Utilization of genome editing technology to knock out \kur{dnd1} gene in sturgeons

In this study, for the first time we used CRISPR/Cas9 gene editing technology in sturgeons i.e., sterlets (Acipenser ruthenus). The sequences of sgRNA and primers were designed based on published dnd1 sterlet sequence. Each pair of sgRNA oligos after ligation ready duplex DNA fragment was cloned into vector pX330-U6-Chimeric_BB-CBh-hSpCas9 backbone and thereafter the transformation to competent cells Escherichia coli DH5 was done. The plasmid carried sgRNA was extracted for downstream applications. We diluted extracted plasmid with 10% of 2 M KCl and injection into animal pole of fertilized eggs of sterlets at one to four-cell-stage, 4 hours post fertilization (hpf). At the same time, second microinjection with 2.5% FITC-biotin-dextrans was injected into vegetal pole for labelling PGCs. In the control groups, the eggs were only injected by 2.5% FITC into vegetal pole. PGCs of sterlet were visualized and photographed using a uorescent stereo microscope Leica M165 FC. To confirm the presence or deletion/insertion occurring in the target gene, we used MCE-202 MultiNA microchip electrophoresis system for DNA analysis, in which the targeted gene after amplifying by PCR was analyzed. Mutations in both treated and control embryos of sterlet were further assessed by Sanger sequencing of the PCR product. In present study, we successfully established basic protocols such as preparation of competent cells, construction of vector carrying sgRNA and its transformation into competent cells to carry out the CRISPR/Cas9 technology in sturgeons. Less number of PGCs was observed in embryos that were treated with CRISPR/Cas9; however, sequencing did not provide us a reliable evidence for mutation of the targeted gene probably due to an unspecific PCR. Therefore, more authentication of dnd1 knockout should be done in future by more specific PCR and repeated sequencing. Teacher questions supporting communication in the classroom (on the material of ELT)

The diploma thesis deals with the teacher questions in classroom interaction in the teaching of English at the second stage of primary school. Many studies show that quality teacher questions develop pupils' communication in foreign language teaching, increase the quality and quantity of the pupil's language. The theoretical part of the thesis first presents the position of English as a school subject within the Framework Educational Programme. The most extensive part of the thesis deals with the terms of classroom communication and interaction known as IRE or IRF. Based on the literature, the thesis focuses on initiation in the form of a question and describes the types of questions according to different typologies and tries to find out which teacher questions are most effective in classroom interaction. The practical part is based on video recordings of English language lessons at the second stage of primary school. This part analyses the questions based on the described typology and observes which types of teacher questions develop classroom communication and which, on the contrary, limit it. Diplomová práce se zabývá otázkami učitele ve třídních interakcích ve výuce anglického jazyka na 2. stupni ZŠ. Mnoho výzkumů totiž ukazuje, že kvalitní otázky učitele vedou k rozvoji komunikace u žáků v cizojazyčné výuce, zvyšují kvalitu a množství žákova jazyka. Teoretická část práce nejprve představuje postavení angličtiny jako školního předmětu v Rámcovém vzdělávacím programu. Nejrozsáhlejší část práce se zabývá vymezením pojmů třídní komunikace a interakce mezi učitelem a žákem nazývanou v anglicky psané literatuře jako IRE nebo IRF. Práce se zaměřuje na iniciaci ve formě otázky a na základě odborné literatury popisuje typy otázek podle různých typologií a zabývá se tím, které otázky učitele jsou ve třídní interakci nejefektivnější. Praktická část vychází z videonahrávek hodin anglického jazyka na 2. stupni ZŠ. Otázky analyzuje na základě popsané typologie a sleduje, které typy otázek nejvíce rozvíjejí komunikaci ve třídě a které ji naopak limitují. Analysis of the dicyemid diversity using amplicon sequencing

Using amplicon sequencing, this study reveals dicyemid diversity around the world as well as within individual cephalopod hosts. A wide number of samples of multiple species were collected to reflect true dicyemid diversity. Heterogeneity primers were designed in order to improve the sequencing performance of the Illumina platform. A complex bioinformatic pipeline was implemented to process non-overlapping reads. The species delimitation methods were used to categorize occurring dicyemid types. Using the output of implemented methods, dicyemid diversity was assessed, proposing new trends opposing the current methodology. This thesis deals with the issue of using songs in English language teaching at lower secondary schools. The thesis comprises of two parts a theoretical one and a practical one. In the theoretical part, I mostly describe and explain the basic terms used, such as song, music and rhythm. The teaching methodology and song activities that can be used in English lessons are also included in this part. Further, I also deal with the influence of music on students and the atmosphere in the classroom in connection with music. Based on these findings, I prepared four English lesson plans in accordance with Educational Framework and school curriculum. These lessons also correspond to the topics or grammatical structures that the students currently deal with in their lessons. The main focus is placed on the skills and subskills that are developed by the particular activities. All these lessons were put into practise at lower secondary school level. Subsequently, I also give feedback on these lessons. Another part of this thesis consists of an analysis of English textbooks with respect to the usage of songs and song activities in lessons. For this purpose, four volumes of the textbook Project Third Edition were chosen. These textbooks were analysed from the point of view of the main educational aims and types of activities, mostly with respect to the skills and subskills practised via these songs. Tato diplomová práce se zabývá studiem a implementací písní ve výuce anglického jazyka na druhém stupni základních škol. Práce je rozdělena na dvě části teoretickou a praktickou. V teoretické části popisuji a rozvíjím základní pojmy jako píseň, hudba a rytmus společně s metodikou výuky a hudebními aktivitami, které mohou být použity ve výuce anglického jazyka. Dále se zde zabývám také vlivem hudby na studenty a atmosférou ve třídě ovlivněnou právě hudbou. Na základě těchto teoretických podkladů jsem připravila čtyři plány vyučovacích hodin, které jsou také v souladu s RVP a ŠVP a odpovídají tématům či rozvíjejí gramatické struktury, kterými se žáci v běžných hodinách zabývají. Hlavní důraz je kladen na řečové dovednosti, které dané aktivity rozvíjejí. Tyto hodiny byly odučeny na druhém stupni ZŠ a následně také reflektovány. Další částí této práce je analýza učebnic z hlediska využití písní a písňových aktivit. Pro tyto účely byly vybrány čtyři díly učebnice Project 3.edice. Tyto učebnice byly analyzovány z hlediska výukových cílů, typu aktivit a především z hlediska řečových dovedností, které jsou danými aktivitami rozvíjeny. The present thesis aims to analyse a narrative interaction between a child (Natálka) and an adult with regard to discursive strategies employed in the context of imagination and creativity. I applied the method of Conversation Analysis to the examination of the transcribed audio recordings that were taken in 2018 and at the turn of 2020 during the interaction between Natálka and myself. An analysis of the data identified that in the child's repertoire the following practices were present: (1) linking two or more episodes in a conversation, (2) asking questions, (3) separating individual topics, and (4) using other discourse elements; concretely humour, exaggeration, and negotiation techniques. The first part of the thesis sets out the discursive practices examined and argues for the insights these provide into the imagination and creativity of a child. The analysis, which is undertaken in the form of a case study, subsequently maps the use of the selected strategies developed over two years of Natálka's life and characterises how these changes reflect the child's cognitive development. Tato diplomová práce si klade za cíl analyzovat narativní interakci mezi dítětem (Natálkou) a dospělou osobou s ohledem na diskurzivní strategie v kontextu představivosti a kreativity. Ve své práci jsem použila metodu konverzační analýzy, kterou jsem následně aplikovala na transkripční přepisy audio nahrávek, jež byly pořízeny během dvou časových obdobích: v roce 2018 a na přelomu roku 2020 při interakci mezi Natálkou a mnou. Analýza dat odhalila, že v repertoáru dítěte byly přítomny následující praktiky: (1) spojování dvou či více epizod ve vyprávění, (2) kladení otázek, (3) oddělování jednotlivých témat a (4) použití dalších diskurzivních elementů, konkrétně humoru, zveličování či nadsázky a vyjednávacích taktik. První část práce se podrobně věnuje diskurzivním praktikám a vysvětluje rozdíly mezi představivostí a kreativitou. Tato případová studie následně mapuje jakým způsobem se v průběhu dvou let vyvinulo použití zkoumaných strategií a charakterizuje, jak tyto změny reflektují kognitivní vývoj dítěte.
