function of the program that was created. Tato práce poskytuje obecný přehled vlastností a možností podvodní bezdrátové komunikace a jejích konfigurací. Obsahuje také informace o indexu lomu obecně a ve vodním médiu, dále jeho stanovení pro určité prostředí a jeho vlivu na podvodní bezdrátovou komunikaci. Na závěr je uveden přehled struktury a funkce vytvořeného programu.

The main goal of this work is to increase the quality of object tracking in three-dimensional space using the fusion of radar and video data obtained from traffic monitoring scenes. The thesis presents several methods dealing with collecting point pairs from both sensor coordinate systems for spatial calibration of sensors, with an emphasis on automation. It solves the association problem of multiple detected objects at once using a modified Hungarian algorithm. The work also shows some methods of predicting the distance of objects detected by video using a dataset obtained from radar. Output from the fusion model provides spatially extended tracks and more accurate counting of tracked objects than radar or video models alone, which can be used for further analysis of traffic in the observed scene. Hlavním cílem této práce je zvýšení kvality sledovaní objektů v třírozměrném prostoru pomocí fúze radarových a video dat získaných ze scén sledujících dopravu. Práce uvádí několik metod zabývajících se sbíráním párů bodů ze souřadnicových systémů obou senzorů pro prostorovou kalibraci s důrazem na automatizaci. Práce dále řeší problém asociace více detekovaných objektů naráz za pomoci modifikovaného Maďarského algoritmu. Práce rovněž ukazuje některé způsoby predikce vzdálenosti objektů detekovaných videem pomocí datasetu získaného z radarových dat. Výstup z fúzního modelu poskytuje prostorově rozšířené sledování dráhy a přesnější počítání sledovaných objektů než radarové a video modely samotné, což může být využito pro další analýzu dopravy v zkoumané oblasti.

Rekonstrukce a vylepšení poškozené části snímků otisků prstů

This thesis deals with the problem of fingerprint image reconstruction with focus on non- recoverable regions affected by various skin diseases. A generative adversarial network with learnable convolutional gabor filter layer was trained on preprocessed dataset of real fingerprint images. The work demonstrates that the trained model can reliably repair small corrupted regions of arbitrary shapes and in case of larger holes, the global quality score of reconstructed fingerprints evaluated by MINDTCT module from NIST biometric image software is increased compared to original fingerprint. A standardized format for fingerprint images that helped stabilize the results when training generative models is proposed. Táto práca sa zaoberá problémom rekonštrukcie snímkov odtlačkov prsta so zameraním na neobnovyteľné oblasti poškodené rôznymi kožnými ochoreniami. Generatívne súperi- ace siete s trénovateľnou konvolučnou vrstvou s gaborovými filtrami bola natrénovaná na dátovej sade reálnych snímkov odtlačkov prsta. Práca predvádza že natrénovaný model vie spoľahlivo rekonštruovať malé oblasti ľubovoľného tvaru a v prípade väčších oblastí, globálne skóre kvality rekonštruovaného odtlačku prsta získané využitím softvéru NIST biometric image software sa v porovnaní s originálnym snímkom navýšilo. Je navrhnutý štandardizovaný formát pre snímky odtlačkov prsta ktorý pomohol stabilizovat trénovanie generatívnych súperiacich sietí.
